Methanobactin (mb) is a novel chromopeptide that appears to function as the extracellular component of a copper acquisition system in methanotrophic bacteria. To examine this potential physiological role, and to distinguish it from iron binding siderophores, the spectral (UV–visible absorption, circular dichroism, fluorescence, and X-ray photoelectron) and thermodynamic properties of metal binding by mb were examined. In the absence of Cu(II) or Cu(I), mb will bind Ag(I), Au(III), Co(II), Cd(II), Fe(III), Hg(II), Mn(II), Ni(II), Pb(II), U(VI), or Zn(II), but not Ba(II), Ca(II), La(II), Mg(II), and Sr(II). The results suggest metals such as Ag(I), Au(III), Hg(II), Pb(II) and possibly U(VI) are bound by a mechanism similar to Cu, whereas the coordination of Co(II), Cd(II), Fe(III), Mn(II), Ni(II) and Zn(II) by mb differs from Cu(II). Consistent with its role as a copper-binding compound or chalkophore, the binding constants of all the metals examined were less than those observed with Cu(II) and copper displaced other metals except Ag(I) and Au(III) bound to mb. However, the binding of different metals by mb suggests that methanotrophic activity also may play a role in either the solubilization or immobilization of many metals in situ.  相似文献

Bacterial communities in two shallow eutrophic aquatic ecosystems (eastern China) were studied using culture-dependent methods, and their correlations with the other water parameters were analyzed. Although the values of the comprehensive trophic state index in Xizi Lake and Hangzhou Canal were almost identical, the abundances of cultivable bacterial communities, such as protein-hydrolyzing bacteria (PHB), fecal coliforms (FC), nitrogen-utilizing bacteria, phosphate-mineralizing bacteria, and cellulose-decomposing bacteria (CDB), differed significantly. They were much less in Xizi Lake than in Hangzhou Canal. Correlation analyses indicated that the abundances of physiological groups of bacteria were determined mainly by the biomasses of phytoplankton and zooplankton, rather than by the utilized substrates. Xizi Lake was an algae-dominated aquatic ecosystem, a situation that mainly arose from the influx of inorganic nutrients, and the Hangzhou Canal was bacteria-dominated due to the influx of organic sewage. Molecular characterization and phylogenetic analysis of isolates showed that there were 6 phylogenetic lineages out of 15 isolates screened from Xizi Lake, including γ-proteobacteria (5), β-proteobacteria (3), α-proteobacteria (2), actinobacteria (1), firmicutes (4), and bacteroidetes (1). While in Hangzhou Canal there were only 4 bacterial groups among 22 isolates, γ-proteobacteria comprised about 82%, α-proteobacteria made up 9%, firmicutes and bacteroidetes made up only 4.5%, respectively, and no β-proteobacteria were found. Enterobacteriaceae were the principal bacteria components in the two aquatic ecosystems, especially in the sewage-polluted Hangzhou Canal. 本研究探讨利用荧光定量PCR技术评价Sindbis病毒经亚甲蓝光化学处理后灭活效果的可行性。研究采用不同光照强度对Sindbis病毒进行亚甲蓝光化学灭活处理,并用SYBR Green I荧光定量PCR对Sindbis病毒的cDNA进行扩增,同时以细胞病变法做平行对照以测定病毒残余滴度。结果显示在亚甲蓝光化学处理过程中,随着光照强度的增强,病毒残余滴度由6.50 LgTCID50/mL逐渐降低至检测限以下,同时病毒核酸的拷贝数显著下降(P<0.05),并与病毒感染性的降低呈线性相关(R2>0.98)。以上结果表明,亚甲蓝光化学灭活法对Sindbis病毒核酸有破坏作用,病毒核酸损伤程度随光照强度的增强而增加,且与病毒感染性的降低存在相关性,提示荧光定量PCR技术评价亚甲蓝光化学法的病毒灭活效果具有可行性。  相似文献

Mannan components of C. albicans (5 mg/kg, i.p.) and S. cerevisiae (2.5 mg/kg, i.p.) cell walls produced pyrogenic responses which were completely inhibited by indomethacin (5 mg/kg, s.c.) pretreatment in rats. A non-selective NOS inhibitor, L-NAME (10 mg/kg, s.c.), also inhibited the pyrogenic effectiveness of C. albicans mannan, whereas it was ineffective on the fever induced by S. cerevisiae mannan. A selective elevation in the serum TNF-alpha levels was observed at the initial phase of the fever due to S. cerevisiae mannan, whereas there was no significant change on the serum levels of TNF-alpha, IL-1beta and IFN-gamma during the latent period or at the initial phase of the fever induced by C. albicans mannan. Injections of N-linked and/or O-linked oligomannosides of the either mannan did not cause any significant change in the body temperature and serum cytokine levels. These data suggest that the mannan components of C. albicans and S. cerevisiae cell walls produce a prostaglandin-dependent fever in rats. The initial signal for fever seems to be different for each mannan. Data also indicate that integrity of the mannans is necessary for the pyrogenic response.  相似文献

