外源茉莉酸甲酯对辣椒烟粉虱的影响

化学农药仍是目前蔬菜烟粉虱Bemisia tabaci防控的重要措施,研发有效的非化学措施防控烟粉虱是蔬菜绿色生产亟待解决的问题。本文以苏椒15号（感性品种）和新苏椒五号（抗性品种）辣椒Capsicum annuum L.为对象,通过在辣椒上喷施茉莉酸甲酯,研究外源茉莉酸甲酯对烟粉虱发育历期、存活率、消化酶活性和田间烟粉虱种群动态的影响。研究发现,取食茉莉酸甲酯处理的辣椒后烟粉虱的发育历期显著延长、存活率明显下降。其中取食感性品种辣椒和抗性品种辣椒的烟粉虱发育历期分别延长7.66 d和9.66 d,存活率分别下降10.5%和9.89%;取食感性品种辣椒的烟粉虱主要是延长卵期、4龄若虫期和伪蛹期,而取食抗性品种辣椒的烟粉虱各历期均延长。外源茉莉酸甲酯对烟粉虱体内消化酶的活性也有明显的影响,取食茉莉酸甲酯处理的辣椒后烟粉虱的消化酶活性显著下降,取食感性品种辣椒和抗性品种辣椒24 h后,胰蛋白酶活力分别较对照下降了61.86%和45.0%,糜蛋白酶活性分别较对照下降了47.29%和40.11%。大田辣椒上喷施茉莉酸甲酯后,烟粉虱的种群数量迅速下降,但在抗性品种和感性品种上烟粉虱的校正虫口减退率没有明显的差异。研究发现,外源茉莉酸甲酯处理,可以延缓烟粉虱的发育速率,降低胰蛋白酶和糜蛋白酶的活性,对田间烟粉虱种群也有较好的控制作用。     

藏山羊KLF8基因克隆及组织表达分析

本试验旨在获得藏山羊KLF8基因序列,并分析其生物学特征,同时阐明该基因在不同组织中的表达情况。利用RT-PCR技术克隆藏山羊KLF8基因序列,利用实时荧光定量PCR (quantitative real-time PCR,qPCR)检测其在藏山羊各个组织中的表达丰度。结果表明,获得藏山羊KLF8基因序列1 069 bp,其中包含CDS区1 008 bp,5'UTR序列28 bp和3'UTR序列33 bp,共编码335个氨基酸,为不稳定亲水碱性蛋白。KLF8基因在藏山羊的肺脏组织中表达水平最高,极显著高于其他组织(p<0.01)。本研究为进一步阐明KLF8基因在藏山羊中的生物学功能提供了依据。     

韭菜迟眼蕈蚊雄虫日龄对其交配能力和雌虫生殖力的影响

为了提高韭菜迟眼蕈蚊性信息素监控水平,本文在25℃、RH70%±5%、光周期L∶D=14∶10的条件下研究韭菜迟眼蕈蚊不同日龄雄虫的交配能力及其对雌虫生殖力的影响。结果表明,未交配雄虫平均寿命4.5d,雌虫4.2d,雌雄成虫的平均寿命无显著差异。雄虫一生最多可交配13次,且随着雄虫日龄增加,雌虫交配成功率逐渐减少。随着雄虫交配经历的增加,雌虫交配的时间延长,最长达97min。雄虫的日龄并不影响与之交配雌虫的产卵量和卵孵化率,但是雄虫的交配经历与雌虫的产卵量和卵孵化率相关,尤其当雄虫交配经历超过8次时,与之交配的雌虫的产卵量和卵孵化率显著下降。研究阐明了韭菜迟眼蕈蚊的生殖行为特征,为其性信息素应用提供了参考。     

水牛CD14 cDNA的克隆、表达及鉴定

目的:克隆水牛白细胞分化抗原14(buffalo cluster of differentiation antigen14,bCD14)基因,表达bCD14蛋白,并进行Western Blot鉴定.方法:采用RT-PCR方法从水牛外周血白细胞中扩增bCD14基因,构建重组质粒pET28a-bCD14,转化入E coli BL21,IPTG诱导表达,对表达蛋白进行可溶性分析及Western blot鉴定.结果:bCD14基因含有一个1 122bp的开放阅读框,编码373个氨基酸;与印度水牛、挪威大鼠和人CD14的cDNA序列同源性分别为97.95%、68.78%、78.60%,氨基酸同源性分别为96.78%、61.27%、72.34%;主要以包涵体形式表达,表达蛋白经Western Blot鉴定,得到了一条约46 kD的特异性条带.结论:该文成功克隆了bCD14基因,表达了bCD14蛋白,为进一步揭示水牛抵抗革兰氏阴性菌感染的免疫机制奠定了基础.     

