A series of substituted guanidine derivatives were prepared and evaluated as potent and selective inhibitors of mitochondrial F(1)F(0) ATP hydrolase. The initial thiourethane derived lead molecules possessed intriguing in vitro pharmacological profiles, though contained moieties considered non-drug-like. Analogue synthesis efforts led to compounds with maintained potency and superior physical properties. Small molecules in this series which potently and selectivity inhibit ATP hydrolase and not ATP synthase may have utility as cardioprotective agents. 相似文献

The G2/M checkpoint is an attractive pathway for targeting and sensitizing tumor cells to cancer treatment. Abrogation of
the G2/M checkpoint by targeting molecules, such as checkpoint kinase 1 (chk1), increases DNA breakage and sensitizes tumor
cells to anti-tumoral agents. However, most of the previously described G2/M abrogators are actually targeting the G2-M border
checkpoints rather than mitotic checkpoints. This prompted us to test the effects of combined targeting of chk1 and a critical
regulator of mitosis, polo-like kinase 1 (plk1). Chk1 and plk1 were found to be co-expressed in 70% of primary neoplastic
tissues we examined. Asynchronized tumor cells were treated with different DNA damaging-agents to activate G1/S, S or G2/M
checkpoints. Either chk1 or plk1-specific antisense oligodeoxynucleotides (ASODN) enhanced DNA damaging agent-induced apoptosis.
When used in combination, however, chk1- plus plk1-specific ASODN failed to produce synergistic effects. Moreover, selective
targeting of plk1 or chk1 in tumor xenografts of mice by oncolytic adenovirus mutants demonstrated potent anti-tumoral efficacy
in the presence of low dose cisplatin. Again, combined targeting of chk1 and plk1 did not further enhance anti-tumoral efficacy.
We concluded that combined targeting of chk1 and plk1 was not superior to either targeting chk1 or plk1 alone, which suggested
that chk1 and plk1 silencing might overlap in their mechanism of action. Whether combined targeting of chk1 with other, more
specific mitotic regulators would synergistically sensitize tumor to anti-neoplastic therapeutics needs to be further clarified.

Discoidin domain receptor 2 (DDR2) is a kind of protein tyrosine kinases associated with cell proliferation and tumor metastasis,
and collagen, identified as a ligand for DDR2, up-regulates matrix metallloproteinase 1 (MMP-1) and MMP-2 expression in cellular
matrix. To investigate the roles of DDR2 in destruction of cartilage in rheumatoid arthritis (RA) and tumor metastasis, we
tried to express extracellular domain of DDR2 fused with a His tag to increase protein solubility and facilitate purification
(without signal peptide and transmembrane domain, designated DR) in Pichia pastoris, purify the expressed protein, and characterize its function, for purpose of future application as a specific DDR2 antagonist.
Two clones of relative high expression of His-DR were obtained, After purification by a Ni-NTA (nitric-tri-acetic acid) chromatographic
column, soluble fused His-DR over 90% purity were obtained. Competitive binding inhibition assay demonstrated that expressed
His-DR could block the binding of DDR2 and natural DDR2 receptors on NIT3T3 and synovial cell surfaces. Results of RT-PCR,
Western blotting, and gelatinase zymography showed that His-DR was capable of inhibiting MMP-1 and MMP-2 secretion from NIT3T3
cells and RA synoviocytes stimulated by collagen II. For MMP-1, the inhibitory effect was displayed at the levels of mRNA
and protein, whereas for MMP-2 it was demonstrated at the level of protein physiological activity. All these findings suggested
that the fused expressed His-DR inhibited the activity of natural DDR2, and relevant MMP-1 and MMP-2 expression in synoviocytes
and NIH3T3 cells provoked by collagen II.

Endostatin, a 20-kDa C-terminal fragment derived from type XVIII collagen, is a potent angiogenesis inhibitor and an antitumor
factor. To improve the production of recombinant human endostatin on increasing demand in clinical practice, we constructed
an artificial gene encoding its mature peptide sequence in human collagen XVIII. The synthetic gene consisted of 20 codons
in preference in methylotropic yeast—Pichia pastoris and was cloned into expression vector pPICZαA; and the recombinant protein was expressed in P. pastoris strain SMD1168 and purified to near homogeneity using heparin affinity chromatography. The amount of expressed recombinant
protein in cultural media using described strategy was 80 mg/l in shake flask cultivation and 435 mg/l in high-density bioreactor
fermentation. Methylthiazolium assay demonstrated that human endostatin expressed in P. pastoris using artificial synthetic gene of preference in P. pastoris was able to inhibit the acidic fibroblast growth factor-induced proliferation of endothelial cells in vitro. 相似文献