A novel hybrid of small core@shell structured CoSx@Cu2MoS4 uniformly hybridizing with a molybdenum dichalcogenide/N,S‐codoped graphene hetero‐network (CoSx@Cu2MoS4‐MoS2/NSG) is prepared by a facile route. It shows excellent performance toward the oxygen reduction reaction (ORR), oxygen evolution reaction (OER), and hydrogen evolution reaction (HER) in alkaline medium. The hybrid exhibits rapid kinetics for ORR with high electron transfer number of ≈3.97 and exciting durability superior to commercial Pt/C. It also demonstrates great potential with remarkable stability for HER and OER, requiring low overpotential of 118.1 and 351.4 mV, respectively, to reach a current density of 10 mA cm?2. An electrolyzer based on CoSx@Cu2MoS4‐MoS2/NSG produces low cell voltage of 1.60 V and long‐term stability, surpassing a device of Pt/C + RuO2/C. In addition, a Zn‐air battery using cathodic CoSx@Cu2MoS4‐MoS2/NSG catalyst delivers a high cell voltage of ≈1.44 V and a power density of 40 mW cm?2 at 58 mA cm?2, better than the state‐of‐the‐art Pt/C catalyst. These achievements are due to the rational combination of highly active core@shell CoSx@Cu2MoS4 with large‐area and high‐porosity MoS2/NSG to produce unique physicochemical properties with multi‐integrated active centers and synergistic effects. The outperformances of such catalyst suggest an advanced candidate for multielectrocatalysis applications in metal‐air batteries and hydrogen production.  相似文献

半干旱沙漠中樟子松和沙柳造林后土壤有机碳及其组分的小尺度空间分布 半干旱沙漠造林有助于改善土壤功能以及增加土壤有机碳(SOC)固定,但人们对造林后SOC及其不稳定(LOC)组分的小尺度空间分布了解甚少。本研究以毛乌素沙地东南缘樟子松(Pinus sylvestris)和沙柳 (Salix psammophila)为研究对象,量化了距离树体20、80、150和240 cm处SOC、LOC组分及其相关变量的小尺度空间分布。研究结果表明,沙柳和樟子松造林显著提高了SOC、总氮(TN)、可溶性有机碳 (DOC)、微生物碳(MBC)和易氧化有机碳(ROOC)含量;在距离树体20 cm处,0–100 cm土层樟子松SOC 储量比沙柳高27.21%;在距离树体80和150 cm处,沙柳SOC储量分别比樟子松高5.50%和5.66%;与流 沙地相比,在距离树体20、80、150 和240 cm处,沙柳和樟子松SOC储量显著增加了94.90%、39.50%、 27.10%和18.50%;沙柳和樟子松ROOC分别占SOC的14.09%和18.93%。总之,造林促进了半干旱流沙地SOC的积累,樟子松比沙柳分配更多的有机质到距离树体<80 cm范围内的土体中。  相似文献

Metastasis is the primary cause of an unfavourable prognosis in patients with malignant cancer. Over the last decade, the role of proteinases in the tumour microenvironment has attracted increasing attention. As a sensor of proteinases, proteinase-activated receptor 2 (PAR2) plays crucial roles in the metastatic progression of cervical cancer. In the present study, the expression of PAR2 in multiple types of cancer was analysed by Gene Expression Profiling Interactive Analysis (GEPIA). Kaplan-Meier plotter was used to calculate the correlation between survival and the levels of PAR2, Grb-associated binding protein 2(Gab2) and miR-125b. Immunohistochemistry (IHC) was performed to examine PAR2 expression in a tissue microarray (TMA) of CESCs. Empower Stats was used to assess the predictive value of PAR2 in the metastatic potential of CESC. We found that PAR2 up-regulation was observed in multiple types of cancer. Moreover, PAR2 expression was positively correlated with the clinicopathologic characteristics of CESC. miR-125b and its target Gab2, which are strongly associated with PAR2-induced cell migration, are well-characterized as predictors of the prognostic value of CESC. Most importantly, the Cancer Genome Atlas (TCGA) data set analysis showed that the area under the curve (AUC) of the PAR2 model was significantly greater than that of the traditional model (0.833 vs 0.790, P < .05), demonstrating the predictive value of PAR2 in CESC metastasis. Our results suggest that PAR2 may serve as a prognostic factor for metastasis in CESC patients.  相似文献

The use of human pluripotent stem cell-derived cardiomyocytes (hPSC-CMs) is limited in drug discovery and cardiac disease mechanism studies due to cell immaturity. Micro-scaled grooves can promote the maturation of cardiomyocytes by aligning them in order, but the mechanism of cardiomyocytes alignment has not been studied. From the level of calcium activity, gene expression and cell morphology, we verified that the W20H5 grooves can effectively promote the maturation of cardiomyocytes. The transient receptor potential channels (TRP channels) also play an important role in the maturation and development of cardiomyocytes. These findings support the engineered hPSC-CMs as a powerful model to study cardiac disease mechanism and partly mimic the myocardial morphological development. The important role of the TRP channels in the maturation and development of myocardium is first revealed.  相似文献