GST-HRB融合蛋白的表达与纯化

构建GST-HRB重组质粒,进行融合蛋白的表达、纯化及鉴定.利用PCR扩增及基因重组技术,以pcDNA-3.1-HRB为模板扩增出HRB全基因序列,并将其插入带有GST(谷胱甘肽巯基转移酶)标签的原核表达载体pGEX-6P-1中,构建GST-HRB融合蛋白表达质粒.然后,将重组质粒GST-HRB转化至大肠杆菌Rosseta进行融合蛋白的表达.利用GST琼脂糖珠进行融合蛋白的纯化,最后应用SDS-PAGE电泳和Western blotting鉴定纯化的融合蛋白.结果表明,成功构建pGEX-6P-1-HRB原核表达载体,表达及纯化了GST-HRB融合蛋白.     

A multi‐year dormancy strategy in a cabbage beetle population in southeastern China

The life cycle of the cabbage beetle Colaphellus bowringi in southeastern China is complex due to four options for adult development: summer diapause, winter diapause, prolonged diapsuse, and nondiapause. However, detailed information on the multi‐year emergence patterns of diapausing individuals in this beetle has not been documented. In this study, we monitored the adult emergence patterns of diapausing individuals and estimated the influence of the diapause‐inducing temperature and photoperiod on the incidence of prolonged diapause under seminatural conditions for several years. The duration of diapause for adults collected from the vegetable fields in different years varied from several months to 5 years. Approximately 25.9%–29.2% of individuals showed prolonged diapause (emergence more than 1 year after entering diapause) over the 5 years of observation. Furthermore, regardless of insect age, the emergence of diapausing adults from the soil always occurred between mid‐February and March in spring and between late August and mid‐October in autumn, when the host plants were available. The influence of diapause‐inducing temperatures (22, 25, and 28°C) combined with different photoperiods (L:D 12:12 h and L:D 14:10 h) on diapause duration was tested under seminatural conditions. Pairwise comparisons of diapause duration performed by the log‐rank test revealed that the low temperature of 22°C combined with the long photoperiod of L:D 14:10 h induced the longest diapause duration, whereas the low temperature of 22°C combined with the short photoperiod of L:D 12:12 h induced the highest proportion of prolonged diapause. This study indicates that C. bowringi adopts a multi‐year dormancy strategy to survive local environmental conditions and unpredictable risks.     

Newer applications of the histological stain prepared from Pterocarpus santalinus

A histological stain prepared from the heartwood of Pterocarpus santalinus Linn. has been found to be an excellent nuclear stain for various cells of animal and plant origin. As an elastic tissue stain, the results are comparable to standard elastic tissue stains. The striations of voluntary muscle fibers are well shown. The Nissl granules and fibers of cranial nerves in the pons are visualized. When counterstained with light green, it differentially stains muscle and fibrous tissue. The stain can be used as counterstain with certain histochemical procedures with satisfactory results. The preparation and use of this versatile stain are described.     

3,5-dibromo-2'-chloro-4'-isothiocyanato salicylanilide in the control of Hymenolepis nana in rats--a pilot study

The efficiency of a new anticestode compound, 3,5-dibromo-2'-chloro-4'-isothiocyanato salicylanilide (GUPTA et al. 1980) for mass eradication of hymenolepiasis from rat colonies was assessed in a pilot study. The drug was administered with milk or food. For effective eradication, depending on the size of animals, single or multiple doses of 100 to 500 mg of the drug/kg body-weight, mixed in milk, or 50 mg/kg in feed were necessary. Better results were obtained when the compound was given in feed as it ensured effective consumption irrespective of the seasonal variations. The efficacy of the compound was also assessed on cysticercoids in intermediate hosts (Tribolium confusum) by feeding them on flour medicated at 5%, 1% and 0.1% levels. None of the concentrations killed the larvae or arrested their development.
