Porphyromonas gingivalis is a gram-negative, anaerobic coccobacillus that has been implicated as a major etiological agent in the development of chronic periodontitis. In this paper, we report the characterization of a protein, IhtB (iron heme transport; formerly designated Pga30), that is an outer membrane hemin-binding protein potentially involved in iron assimilation by P. gingivalis. IhtB was localized to the cell surface of P. gingivalis by Western blot analysis of a Sarkosyl-insoluble outer membrane preparation and by immunocytochemical staining of whole cells using IhtB peptide-specific antisera. The protein, released from the cell surface, was shown to bind to hemin using hemin-agarose. The growth of heme-limited, but not heme-replete, P. gingivalis cells was inhibited by preincubation with IhtB peptide-specific antisera. The ihtB gene was located between an open reading frame encoding a putative TonB-linked outer membrane receptor and three open reading frames that have sequence similarity to ATP binding cassette transport system operons in other bacteria. Analysis of the deduced amino acid sequence of IhtB showed significant similarity to the Salmonella typhimurium protein CbiK, a cobalt chelatase that is structurally related to the ATP-independent family of ferrochelatases. Molecular modeling indicated that the IhtB amino acid sequence could be threaded onto the CbiK fold with the IhtB structural model containing the active-site residues critical for chelatase activity. These results suggest that IhtB is a peripheral outer membrane chelatase that may remove iron from heme prior to uptake by P. gingivalis.  相似文献

The incidence of recurrence after endoscopic carpal tunnel release   总被引:5,自引:0,他引:5  
Endoscopic carpal tunnel release has been used to decompress the median nerve in carpal tunnel syndrome for over the past decade, with an advantage (over the traditional "open" release) being decreased pain in the postoperative period. The goals of this study were to attempt to define the recurrence rate after endoscopic carpal tunnel release and to determine if it differs from that of open technique. The charts of 191 consecutive carpal tunnel syndrome patients treated operatively at the University of Missouri were reviewed. For this study, recurrent carpal tunnel syndrome was defined as documented cases in which the symptoms had resolved following surgical release but subsequently recurred, requiring surgical rerelease of the carpal tunnel. All endoscopic releases were performed using the Chow two-portal technique. Statistical analysis was performed using Fisher's exact test. A total of 103 patient hands had open carpal tunnel releases; 88 were endoscopically released. Total follow-up time (from the initial release) averaged 29 months for the open group and 22 months for the endoscopic group. There were no recurrences in the open group and six recurrences in the endoscopic group (7 percent, p = 0.008). All six recurrences were in worker's compensation patients. The median time between endoscopic release and rerelease was 8.5 months. There seems to be a statistically higher incidence of recurrence of carpal tunnel syndrome after endoscopic release compared with the traditional "open" release in our cases. Although the pathogenesis of this increased rate of recurrence is not clear, this should be considered when planning surgical release of the volar carpal ligament for carpal tunnel syndrome.  相似文献

Lysophosphatidylcholine (LPC) is the major bioactive lipid component of oxidized LDL, thought to be responsible for many of the inflammatory effects of oxidized LDL described in both inflammatory and endothelial cells. Inflammation-induced transformation of vascular smooth muscle cells from a contractile phenotype to a proliferative/secretory phenotype is a hallmark of the vascular remodeling that is characteristic of atherogenesis; however, the role of LPC in this process has not been fully described. The present study tested the hypothesis that LPC is an inflammatory stimulus in coronary artery smooth muscle cells (CASMCs). In cultured human CASMCs, LPC stimulated time- and concentration-dependent release of arachidonic acid that was sensitive to phospholipase A2 and C inhibition. LPC stimulated the release of arachidonic acid metabolites leukotriene-B4 and 6-keto-prostaglandin F, within the same time course. LPC was also found to stimulate basic fibroblast growth factor release as well as stimulating the release of the cytokines GM-CSF, IL-6, and IL-8. Optimal stimulation of these signals was obtained via palmitic acid-substituted LPC species. Stimulation of arachidonic acid, inflammatory cytokines and growth factor release, implies that LPC might play a multifactorial role in the progression of atherosclerosis, by affecting inflammatory processes.  相似文献

