Plasma and urinary levels of "urotensin(U)-II-like" substances determined in healthy human volunteers were 12.4 +/- 0.6 ng/ml and 2.2 +/- 0.3 ng/ml by RIA, an order of magnitude lower than that seen by RRA, 167.5 +/- 9.5 ng/ml and 65.2 +/- 4.3 ng/ml. HPLC demonstrated the existence of at least three prominent activity peaks in plasma and urine, the more hydrophobic of which did not co-elute with U-II, degradation products or URP. RRA and RIA recognized these peaks with contrasting efficacy. As such, published levels of "U-II-like" activity should be interpreted with caution until a better understanding is obtained regarding what species specific RIA and RRA assay reagents interact with.  相似文献

Conifer somatic embryo germination and early seedling growth are fundamentally different than in their zygotic counterparts in that the living maternal megagametophyte tissue surrounding the embryo is absent. The megagametophyte contains the majority of the seed storage reserves in loblolly pine and the lack of the megagametophyte tissue poses a significant challenge to somatic embryo germination and growth. We investigated the differences in seed storage reserves between loblolly pine mature zygotic embryos and somatic embryos that were capable of germination and early seedling growth. Somatic embryos utilized in this study contained significantly lower levels of triacylglycerol and higher levels of storage proteins relative to zygotic embryos. A shift in the ratio of soluble to insoluble protein present was also observed. Mature zygotic embryos had roughly a 3:2 ratio of soluble to insoluble protein whereas the somatic embryos contained over 5-fold more soluble protein compared to insoluble protein. This indicates that the somatic embryos are not only producing more protein overall, but that this protein is biased more heavily towards soluble protein, indicating possible differences in metabolic activity at the time of desiccation.  相似文献

The purpose of this study was to examine the acute effect of resistance exercise (RE) on muscle androgen receptor (AR) and glucocorticoid receptor (GR) protein content. Fifteen resistance-trained men (n = 8; 21 ± 1 years, 175.3 ± 6.7 cm, 90.8 ± 11.6 kg) and women (n = 7; 24 ± 5 years, 164.6 ± 6.7 cm, 76.4 ± 15.6 kg) completed 6 sets of 10 repetitions of heavy squats. Blood samples were obtained before RE, after 3 and 6 sets of squats, and 5, 15, 30 and 70 min after RE. Muscle biopsies from the vastus lateralis were obtained before RE, and 10 min and 70 min after RE. Blood samples were analyzed for total and free testosterone concentrations and muscle samples were analyzed for AR and GR protein content. Circulating total testosterone increased significantly (p  0.05) in men and free testosterone increased in men and women with exercise. AR was significantly reduced at 70 min post-exercise in men and at 10 min post-exercise in women compared to pre-exercise. There were no changes in GR following RE, but GR was significantly higher in women compared to men. These findings support a current paradigm for stabilization followed by a reduction and then a rebound in the acute AR response to RE but demonstrate that gender differences exist in the timeline of the AR response.  相似文献

The purpose of this study was to examine changes in body composition (BC) and physical performance tests (PT) resulting from a competitive season in soccer. Twenty-five male collegiate players (age = 19.9 +/- 1.3 years; height = 177.6 +/- 6.4 cm; body mass = 77.6 +/- 8.6 kg, and percentage body fat = 12.8 +/- 5.2%) were tested before (PRE) and after (POST) the 2003-2004 National Collegiate Athletic Association season. The following tests were performed: BC (anthropometric and dual energy x-ray absorptiometry measurements), vertical jump (VJ), 9.1-m (9 m) and 36.5-m (36 m) sprint, lower-body power (LP), total body power (TP), and cardiorespiratory endurance (VO(2)max). Training was divided into soccer-specific training: field warm-up drills, practices, games, and additional conditioning sessions. A daily, unplanned, nonlinear periodization model was used to assign session volume and intensity for strength sessions (total repetitions < or =96 and workload was > or =80% of 1 repetition maximum). For the entire team, body mass significantly increased by 1.5 +/- 0.4 kg from PRE to POST due to a significant increase in total lean tissue (0.9 +/- 0.2 kg). Regionally, lean tissue mass significantly increased in the legs (0.4 +/- 0.0 kg) and trunk (0.3 +/- 0.1 kg). Physical performance variables were very similar for the entire team at PRE and POST; VJ (cm) = 61.9 +/- 7.1 PRE vs. 63.3 +/- 8.0 POST, 9.1-m (s) = 1.7 +/- 0.1 PRE and POST, 36.5-m (s) = 5.0 +/- 0.2 PRE and POST, predicted VO(2)max (ml.kg.min(-1))= 59.8 +/- 3.3 PRE vs. 60.9 +/- 3.4 POST. The only significant improvements across the season were for TP (17.3%) and for LP (10.7%). In conclusion, soccer athletes who begin a season with a high level of fitness can maintain, and in some cases improve, body composition and physical performance from before to after a competitive season. A correct combination of soccer-specific practices and strength and conditioning programs can maintain and develop physical performance, allowing a soccer athlete to perform optimally throughout pre-, in-, and postseason play.  相似文献

