In species acting as hosts of infectious agents, the extent of gene flow between populations is of particular interest because the expansion of different infectious diseases is usually related to the dispersal of the host. We have estimated levels of gene flow among populations of the sigmodontine rodent Oligoryzomys flavescens, in which high titers of antibodies have been detected for a Hantavirus in Argentina that produces a severe pulmonary syndrome. Enzyme polymorphism was studied by means of starch gel electrophoresis in 10 populations from the area where human cases of Hantavirus have occurred. Genetic differentiation between populations was calculated from FST values with the equation Nm = [(1/FST−1]/4. To assess the relative importance of current gene flow and historical associations between populations, the relationship of population pairwise log Nm and log geographic distance was examined. Low FST (mean = 0.038) and high Nm (15.27) values suggest high levels of gene flow among populations. The lack of an isolation by distance pattern would indicate that this species has recently colonized the area. The northernmost population, located on the margin of a great river, shows very high levels of gene flow with the downstream populations despite the large geographic distances. Passive transport of animals down the river by floating plants would promote unidirectional gene flow. This fact and the highest mean heterozygosity of that northernmost population suggest it is a center of dispersal within the species' range. Chronic obstructive pulmonary disease (COPD) is a complex disease influenced by genetic and environmental factors. Cystic fibrosis transmembrane conductance regulator (CFTR) protein is an important component of the lung tissue homeostasis, involved in the regulation of the rate of mucociliary clearance. As it is known that certain CFTR variants have consequences on the function of CFTR protein, the aim of this study was to examine the possible role of F508del, M470V, Tn locus, and R75Q variants in COPD development and modulation. Total number of 86 COPD patients and 102 control subjects were included in the study. Possible association between COPD susceptibility, severity, and onset of the disease and allele or genotype of four analyzed CFTR variants was examined. No associations were detected between COPD development, onset of the disease and tested CFTR alleles and genotypes. However, VV470 genotype was associated with mild/moderate COPD stages in comparison to severe/very severe ones (OR = 0.29, 95%CI = 0.11-0.80, p = 0.016). Our study showed that patients with VV470 genotype had a 3.4-fold decreased risk for the appearance of severe/very severe COPD symptoms, and the obtained results indicate that this genotype may have a protective role. These results also suggest the importance of studying CFTR gene as a modifier of this disease.  相似文献

Knowledge of cell mechanical properties, such as elastic modulus, is essential to understanding the mechanisms by which cells carry out many integrated functions in health and disease. Cellular stiffness is regulated by the composition, structural organization, and indigenous mechanical stress (or prestress) borne by the cytoskeleton. Current methods for measuring stiffness and cytoskeletal prestress of living cells necessitate either limited spatial resolution but with high speed, or spatial maps of the entire cell at the expense of long imaging times. We have developed a novel technique, called biomechanical imaging, for generating maps of both cellular stiffness and prestress that requires less than 30 s of interrogation time, but which provides subcellular spatial resolution. The technique is based on the ability to measure tractions applied to the cell while simultaneously observing cell deformation, combined with capability to solve an elastic inverse problem to find cell stiffness and prestress distributions. We demonstrated the application of this technique by carrying out detailed mapping of the shear modulus and cytoskeletal prestress distributions of 3T3 fibroblasts, making no assumptions regarding those distributions or the correlation between them. We also showed that on the whole cell level, the average shear modulus is closely associated with the average prestress, which is consistent with the data from the literature. Data collection is a straightforward procedure that lends itself to other biochemical/biomechanical interventions. Biomechanical imaging thus offers a new tool that can be used in studies of cell biomechanics and mechanobiology where fast imaging of cell properties and prestress is desired at subcellular resolution.  相似文献

Monte-Carlo simulations were performed in order to quantify the microbiological risk arising from the presence of Giardia and Cryptosporidium in drinking water. A two-year survey of 45 utilities located on a portion of the St. Lawrence River (Quebec, Canada) allowed us to collect the information required for this assessment: parasite occurrence data, treatment performances and daily unboiled water consumption. For a conventional treatment, the mean annual risk of infection with Cryptosporidium was found to be approximately eight times higher than with Giardia. The Monte-Carlo simulations confirm that the clarification process plays an important role in the reduction of the health risk associated with protozoan parasites. In fact, in the case of Cryptosporidium, the impact of chemical disinfectants (O3 or Cl2) appears to be minimal in the cold water treatment conditions prevailing in Quebec. According to the Monte-Carlo simulations, 45% (for conventional treatment+O3) and 51% (for conventional treatment) of individuals are exposed to a risk of infection with Cryptosporidium higher than the proposed USEPA guideline (1 infection per 10[emsp4 ]000 individuals per year).  相似文献

