Non‐alcoholic fatty liver disease can result in changes to drug metabolism and disposition potentiating adverse drug reactions. Furthermore, arsenite exposure during development compounds the severity of diet‐induced fatty liver disease. This study examines the effects of arsenite potentiated diet‐induced fatty liver disease on hepatic transport in male mice. Changes were detected for Mrp2/3/4 hepatic transporter gene expression as well as for Oatp1a4/2b1/1b2. Plasma concentrations of Mrp and Oatp substrates were increased in arsenic exposure groups compared with diet‐only controls. In addition, murine embryonic hepatocytes and adult primary hepatocytes show significantly altered transporter expression after exposure to arsenite alone: a previously unreported phenomenon. These data indicate that developmental exposure to arsenite leads to changes in hepatic transport which could increase the risk for ADRs during fatty liver disease.  相似文献

Polycomb group (PcG) complexes regulate cellular identity through epigenetic programming of chromatin. Here, we show that SSX2, a germline-specific protein ectopically expressed in melanoma and other types of human cancers, is a chromatin-associated protein that antagonizes BMI1 and EZH2 PcG body formation and derepresses PcG target genes. SSX2 further negatively regulates the level of the PcG-associated histone mark H3K27me3 in melanoma cells, and there is a clear inverse correlation between SSX2/3 expression and H3K27me3 in spermatogenesis. However, SSX2 does not affect the overall composition and stability of PcG complexes, and there is no direct concordance between SSX2 and BMI1/H3K27me3 presence at regulated genes. This suggests that SSX2 antagonizes PcG function through an indirect mechanism, such as modulation of chromatin structure. SSX2 binds double-stranded DNA in a sequence non-specific manner in agreement with the observed widespread association with chromatin. Our results implicate SSX2 in regulation of chromatin structure and function.  相似文献

Glycosylated haemoglobins AIa+Ib and AIc were measured serially in 10 consecutive cases of newly discovered non-acidotic diabetes before and after diet and insulin treatment. The average concentration of Hb AIc was 11.4% in untreated diabetics as compared with 4.3% in healthy controls. With prolonged optimal regulation of blood glucose Hb AIc slowly decreased to a mean concentration of 5.5%. The concentration of Hb AIc was significantly correlated with the fasting blood sugar value. The findings suggest that determining Hb AIc may give valuable information on the regulation of carbohydrate metabolism in the preceding one to two months and thus become an important aid to management.  相似文献

Abstract

SNARE (soluble N‐ethylmaleimide‐sensitive factor attachment protein receptor) proteins are the principal actors of the concluding step of membrane traffic, the fusion of a vesicle with its acceptor membrane. The specificity of their interactions has been studied mostly in vitro and is now waiting for innovative approaches to elucidate the highly complex in vivo situations with which the multicellular organisms have satisfied the need for polarized and regulated secretion or vacuolar/lysosomal/endosomal trafficking. Plants, because of their high number of SNARE genes, their complex vacuolar system and peculiarity of established models for studying polarized and regulated exocytosis (pollen tube, root hair), represent a particularly challenging system for understanding the fine details of SNARE functions. The specificity of SNARE interactions resides in their structure but is certainly modulated by the cellular environment and a number of accessory factors; consequently, only in vivo experiments will reveal the full complexity of SNARE‐regulated processes. This review will highlight the major areas of interest and the approaches to study plant SNAREs specificity in vivo.  相似文献

This study was aimed at developing a process of solid state fermentation (SSF) with the fungi Pleurotus ostreatus and Trametes versicolor on apple processing residues for wastewater decolorization. Both fungi were able to colonize apple residues without any addition of nutrients, material support or water. P. ostreatus produced the highest levels of laccases (up to 9 U g−1 of dry matter) and xylanases (up to 80 U g−1 of dry matter). A repeated batch decolorization experiment was set up with apple residues colonized by P. ostreatus, achieving 50% decolorization and 100% detoxification after 24 h, and, adding fresh wastewater every 24 h, a constant decolorization of 50% was measured for at least 1 month. A continuous decolorization experiment was set up by a packed-bed reactor based on colonized apple residues achieving a performance of 100 mg dye L−1 day−1 at a retention time of 50 h.  相似文献