Compare the most common techniques of reconstruction in patients and detail benefits and risks associated with each. 3. Outline different methods of reconstruction and identify the method considered best for the patient based on timing of the procedures, body type, adjuvant therapies, and other coexisting conditions. 4. Distinguish between some of the different flaps that can be considered for autologous reconstruction. SUMMARY: Breast cancer is unfortunately a common disease affecting millions of women, often at a relatively young age. Reconstruction following mastectomy offers women an opportunity to mollify some of the emotional and aesthetic effects of this devastating disease. Although varying techniques of alloplastic and autologous techniques are available, all strive to achieve the same goal: the satisfactory reformation of a breast mound that appears as natural as possible without clothing and at the very least is normal in appearance under clothing. This article summarizes the various approaches to breast reconstruction and offers a balanced view of the risks and benefits of each, all of which in the end offer the opportunity for excellent and predictable results with a high degree of patient satisfaction.  相似文献

The purpose of this study was to verify the concurrent validity of a bar-mounted Myotest? instrument in measuring the force and power production in the squat and bench press exercises when compared to the gold standard of a computerized linear transducer and force platform system. Fifty-four men (bench press: 39-171 kg; squat: 75-221 kg) and 43 women (bench press: 18-80 kg; squat: 30-115 kg) (age range 18-30 years) performed a 1 repetition maximum (1RM) strength test in bench press and squat exercises. Power testing consisted of the jump squat and the bench throw at 30% of each subject's 1RM. During each measurement, both the Myotest? instrument and the Celesco linear transducer of the directly interfaced BMS system (Ballistic Measurement System [BMS] Innervations Inc, Fitness Technology force plate, Skye, South Australia, Australia) were mounted to the weight bar. A strong, positive correlation (r) between the Myotest and BMS systems and a high correlation of determination (R2) was demonstrated for bench throw force (r = 0.95, p < 0.05) (R2 = 0.92); bench throw power (r = 0.96, p < 0.05) (R2 = 0.93); squat jump force (r = 0.98, p < 0.05) (R2 = 0.97); and squat jump power (r = 0.91, p < 0.05) (R2 = 0.82). In conclusion, when fixed on the bar in the vertical axis, the Myotest is a valid field instrument for measuring force and power in commonly used exercise movements.  相似文献

In goats, bilateral thoracic dorsal rhizotomy (TDR) causes severe ventilatory failure during exercise, followed by progressive functional recovery. We investigated spinal neurochemical changes associated with TDR and/or functional recovery by measuring spinal concentrations of the monoamines serotonin (5-HT), norepinephrine, and dopamine via HPLC. Changes in 5-HT and calcitonin gene-related peptide were visualized with immunohistochemistry. Goat spinal cords were compared 4-15 mo after TDR from T(2) to T(12) (n = 7) with sham-operated (n = 4) or unoperated controls (n = 4). TDR increased the concentration of cervical 5-HT (C(5)-C(6); 122% change), caudal thoracic norepinephrine (T(7)-T(11); 53% change), and rostral thoracic dopamine (T(3)-T(6); 234% change). TDR increased 5-HT-immunoreactive terminal density (dorsal and ventral horns) and nearly eliminated calcitonin gene-related peptide immunoreactivity in the superficial laminae of the dorsal horn in rostral thoracic segments; both effects became less pronounced in caudal thoracic segments. Thus TDR elevates monoamine concentrations in discrete spinal regions, including possible compensatory changes in descending serotonergic inputs to spinal segments not directly affected by TDR (i.e., cervical) but associated with functionally related motor nuclei (i.e., phrenic nucleus).  相似文献

Using immunocytochemical techniques, we report here direct evidence of kynurenine (Kyn) presence and localization in the lung and brain. Kyn is a metabolite of tryptophan and 5-hydroxytryptophan, produced by indoleamine 2,3-dioxygenase (IDO). Whereas IDO has been quantitated in tissues from lung, brain, and other organs, Kyn has only been identified in brain (by HPLC), and its specific localization has not been determined. We reacted alternate serial paraffin sections with antisera raised in rabbits against a L-Kyn-albumin conjugate, and with anti-5HT (serotonin, 5-hydroxytryptamine), using the PAP method. Kyn-like immunoreactivity in the lung was specifically localized to cells of the bronchiolar epithelium resembling basal cells. Taller epithelial cells in the bronchi and dorsal trachea were likewise positive whereas neuroepithelial bodies were negative. Immunoreactivity in the brain was typically localized to cells localized in the ependyma of the walls of all ventricles, and to nerve fibers. The cellular Kyn-like reactivity was totally separate from that of anti-5HT, the latter uniquely staining argyrophil lung neuroendocrine cells and raphae neurons of the brain. Our findings suggest a route of tryptophan metabolism in the lung and brain alternate to the common pathway leading to 5-hydroxyindoleacetic acid via 5-HT. This route is of physiologic and pathologic significance as many metabolites are pharmacologically active.  相似文献
