The determination of U and Th was carried out on pooled samples of muscle tissue of cod (Gadus morhua), herring (Clupea harengus), sprat (Sprattus sprattus) and some other species of fish caught in 1981 in the southern Baltic. The levels of U obtained in the present study are
generally within the ranges presented by other authors for fish from different aquatic regions of the world. The concentrations
of U are similar to those found for Th in Baltic fish analysed. The concentration factors CFU(Th), discrimination factors DFU/Th and Th/U ratios (by weight) were calculated and discussed. These coefficients were also compared with values determined in
other Baltic organisms. Transfer of U and Th along a food chain from potential prey (diet) to fish as predator (consumer)

Chromosomal integration of multicopy transgene inserts in higher plants is often followed by loss of expression. We have analysed whether this inactivation can trigger repeat-induced point mutations (RIP) as has been observed in Neurospora crassa. We have previously characterized transgenic lines of Arabidopsis thaliana containing the hygromycin phosphotransferase (hpt) gene either as a unique sequence in plants expressing the gene, or as multimeric, closely linked repeats in clones that were resistant to hygromycin directly after transformation but exhibited gene inactivation in the subsequent generation. At the sequence level, we have determined the mutation frequencies in the promoter and coding regions of active and inactive copies of transgene inserts after passage through three sexual generations. No RIP-like mutations were found in inactivated genes. Comparison of our data with those from Neurospora suggest that sequence divergence within plant repetitive DNA is either much slower than in Neurospora or is generated by a different mechanism. 相似文献

The PDC (pyruvate dehydrogenase complex) is strongly inhibited by phosphorylation during starvation to conserve substrates for gluconeogenesis. The role of PDHK4 (pyruvate dehydrogenase kinase isoenzyme 4) in regulation of PDC by this mechanism was investigated with PDHK4-/- mice (homozygous PDHK4 knockout mice). Starvation lowers blood glucose more in mice lacking PDHK4 than in wild-type mice. The activity state of PDC (percentage dephosphorylated and active) is greater in kidney, gastrocnemius muscle, diaphragm and heart but not in the liver of starved PDHK4-/- mice. Intermediates of the gluconeogenic pathway are lower in concentration in the liver of starved PDHK4-/- mice, consistent with a lower rate of gluconeogenesis due to a substrate supply limitation. The concentration of gluconeogenic substrates is lower in the blood of starved PDHK4-/- mice, consistent with reduced formation in peripheral tissues. Isolated diaphragms from starved PDHK4-/- mice accumulate less lactate and pyruvate because of a faster rate of pyruvate oxidation and a reduced rate of glycolysis. BCAAs (branched chain amino acids) are higher in the blood in starved PDHK4-/- mice, consistent with lower blood alanine levels and the importance of BCAAs as a source of amino groups for alanine formation. Non-esterified fatty acids are also elevated more in the blood of starved PDHK4-/- mice, consistent with lower rates of fatty acid oxidation due to increased rates of glucose and pyruvate oxidation due to greater PDC activity. Up-regulation of PDHK4 in tissues other than the liver is clearly important during starvation for regulation of PDC activity and glucose homoeostasis. 相似文献

Summary Comparative analysis of the available 3′-portions of the human L1 (LINE-1) family of repeated sequences indicates that all
the sequences can be classified in two major subfamilies. The division is based on patterns of diagnostic bases shared within
L1 subfamilies of sequences but differing between them. The overall ratio of replacement to synonymous positions, occupied
by the diagnostic bases in the large open reading frame of the L1 sequence, is 1.15. This indicates that both subfamilies
were obtained from genes coding for functional proteins. The L1 subfamilies appear to be of different ages and may represent
a “fossil record” of the same active gene at different times in the history of primates. The younger subfamily can be split
further into at least two closely related branches of sequences. The above facts combined with the recent data for the Alu
subfamily structure show that LINE and SINE families of interspersed repeats share discontinuous patterns in their evolution.
These data are consistent with the model that both Alu and L1 families, as well as other pseudogene families, contain active
genes producing discrete layers of pseudogenes throughout the history of primates. Models of evolutionary processes that could
generate these discontinuities are discussed together with the possible biological role of Alu and L1 genes. 相似文献

Current information on barley resistance genes available from scientific papers and on-line databases is summarised. The recent literature contains information on 107 major resistance genes (R genes) against fungal pathogens (excluding powdery mildew), pathogenic viruses and aphids identified in Hordeum vulgare accessions. The highest number of resistance genes was identified against Puccinia hordei, Rhynchosporium secalis, and the viruses BaYMV and BaMMV, with 17, 14 and 13 genes respectively. There is still a lot of confusion regarding symbols for R genes against powdery mildew. Among the 23 loci described to date, two regions Mla and Mlo comprise approximately 31 and 25 alleles. Over 50 R genes have already been localised and over 30 mapped on 7 barley chromosomes. Four barley R genes have been cloned recently: Mlo, Rpg1, Mla1 and Mla6, and their structures (sequences) are available. The paper presents a catalogue of barley resistance gene symbols, their chromosomalocation and the list of available DNA markers useful in characterising cultivars and breeding accessions. 相似文献

