The wings of some insect species are known to fluoresce under illumination by ultraviolet light. Their fluorescence properties are however, not comprehensively documented. In this article, the optical properties of one specific insect, the Trictenotoma childreni yellow longhorn beetle, were investigated using both linear and nonlinear optical (NLO) methods, including one‐ and two‐photon fluorescence and second harmonic generation (SHG). These three distinct optical signals discovered in this beetle are attributed to the presence of fluorophores embedded within the scales covering their elytra. Experimental evidence collected in this study indicates that the fluorophores are non‐centrosymmetric, a fundamental requirement for SHG. This study is the first reported optical behavior of this type in insects. We described how NLO techniques can complement other more convenient approaches to achieve a more comprehensive understanding of insect scales and integument properties. 相似文献

Telomeres are repetitive nucleoprotein structures at the ends of chromosomes. Like most genomic regions consisting of repetitive DNA, telomeres are fragile sites prone to replication fork stalling and generation of chromosomal instability. In particular, abrogation of the TRF1 telomere binding protein leads to stalled replication forks and aberrant telomere structures known as “multitelomeric signals”. Here, we report that TRF1 deficiency also leads to the formation of “ultra-fine bridges” (UFB) during mitosis, and to an increased time to complete mitosis mediated by the spindle assembly checkpoint proteins (SAC). We find that topoisomerase IIα (TopoIIα), an enzyme essential for resolution of DNA replication intermediates, binds telomeres in a TRF1-mediated manner. Indeed, similar to TRF1 abrogation, TopoIIα downregulation leads to telomere fragility and UFB, suggesting that these phenotypes are due to decreased TopoIIα at telomeres. We find that SAC proteins bind telomeres in vivo, and that this is disrupted upon TRF1 deletion. These findings suggest that TRF1 links TopoIIα and SAC proteins in a pathway that ensures correct telomere replication and mitotic segregation, unveiling how TRF1 protects from telomere fragility and mitotic defects. 相似文献

RFLP-based genetic map of rye, developed previously using a cross of lines DS2×RXL10 (F2 generation), was extended with 69 RAPD and 12 isozyme markers. The actual map contains 282 markers dispersed on all seven
chromosomes and spans a distance of 1,140 cM. The efficiency of mapping RAPD markers was close to ten loci per 100-screened
arbitrary primers. A strong selection of polymorphic, intensive and reproducible fragments was necessary to reveal individual
marker loci that could be assigned to rye chromosomes. Newly mapped markers cover a substantial part of the rye genome and
constitute a valuable tool suitable for map saturation, marker-aided selection and phenetic studies. A specific nomenclature
for the RAPD loci mapped on individual rye chromosomes, which could be helpful in managing of accumulating data, is proposed.

Populations of Prosopis glandulosa var. torreyana in the Chihuahuan desert have a fixed dimorphic system of nectar production in which half the individuals produce nectar (are nectarful) and the other half are nectarless. We analyzed the impact of nectar production on different estimates of fitness, comparing nectarful against nectarless individuals in size, mating system, seed traits, and fruit set in a 1-ha scrubland. Of the reproductive individuals (358), 46% were nectarful and 54% were nectarless. Neither tree size nor flowering phenology differed between nectar morphs. Fixation indices (F) for both progeny (F = −0.2) and adults (F = −0.45) were negative, and high heterozygosities were found in adults and progeny (H = 0.45). No differences were found between nectar morphs for F, H, and single (ts = 1.1) and multilocus (tm = 1.03) outcrossing rates. Controlled pollinations showed differences between selfing and control treatments with no differences between nectar morphs. Nectarless individuals produced significantly more pollen grains than did nectar producers, but all other measured floral traits showed no differences. Nectarful trees were visited by pollinators 21 times more often and had a significantly higher overall fruit set than did nectarless trees. No differences between nectar morphs in seed mass or in percentage seed germination were found, but heavier seeds tended to have higher heterozygosities. Both morphs had similar success as females, but nectarless trees had ∼7% higher male function. We discuss three possible scenarios for the evolution of the fixed dimorphism in nectar production, two involving unstable phases (substitution of one morph by the other, and evolution towards dioecy) and one stable scenario (maintenance of the dimorphic system). 相似文献

