Mass spectrometry–based proteomics was employed to analyze urine from eight healthy volunteers during a 21-day bed rest (BR) study. The analysis included trypsinolysis in solution prior to liquid chromatography and tandem mass spectrometry (LC-MS/MS), and spectrum processing using the bioinformatic tools. Relying on 221 IPI indices with scores from 24 to 1700, 169 different proteins were identified. Molecular functions, biological processes, and cell components as the loci of certain protein functioning were determined with the help of UniProt-GOA. Associative interactions networks were constructed using BiNGO. There were 14 proteins identified that were functional in the cardiovascular system mostly. They were annotated, and the dynamics of their occurrence throughout the experiment was considered. The paper is concerned with development of conditions for cultivation of the erythromycin-producing organism and preparation and maintenance of its stable viable protoplasts. Optimal conditions for the culture growth and protoplasting were developed. Two-stage cultivation of the organism on media PB and S provided dense diffuse or diffuse local growth characterized by low differentiation and higher homogenicity. The incubation time at stage I was 66 hours and that at stage II was not more than 24 hours at respective temperatures. The culture was incubated on a shaker in the presence of glycine at the minimum concentration. The presence of glycine in the medium altered the culture cell walls which was evident from changing of staining by Gram from + to +/- . Treatment of such a culture with lysozyme for 30 minutes provided formation of up to 2.10(9) protoplasts per 1. ml. It is possible to maintain the protoplasts in frozen state at -20 degrees C in medium P for 1 month. Under such storage conditions the titer of the viable protoplasts as compared to the initial one decreased only 2-fold after the one-month storage. 相似文献

We have developed a new approach to the analysis of hypomethylated CpG patterns within predetermined, megabase long, genome regions. The approach, which we term Non-methylated Genomic Sites Coincidence Cloning (NGSCC), includes three main steps. First, total genomic DNA is digested with a methylation sensitive restriction endonuclease, such as Hpa II or Hha I. Then the fragments corresponding to the genomic area of interest are selected. To this end the fragmented genome DNA is hybridized with a mixture of clones (BACs, cosmids etc.) representing a given region and digested with the same restriction enzyme(s). A special version of the coincidence cloning procedure was developed to make this hybridization selection highly efficient and specific. Finally, fragments of the locus under study are mapped and sequenced. The technique proved to be efficient and specific. As a test, it was applied to the analysis of hypomethylated CpG patterns along the 1-Mb D19S208-COX7A1 (Chr 19q13.12) locus, on human chromosome 19, in normal testis and in seminoma tissues. Some differences in the distribution of hypomethylated CpGs between the two tissues were demonstrated. The methylation profiles in both tissues revealed a clear trend to clustering of non-methylated sites. We also analyzed the expression of genes located within hypomethylated clusters in both tissues. It was shown that, whereas the expression of some of the genes investigated was correlated with hypomethylation of the region, other genes were expressed regardless of their methylation status. NGSCC thus promises to be a useful approach for the analysis of the role of dynamic epigenetic factors in genome function.Communicated by G. P. Georgiev 相似文献

Polyclithrum ponticum sp. n. is described and P. mugilini Rogers, 1967 is redescribed. Both monogenean species are parasites of Mugil cephalus in the Black Sea. The new species differs from P. mugilini, P. alberti and P. boegeri by the lesser size of anchors, while it is distinguished from P. corallense by the larger size of these structures. P. ponticum sp. n. differs from all formerly described species by the greater length of dorsal connective bar. In both species from the Black Sea, "ear-like" structures situated near the external roots of anchors are described for the first time. It is suggested, that these structures take part in longitudinal, two-lobe folding of the haptor. The process of opening the haptor is probably performed by the additional bars of the haptor (bars 2 and 3 after: Rogers, 1967), joined to each other and with the anchors. The fifth pair of additional bars (Ernst e. a., 2000) derives from the "beard" of ventral connective bar and is united with its basal part. The sixth pair of additional bars (Ernst e. a., 2000) is considered as a typical "ribs" of the haptor, and therefore the "ribs" are represented by three pairs. Differences between marginal hooks of P. ponticum sp. n. and P. mugilini are insignificant, that probably depends on the presence of "ribs" of the haptor. Based on the subdivision of marginal hooks into two groups, the presence of additional supporting structure in the haptor, and the presence of the seminal receptacle, it is suggested that the subfamily Polyclithrinae Rogers, 1967 should include the genera Polyclithrum Rogers, 1967, Swingleus Rogers, 1969, Macrogyrodactylus Mamlberg, 1959, and probably Fundulotrema Hargis, 1955. Based on such characters as the lack of the anchors, the presence of suckers in the haptor, and ovipositing of eggs, it seems to be expedient to use the following taxa in systematics of gyrodactylids: Isancistrinae Fuhrmann, 1928 (genera Isancistrum, Anacanthocotyle); Gyrdicotylinae Vercammen-Grandjean, 1960 (Gyrdicotyle) and Ooegyrodactylinae Harris, 1983 (genera Phanerothecium, Ooegyrodactylus, Nothogyrodactylus, Hyperopletes). 相似文献