The purpose of this study was to determine the impact of a very slow (VS) velocity and a self-selected volitional (VOL) velocity at varying intensities on repetition number, peak force, peak power, and total volume in the squat and shoulder press exercises. On separate testing days, 9 resistance trained men (age: 23.9 +/- 2.5 years; height: 174.8 +/- 6.5 cm; body mass: 80.1 +/- 12.4 kg) performed a squat (SQ) and shoulder press (SP) exercise at 60 or 80% of 1 repetition maximum (1RM) at either VOL or VS (10-second eccentric and 10-second concentric actions) velocity for as many repetitions as possible. Force, power, and volume (repetitions x kg) were also determined. Subjects performed significantly fewer repetitions (p < or = 0.05) in the VS exercises (60% VS SQ 5 +/- 1 vs. VOL SQ 24 +/- 2; 80% VS SQ 2 +/- 0 vs. VOL SQ 12 +/- 1; 60% VS SP 4 +/- 1 vs. VOL SP 14 +/- 2; 80% VS SP 1 +/- 0 vs. VOL SP 6 +/- 1). Peak force and power were significantly higher at the VOL speed (peak force [in newtons]: 60% VS SQ 564.4 +/- 77.3 vs. VOL SQ 1229.0 +/- 134.9 N; 80% VS SQ 457.3 +/- 27.9 vs. VOL SQ 1059.3 +/- 104.7 N; 60% VS SP 321.6 +/- 37.8 vs. VOL SP 940.7 +/- 144.8 N; 80% VS SP 296.5 +/- 24.7 vs. VOL SP 702.5 +/- 57.7 N; and peak power [in watts]: 60% VS SQ 271.2 +/- 40.1 vs. VOL SQ 783.2 +/- 129.1 W; 80% VS SQ 229.3 +/- 49.5 vs. VOL SQ 520.2 +/- 85.8 W; 60% VS SP 91.3 +/- 21.9 vs. VOL SP 706.6 +/- 151.4 W; 80% VS SP 78.1 +/- 19.8 vs. VOL SP 277.6 +/- 46.4 W). VOL speed elicited higher total volume than the VS velocity. The beneficial role of carbohydrate (CHO) supplementation in endurance exercise is well documented. However, only few data are available on the effects of CHO loading on resistance exercise performance. Because of the repetitive use of high-threshold motor units, it was hypothesized that the power output (power-endurance) of multiple sets of jump squats would be enhanced following a high-CHO (6.5 g CHO kg body mass(-1)) diet compared to a moderate-CHO (4.4 g CHO kg body mass(-1)) diet. Eight healthy men (mean +/- SD: age 26.3 +/- 2.6 years; weight 73.0 +/- 6.3 kg; body fat 13.4 +/- 5.0%; height 178.2 +/- 6.1 cm) participated in 2 randomly assigned counterbalanced supplementation periods of 4 days after having their free-living habitual diet monitored. The resistance exercise test consisted of 4 sets of 12 repetitions of maximal-effort jump squats using a Plyometric Power System unit and a load of 30% of 1 repetition maximum (1RM). A 2-minute rest period was used between sets. Immediately before and after the exercise test, a blood sample was obtained to determine the serum glucose and blood lactate concentrations. Resistance exercise intensity is commonly prescribed as a percent of 1 repetition maximum (1RM). However, the relationship between percent 1RM and the number of repetitions allowed remains poorly studied, especially using free weight exercises. The purpose of this study was to determine the maximal number of repetitions that trained (T) and untrained (UT) men can perform during free weight exercises at various percentages of 1RM. Eight T and 8 UT men were tested for 1RM strength. Then, subjects performed 1 set to failure at 60, 80, and 90% of 1RM in the back squat, bench press, and arm curl in a randomized, balanced design. There was a significant (p < 0.05) intensity x exercise interaction. More repetitions were performed during the back squat than the bench press or arm curl at 60% 1RM for T and UT. At 80 and 90% 1RM, there were significant differences between the back squat and other exercises; however, differences were much less pronounced. No differences in number of repetitions performed at a given exercise intensity were noted between T and UT (except during bench press at 90% 1RM). In conclusion, the number of repetitions performed at a given percent of 1RM is influenced by the amount of muscle mass used during the exercise, as more repetitions can be performed during the back squat than either the bench press or arm curl. Bacterial populations can use bet‐hedging strategies to cope with rapidly changing environments. One example is non‐growing cells in clonal bacterial populations that are able to persist antibiotic treatment. Previous studies suggest that persisters arise in bacterial populations either stochastically through variation in levels of global signalling molecules between individual cells, or in response to various stresses. Here, we show that toxins used in contact‐dependent growth inhibition (CDI) create persisters upon direct contact with cells lacking sufficient levels of CdiI immunity protein, which would otherwise bind to and neutralize toxin activity. CDI‐mediated persisters form through a feedforward cycle where the toxic activity of the CdiA toxin increases cellular (p)ppGpp levels, which results in Lon‐mediated degradation of the immunity protein and more free toxin. Thus, CDI systems mediate a population density‐dependent bet‐hedging strategy, where the fraction of non‐growing cells is increased only when there are many cells of the same genotype. This may be one of the mechanisms of how CDI systems increase the fitness of their hosts.  相似文献