Mammalian Hedgehog (HH) signalling pathway plays an essential role in tissue homeostasis and its deregulation is linked to rheumatological disorders. UBR5 is the mammalian homologue of the E3 ubiquitin-protein ligase Hyd, a negative regulator of the Hh-pathway in Drosophila. To investigate a possible role of UBR5 in regulation of the musculoskeletal system through modulation of mammalian HH signaling, we created a mouse model for specific loss of Ubr5 function in limb bud mesenchyme. Our findings revealed a role for UBR5 in maintaining cartilage homeostasis and suppressing metaplasia. Ubr5 loss of function resulted in progressive and dramatic articular cartilage degradation, enlarged, abnormally shaped sesamoid bones and extensive heterotopic tissue metaplasia linked to calcification of tendons and ossification of synovium. Genetic suppression of smoothened (Smo), a key mediator of HH signalling, dramatically enhanced the Ubr5 mutant phenotype. Analysis of HH signalling in both mouse and cell model systems revealed that loss of Ubr5 stimulated canonical HH-signalling while also increasing PKA activity. In addition, human osteoarthritic samples revealed similar correlations between UBR5 expression, canonical HH signalling and PKA activity markers. Our studies identified a crucial function for the Ubr5 gene in the maintenance of skeletal tissue homeostasis and an unexpected mode of regulation of the HH signalling pathway.  相似文献

A short-term short-scale field manipulation experiment was designed to assess the effects of total or partial defaunation, by sediment drying or diesel oil respectively, on polychaete recolonization rates in mangrove sediments of Paranaguá Bay, SE Brazil. After replacement of defaunated cores in the field, disturbed and adjacent undisturbed control sites were subsequently sampled together with adjacent undisturbed control sites on different dates over a 64-day period. Analyses of variance were carried out to test for differences in polychaete populational densities among experimental treatments and sampling dates. Examination of cores impacted by oil showed that most polychaete species kept rather active, quickly reacting to mechanical or light stimuli or moving rapidly. Polychaete recolonization of local mangroves after small-scale disturbance by oil was a fast process, in contrast to that following total defaunation by sediment drying. Rapid recolonization was largely dependent upon the differing mobility of adults of individual species from unaffected areas nearby, epifaunal crawlers or infaunal predators exhibiting the most rapid recovery rates. Conversely, the slow-paced recolonization of completely defaunated areas was due to the modification of sediment properties by drying and to the absence of larvae or juveniles of local species during the experiment.  相似文献

In this work, we identify physical and genetic interactions that implicate E3 identified by differential display (EDD) in promoting spindle assembly checkpoint (SAC) function. During mitosis, the SAC initiates a mitotic checkpoint in response to chromosomes with kinetochores unattached to spindle pole microtubules. Similar to Budding uninhibited by benzimidazoles-related 1 (BUBR1) siRNA, a bona fide SAC component, EDD siRNA abrogated G2/M accumulation in response to the mitotic destabilizing agent nocodazole. Furthermore, EDD siRNA reduced mitotic cell viability and, in nocodazole-treated cells, increased expression of the promitotic progression protein cell division cycle 20 (CDC20). Copurification studies also identified physical interactions with CDC20, BUBR1, and other components of the SAC. Taken together, these observations highlight the potential role of EDD in regulating mitotic progression and the cellular response to perturbed mitosis.  相似文献

The regulatory functions of Rab proteins in membrane trafficking lie in their ability to perform as molecular switches that oscillate between a GTP- and a GDP-bound conformation. The role of tomato LeRab11a in secretion was analyzed in tobacco protoplasts. Green fluorescent protein (GFP)/red fluorescent protein (RFP)-tagged LeRab11a was localized at the trans-Golgi network (TGN) in vivo. Two serines in the GTP-binding site of the protein were mutagenized, giving rise to the three mutants Rab11S22N, Rab11S27N and Rab11S22/27N. The double mutation reduced secretion of a marker protein, secRGUS (secreted rat beta-glucuronidase), by half, whereas each of the single mutations alone had a much smaller effect, showing that both serines have to be mutated to obtain a dominant negative effect on LeRab11a function. The dominant negative mutant was used to determine whether Rab11 is involved in the pathway(s) regulated by the plasma membrane syntaxins SYP121 and SYP122. Co-expression of either of these GFP-tagged syntaxins with the dominant negative Rab11S22/27N mutant led to the appearance of endosomes, but co-expression of GFP-tagged SYP122 also labeled the endoplasmic reticulum and dotted structures. However, co-expression of Rab11S22/27N with SYP121 dominant negative mutants decreased secretion of secRGUS further compared with the expression of Rab11S22/27N alone, whereas co-expression of Rab11S22/27N with SYP122 had no synergistic effect. With the same essay, the difference between SYP121- and SYP122-dependent secretion was then evidenced. The results suggest that Rab11 regulates anterograde transport from the TGN to the plasma membrane and strongly implicate SYP122, rather than SYP121. The differential effect of LeRab11a supports the possibility that SYP121 and SYP122 drive independent secretory events.  相似文献