The reproductive biology of Cryptogramma crispa, a tetraploid species with a broad circumboreal and alpine distribution, growing mainly in siliceous boulder fields and crevices, was studied in the laboratory by growing gametophytes in plates with both solidified agar media and sterilized soil. In addition, an electrophoretic study of isozymes was carried out on frond samples from five natural populations, as an additional source of evidence concerning the breeding system and the genetic structure of sporophyte populations. Populations throughout the Iberian range of the species were selected for this study, and a Scottish population was included to represent plants from outside our local area and ecology. The morphological development of gametophytes is of the Adiantum type. All multispore cultures developed into a bigametophytic system, consisting in most cases of male and female prothalli. This pattern of sexual expression provides evidence for outcrossing as the main breeding system in this species. Moreover, there is good evidence that the species possesses an antheridiogen system to promote outcrossing. The long time needed by gametophytes to produce gametangia, and afterwards to fertilize and produce sporophytes, might be the primary reason why so few young sporophytes are found in the wild. The values of the percentage of polymorphic loci and the similarity levels obtained from the isozyme analyses indicate a level of genetic variability that would be expected in an outcrossing species. Long-lived perennials are a species-rich, ecologically important component of the North American deserts, yet we know little about their genetic structure, information important for their conservation. Agave victoriae-reginae is an endemic of the Chihuahuan Desert of northern Mexico that is endangered by collection for the ornamental trade. We examined levels and patterns of variation at ten polymorphic allozyme loci in ten populations representing the range of the species. Levels of genetic variation (mean He= 0.335) and differentiation (mean FST = 0.236) were high. Phenetic clustering suggested the existence of at least three distinct groups of populations. If this pattern of variation is representative of other long-lived desert perennials, it may explain the species richness of this group and will pose a real challenge to gene conservation efforts. 相似文献

At fixed lung volume (VL), alterations in surface tension change alveolar surface area (S) and lung recoil (PL). Wilson (26), using data from fixed lungs (1, 9), quantified the isovolume change in S with PL. We reexamined this question in fresh excised rabbit lungs, with two important differences. First, we measured fractional changes in S by using diffuse light scattering, avoiding the potential upset of the balance of tissue and surface forces during fixation. Second, we altered surface tension by ventilating the lungs with nebulized polydimethylsiloxane, with much less residual fluid compared with lavage. We found that S decreased at low and mid VL (treatment surface tension > control) by about half of Wilson's estimates and was nearly unaffected by treatment at high VL. This suggests that with increased surface tension there is 1) greater septal retraction in lungs fixed by vascular perfusion compared with unfixed lungs and 2) a greater increase in PL and less loss of S than would have been predicted. 相似文献

We present a general framework for modelling adaptive trait dynamics in which we integrate various concepts and techniques from modern ESS-theory. The concept of evolutionarily singular strategies is introduced as a generalization of the ESS-concept. We give a full classification of the singular strategies in terms of ESS-stability, convergence stability, the ability of the singular strategy to invade other populations if initially rare itself, and the possibility of protected dimorphisms occurring within the singular strategy's neighbourhood. Of particular interest is a type of singular strategy that is an evolutionary attractor from a great distance, but once in its neighbourhood a population becomes dimorphic and undergoes disruptive selection leading to evolutionary branching. Modelling the adaptive growth and branching of the evolutionary tree can thus be considered as a major application of the framework. A haploid version of Levene's soft selection model is developed as a specific example to demonstrate evolutionary dynamics and branching in monomorphic and polymorphic populations. 相似文献