Previous studies showed that nitricoxide synthase (NOS) and oxidative stress can induce skeletal muscle atrophy in the muscular
dystrophy and inclusion-body myopathy. There is a correlation between NOS and oxidative stress. However, it is not clear,
whether there are some changes of the NOS activity in prolonged alcoholic myopathy (PAM), and whether NOS activity has relation
to amyotrophy of PAM. We established experimental alcoholic myopathy model of rats by prolonged alcohol intake. We found that
there is a reduction in GSH-px (P < 0.05) and an increase of SOD (P < 0.05), MDA (P < 0.05) and iNOS (P < 0.05) in the plantaris of the experimental group by spectrophotometer. In the soleus of the experimental group, except
for MDA showed an increase (P < 0.05), the other enzymes showed no obvious difference (P > 0.05). The immunohistochemistry results showed that there was obvious expression of iNOS in the cytoplasm of plantaris
in the experimental group and there was no expression of iNOS in the control group. There was a decrease of nNOS expression
on the membranes of the plantaris cells in the experimental group by immunofluorescence. Meanwhile, we found the expression
of nNOS in some cytoplasm. Our results suggested that NOS might be an important factor during the development of PAM. We could
infer that there are some disturbances with regard to output and scavenging of free radical in PAM. Alcohol can induce the
oxidative stress reaction and further result in imbalance of the oxidant-antioxidant status in the organism.

The single channel properties of cloned P2X2 purinoceptors expressed in human embryonic kidney (HEK) 293 cells and Xenopus oocytes were studied in outside-out patches. The mean single channel current-voltage relationship exhibited inward rectification in symmetric solutions with a chord conductance of approximately 30 pS at -100 mV in 145 mM NaCl. The channel open state exhibited fast flickering with significant power beyond 10 kHz. Conformational changes, not ionic blockade, appeared responsible for the flickering. The equilibrium constant of Na+ binding in the pore was approximately 150 mM at 0 mV and voltage dependent. The binding site appeared to be approximately 0.2 of the electrical distance from the extracellular surface. The mean channel current and the excess noise had the selectivity: K+ > Rb+ > Cs+ > Na+ > Li+. ATP increased the probability of being open (Po) to a maximum of 0.6 with an EC50 of 11.2 microM and a Hill coefficient of 2.3. Lowering extracellular pH enhanced the apparent affinity of the channel for ATP with a pKa of approximately 7.9, but did not cause a proton block of the open channel. High pH slowed the rise time to steps of ATP without affecting the fall time. The mean single channel amplitude was independent of pH, but the excess noise increased with decreasing pH. Kinetic analysis showed that ATP shortened the mean closed time but did not affect the mean open time. Maximum likelihood kinetic fitting of idealized single channel currents at different ATP concentrations produced a model with four sequential closed states (three binding steps) branching to two open states that converged on a final closed state. The ATP association rates increased with the sequential binding of ATP showing that the binding sites are not independent, but positively cooperative. Partially liganded channels do not appear to open. The predicted Po vs. ATP concentration closely matches the single channel current dose-response curve. 相似文献

Compsilura concinnata (Meigen) is one of the most famous, most polyphagous and most widely distributed tachinid flies (Diptera, Tachinidae) in the world. This species is well known as a biocontrol agent of some injurious pests of cultural and wild plants and has been introduced from Europe to the United States to control mainly the gypsy moth. Recently we found three new species very closely resembling C. concinnata from Southeast and East Asia: C. lobata sp. nov. (Japan and Thailand), C. malayana sp. nov. (Malaysia) and C. pauciseta sp. nov. (Japan and Taiwan). Additionally, C. samoaensis Malloch is treated as a junior synonym of C. concinnata based on the examination of the type specimen. The genetic differences in the mitochondrial COI gene data are examined to assess the accuracy of species delimitation of Compsilura. The male postabdominal characters of these species are illustrated. The piercing female postabdomen of C. concinnata is illustrated and compared to those of other members belonging to the Blondelia group including Blondelia Robineau-Desvoidy, Celatoria Coquillett, Eucelatoria Townsend and Vibrissina Rondani. 相似文献

Primary ciliary dyskinesia (PCD) is a group of genetically and clinically heterogeneous disorders with motile cilia dysfunction. It is clinically characterized by oto-sino-pulmonary diseases and subfertility, and half of the patients have situs inversus (Kartagener syndrome). To identify the genetic cause in a Han-Chinese pedigree, whole-exome sequencing was conducted in the 37-year-old proband, and then, Sanger sequencing was performed on available family members. Minigene splicing assay was applied to verify the impact of the splice-site variant. Compound heterozygous variants including a splice-site variant (c.1974-1G>C, rs1359107415) and a missense variant (c.7787G>A, p.(Arg2596Gln), rs780492669), in the dynein axonemal heavy chain 11 gene (DNAH11) were identified and confirmed as the disease-associated variants of this lineage. The minigene expression in vitro revealed that the c.1974-1G>C variant could cause skipping over exon 12, predicted to result in a truncated protein. This discovery may enlarge the DNAH11 variant spectrum of PCD, promote accurate genetic counselling and contribute to PCD diagnosis. 相似文献
