Telomerase is a key component of the telomere length maintenance system in the majority of eukaryotes. Telomerase displays maximal activity in stem and cancer cells with high proliferative potential. In humans, telomerase activity is regulated by various mechanisms, including the interaction with telomere ssDNA overhangs that contain a repetitive G‐rich sequence, and with noncoding RNA, Telomeric repeat‐containing RNA (TERRA), that contains the same sequence. So these nucleic acids can compete for telomerase RNA templates in the cell. In this study, we have investigated the ability of different model substrates mimicking telomere DNA overhangs and TERRA RNA to compete for telomerase in vitro through a previously developed telomerase inhibitor assay. We have shown in this study that RNA oligonucleotides are better competitors for telomerase that DNA ones as RNA also use an alternative binding site on telomerase, and the presence of 2′‐OH groups is significant in these interactions. In contrast to DNA, the possibility of forming intramolecular G‐quadruplex structures has a minor effect for RNA binding to telomerase. Taking together our data, we propose that TERRA RNA binds better to telomerase compared with its native substrate – the 3′‐end of telomere DNA overhang. As a result, some specific factor may exist that participates in switching telomerase from TERRA to the 3′‐end of DNA for telomere elongation at the distinct period of a cell cycle in vivo. Geotemporal information, information associated with geographical space and time, has always been critical to climate and environmental science. However, this information is certainly not universally or easily accessible. In fact, obtaining and using geotemporal information often comes with a considerable technical overheads, impeding research progress. To address this, we introduce FetchClimate: a cloud service designed to provide easy, universal access to geotemporal information. FetchClimate enables and accelerates the use of geotemporal information by enabling it to be accessed programmatically from a Web service (such as the statistical software R) or non‐programmatically using a Web browser. We intend the service to accelerate the pace of ecological and environmental research by eliminating the technical overheads currently needed to obtain geotemporal information. Objective

Myelin oligodendrocyte glycoprotein (MOG) is one of the major autoantigens in multiple sclerosis (MS), therefore selective depletion of autoreactive lymphocytes exposing MOG-specific B cell receptors (BCRs) would be beneficial in terms of MS treatment.

Results

Using E. coli we generated an efficient protocol for the purification of the recombinant immunotoxin DT-MOG composed of the extracellular Ig-like domain of MOG fused in frame with the catalytic and translocation subunits of diphtheria toxin (DT, Corynebacterium diphtheriae) under native conditions with a final yield of 1.5 mg per liter of culture medium. Recombinant DT-MOG was recognized in vitro by MOG-reactive antibodies and has catalytic activity comparable with wild-type DT.

Conclusion

Enhanced pharmacokinetics (mean residence time in the bloodstream of 61 min) and minimized diminished nonspecific toxicity (LD50 = 1.76 mg/kg) of the DT-MOG makes it a potential candidate for the immunotherapy of MS.

Insect- and bird-size drones—micro air vehicles (MAV) that can perform autonomous flight in natural and man-made environments are now an active and well-integrated research area. MAVs normally operate at a low speed in a Reynolds number regime of 104–105 or lower, in which most flying animals of insects, birds and bats fly, and encounter unconventional challenges in generating sufficient aerodynamic forces to stay airborne and in controlling flight autonomy to achieve complex manoeuvres. Flying insects that power and control flight by flapping wings are capable of sophisticated aerodynamic force production and precise, agile manoeuvring, through an integrated system consisting of wings to generate aerodynamic force, muscles to move the wings and a control system to modulate power output from the muscles. In this article, we give a selective review on the state of the art of biomechanics in bioinspired flight systems in terms of flapping and flexible wing aerodynamics, flight dynamics and stability, passive and active mechanisms in stabilization and control, as well as flapping flight in unsteady environments. We further highlight recent advances in biomimetics of flapping-wing MAVs with a specific focus on insect-inspired wing design and fabrication, as well as sensing systems.This article is part of the themed issue ‘Moving in a moving medium: new perspectives on flight’.  相似文献

One of the leitmotifs of the ecophysiological research on ectotherms is the variation and evolution of thermal reaction norms for biological rates. This long‐standing issue is crucial both for our understanding of life‐history diversification and for predicting the phenology of economically important species. A number of properties of the organism's thermal phenotype have been identified as potential constraints on the evolution of the rate–temperature relationship. This comparative study addresses several such constraints by testing whether the actual interspecific variation of thermal reaction norms across nearly hundred leaf beetle species agrees with the expected patterns. The results show that developmental rate and its temperature‐dependent parameters are similar in closely related species and that the variation pattern depends on the taxonomic scale, the thermal reaction norms being mostly parallel for the representatives of distant subclades but intersecting more often farther down the phylogenetic tree. The parallel shift disagrees with the putative ubiquity of a positive slope–threshold relationship, whereby thermal reaction norms should normally intersect, and even more contradicts with the common‐intersection hypothesis. The ability to develop in cooler conditions is not traded off at higher temperatures, which is an exception to the “warmer is better” principle. A comparison of high‐ and low‐quality data indicates that some of these discrepancies with earlier findings may stem from a likely presence of noise in previous analyses, which may have affected the variation patterns observed. Overall, the failure to support the universality of the predicted patterns suggests that the evolution of thermal reaction norms in leaf beetles has largely overcome the hypothesized constraints.  相似文献

Capsule: Sympatric Marsh Warblers Acrocephalus palustris and Blyth’s Reed Warblers Acrocephalus dumetorum differ significantly in their life history traits.

Aims: To provide a direct comparison of demographic parameters among two sympatric populations of the closely related Marsh Warbler and Blyth’s Reed Warbler.

Methods: We examined breeding phenology and reproductive traits at a 25?ha study plot. We use program MARK to estimate daily nest survival and adult apparent survival rates.

Results: On average, Marsh Warblers laid the first eggs 3 days later than Blyth’s Reed Warblers. Mean clutch size in the Marsh Warbler was significantly lower than in the Blyth’s Reed Warbler. There are no significant differences between the two species for nest daily survival, duration of incubation and nestling periods. Apparent survival of adults was slightly higher in Marsh Warblers than in Blyth’s Reed Warblers.

Conclusion: Our findings suggest that two ecologically similar sympatric species differ significantly in terms of life history traits. We assume that observed differences could be the result of adaptations to environmental factors in the central parts of the species’ ranges or due to differences in mortality on migratory pathways or wintering grounds.  相似文献