Summary Jasmonic acid (JA) effects on in vitro tuberization of potato nodal explants cvs. Sangre and Russet Burbank were tested under liquid and solid media conditions
and 0,8, and 16h photoperiod. Explants taken from stock plants grown on 2.5μM JA-supplemented medium tuberized first, particularly in darkness. The most pronounced benefits of the JA pretreatment were
recorded under 16h photoperiod, which is known to inhibit tuberization. Cultivar Sangre benefited from the JA preconditioning
of stock plants more than Russet Burbank. Russet Burbank required the JA supplement in tuberization media to reach the same
degree of stimulation. Overall, microtubers produced either from JA preconditioned stock plants or on the JA-containing tuberization
media were more uniform and larger than from other treatments. Eight hours photoperiod was by far the best treatment for the
production of high-quality uniform microtubers. JA conditioning of stock plants prior to taking explants for tuberization

Taking into account that fructophilic lactic acid bacteria (FLAB) can play an important role in the health of honey bees and can be used as probiotics, phenotypic properties of probiotic interest of Lactobacillus kunkeei (12 strains) and Fructobacillus fructossus bacteria (2 strains), isolated from Apis mellifera gastrointestinal tract, have been studied. We have evaluated survival of tested FLAB in honey bee gut, their susceptibility to antibiotics (ampicillin, erythromycin, tylosin), cell surface hydrophobicity, auto-aggregation ability, co-aggregation with model pathogenic bacteria, biofilm formation capacity, and effect of studied FLAB, added to sucrose syrup bee diet, on longevity of honey bees. The tested FLAB exhibited good gastrointestinal tract tolerance and high antibiotic susceptibility, which are important criteria in the screening of probiotic candidates. It was also found that all FLAB studied have high cell surface hydrophobicity and fulfil next selection criterion for their use as probiotics. Symbionts of A. mellifera showed also auto- and co-aggregation capacities regarded as valuable features for biofilm formation and inhibition of pathogens adhesion to the bee gut cells. Biofilm-development ability is a desired characteristic of probiotic lactic acid bacteria. As indicated by quantitative crystal violet-stained microplate assay and confocal laser scanning microscopy imaging, all studied A. mellifera gut isolates exhibit a biofilm positive phenotype. Moreover, it was also documented, on honey bees kept in cages, that supplementation of A. mellifera sucrose diet with FLAB decreases mortality and improves significantly longevity of honey bees. A rare marine tardigrade Halobiotus crispae Kristensen, 1982 was found abundant in intertidal flats at Josephbukta, Bellsund fiord, West Spitsbergen. This is the first record of this species from the Svalbard Archipelago. So far, the species has been found only in seven other localities in the northern hemisphere (between 56° and 69°N). The present record greatly extends the known geographical range to the north and also indicates a much wider distribution of the species than formerly reported. Moreover, a potential indicative character of this species and of the study area in monitoring climatic changes is discussed. 相似文献

A new species of the genus Mesostoma Ehrenberg 1935, M. zariae n.sp. is described and its relationship discussed. The new species belongs to the M. lingua species-group and is characterised by the presence of two kinds of prostate secretions and a pear-shaped penis papilla. It occurs in small standing or slowly running waters at Zaria, Nigeria. It has previously been proven (Mead 1978) that this species is predatory on the aquatic stages of mosquitoes. 相似文献

The objective of this study was to assess the dynamics of oxidative damage to cellular macromolecules such as proteins, lipids and DNA under conditions of oxidative stress triggering early stages of estrogen-dependent carcinogenesis. A rodent model of carcinogenesis was used. Syrian hamsters were sacrificed after 1, 3, 5 h and one month from the initial implantation of estradiol. Matching control groups were used. Kidneys as target organs for estradiol-mediated oxidative stress were excised and homogenized for biochemical assays. Subcellular fractions were isolated. Carbonyl groups (as a marker of protein oxidation) and lipid hydroxyperoxides were assessed. DNA was isolated and 8-oxodGuo was assessed. Electron paramagnetic resonance spectroscopy was used to confirm the results for lipid peroxidation. Exposition to estradiol in the rodent model leads to damage of macromolecules of the cell, including proteins and DNA, but not lipids. Proteins appear to be the primary target of the damage but are closely followed by DNA. It has previously been speculated that protein peroxides can increase DNA modifications. This time sequence was observed in our study. Nevertheless, the direct relation between protein and DNA damage still remains unsolved. 相似文献