The effects of mature benthivorous cyprinid fish and theirrecruitment on sediment resuspension, turbidity, phyto- andzooplankton, and benthic macroinvertebrates were studied in fourexperimental ponds. The ponds were stocked with bream (Abramisbrama L.), white bream (Blicca bjoerkna L.), roach (Rutilus rutilus L.) and wild carp (Cyprinus carpio L.) of3+–5+ age classes at standing crop biomass varying from 0 to500 kg ha-1. Cyprinids caused an increase in sedimentresuspension and in turbidity, in proportion to their biomass. Meancrustacean biomass did not significantly affect phytoplanktonbiomass due to intense grazing by fish during spring. Ponds withhigh fish stocks showed reduced midge biomass and vegetation coverand increased biomass of predatory invertebrates.  相似文献

The tapeworm Proteocephalus macrocephalus (Creplin, 1825), a parasite of the eel Anguilla anguilla, was characterised using morphological and genetic characters. The species is morphologically typified mainly by features of the scolex (i.e. shape and size of an apical organ which is formed early during the development in an intermediate host) and strobila (i.e. regularly rectangular proglottides, morphology of the vagina and anterior position of the seminal receptacle). Genetically, P. macrocephalus was compared with two congeners, P. percae and P. exiguus. Characters enabling differentiation of P. macrocephalus were as follows: fixed allelic differences at Got, Pgm and Gapdh loci, metrical features of chromosomes Nos 1, 2 and 7, and species-specific fragments obtained by RAPD method using 13 random primers.  相似文献

The effect of in-place regeneration of protein A adsorbents on protein adsorption characteristics is investigated. Regeneration with sodium hydroxide and time of exposure determined the protein capacity of the adsorbent, but no effect was observed on the adsorbent protein affinity. Fixed-bed adsorption of human immunoglobulin G was studied. Breakthrough curves were measured for protein adsorption on fixed-bed columns. These data were analyzed by a simple kinetic model to determine the rate constants for the adsorption process. It was found that forward adsorption rate constant remained constant along the chemical treatment exposure time. Protein A adsorbent selectivity was determined using mouse serum immunoglobulins G 1 and G 3 . Column linear gradient elution showed that adsorbent selectivity decreased with the exposure time chemical treatment. The implications of these results on the design and optimization of protein A chromatographic process are discussed.  相似文献

We addressed the following questions: (a) May different seed bank functional types (SBFT), defined on the basis of singularities of their persistence in soil, be identified in the Festuca pallescens grasslands? (b) Do different above-ground vegetation states modify the expression of SBFT singularities? (c) Is the potential for vegetation recovery from the soil seed bank associated with the characteristics of their SBFT? To address these questions we examined the sizes of immediately and non-immediately-germinable seed banks (IGSB and NIGSB, respectively) in four different vegetation states of the steppe of F. pallescens resulting from the combined effects of topography and grazing. The results show that four lifeforms (perennial grasses, perennial graminoid non-grasses, annual dicots and annual grasses) display three characteristic SBFT. These are: (1) a transient type constituted by perennial grasses (SBFT1), (2) an intermediate type represented by annuals (SBFT2), and (3) a persistent type characteristic of one perennial graminoid non-grass (SBFT3). Other lifeforms (perennial dicots and biennials) could not be assigned to any of these types or characterized into a definite one. Seed bank functional types do not change among vegetation states in terms of the relative proportions of IGSB and NIGSB, although differences in the absolute and the relative size of each SBFT could be found. Species with persistent or mixed persistent/non-persistent seed bank types (SBFT2, SBFT3, etc.) colonize habitats disturbed by grazing with more success than those with non-persistent seed banks (SBFT1). Species with persistent seed banks are over-represented in the soil seed bank in relation to their contribution to above-ground cover. Management strategies for conservation in these grasslands could conveniently be targeted to increase the seed rain of perennial grasses as well as to the creation of suitable micro-environmental conditions to enhance their establishment.  相似文献

Vanadium compounds are shown to have a mitogenic effect on fibroblast cells. The effects of vanadate, vanadyl and pervanadate on the proliferation and morphological changes of Swiss 3T3 cells in culture are compared. Vanadium derivatives induced cell proliferation in a biphasic manner, with a toxic-like effect at doses over 50mM, after 24h of incubation. Vanadyl and vanadate were equally potent at 2.5–10mM. At 50mM vanadate inhibited cell proliferation, whereas slight inhibition was observed at 100mM of vanadyl. At 10mM pervanadate was as potent as vanadate and vanadyl in stimulating fibroblast proliferation, but no effect was observed at lower concentrations. A pronounced cytotoxic-like effect was induced by pervanadate at 50mM. All of these effects were accompanied by morphological changes: transformation of fibroblast shape from polygonal to fusiform; retraction with cytoplasm condensation; and loss of lamellar processes. The magnitude of these transformations correlates with the potency of vanadium derivatives to induce a cytotoxic-like effect: pervanadate>vanadate>vanadyl. These data suggest that the oxidation state and coordination geometry of vanadium determine the degree of the cytotoxicity.  相似文献