One hundred psychiatric patients were treated with chlorpromazine, alone or combined with reserpine. Fifty-six per cent of patients with chronic schizophrenic reactions showed moderate or pronounced improvement when treated with chlorpromazine alone. The results of treatment with the combined drugs were not so good as that. Indications are that treatment of patients with chronic schizophrenic reactions is more efficacious with these drugs than with other forms of somatic therapy.Complications of treatment were far greater with combined use of chlorpromazine and reserpine. For this reason, the combination appears to have limited usefulness. The Parkinson syndrome was the most frequent complication of large doses of these drugs. It appears to be a toxic reaction, requiring reduction in dosage. Jaundice appears to be neither a frequent nor a serious complication of treatment.  相似文献

Neurosecretory pathways were examined in normal male rats by the use of the immunoperoxidase technique for the localization of neurophysin in Bouin-fixed, deparaffinized sections. Using this technique two projections of extrahypothalamic neuorosecretory fibers can be traced to the sites of origin of the choid plexuses of both horns of the lateral ventricles. Neurophysin-containing axons originating primarily from the paraventricular field course dorsolaterad to enter the choroid fissure of the dorsal horn. A caudally direced group of fibers course ventrolaterad to enter the ventral horn choroid fissures. The supraoptic nuclear field is the primary contributor to the latter group. Scattered neurosecretory neurons are found in association with both pathways, usually in contact with blood vessels supplying the choroid plexuses, or in the telencephalic subependymal stroma. Neurosecretory fibers and terminals occur within the choroid fissures and juxtaventricular neuropil. The neurosecretory terminals in the choroid fissures appear as Herring-body type neurohemal organs; in the neuropil they appear as punctate peri-neuronal desities suggestive of synaptic contacts. Thes morphologic findings are discussed in relation to reports indicating the presence of antidiuretic, vasopressin-like activity in cerebrospinal fluid and choroid plexus extraxts together with ultrastructural evidence supportive of vasopressin-mediated transchoroidal cerebrospinal fluid absorption. These results and those of others indicated the possible involvement of the neurosecretory system in the regulation of brain interstitial-ventricular cerebrospinal fluid dynamics.  相似文献
