Elucidation of mechanisms underlying plant tolerance to cadmium, a widespread toxic soil pollutant, and accumulation of Cd in plants are urgent tasks. For this purposes, the pea (Pisum sativum L.) mutant SGECdt (obtained by treatment of the laboratory pea line SGE with ethylmethane sulfonate) was reciprocally grafted with the parental line SGE, and four scion/rootstock combinations were obtained: SGE/SGE, SGECdt/SGECdt, SGE/SGECdt, and SGECdt/SGE. They were grown in hydroponics in the presence of 1 μM CdCl2 for 30 d. The SGE and SGECdt scions on the SGECdt rootstock had a higher root and shoot biomass and an elevated root and shoot Cd content compared with the grafts having SGE rootstock. Only the grafts with the SGE rootstock showed chlorosis and roots demonstrating symptoms of Cd toxicity. The content of nutrient elements in roots (Fe, K, Mg, Mn, Na, P, and Zn) was higher in the grafts having the SGECdt rootstock, and three elements, namely Ca, Fe, and Mn, were efficiently transported by the SGECdt root to the shoot of these grafts. The content of other measured elements (K, Mg, Na, P, and Zn) was similar in the root and shoot in all the grafts. Then, the non-grafted plants were grown in the presence of Cd and subjected to deficit or excess concentrations of Ca, Fe, or Mn. Exclusion of these elements from the nutrient solution retained or increased differences between SGE and SGECdt in growth response to Cd toxicity, whereas excess of Ca, Fe, or Mn decreased or eliminated such differences. The obtained results assign a principal role of roots to realizing the increased Cd-tolerance and Cdaccumulation in the SGECdt mutant. Efficient translocation of Ca, Fe, and Mn from roots to shoots appeared to counteract Cd toxicity, although Cd was actively taken up by roots and accumulated in shoots. 相似文献

Fragmentation and habitat loss pose major threats to global biodiversity. Especially forest dwelling species with small ranges and high habitat specialisation are affected by ongoing land use change. Building projects for infrastructural purposes, expanding settlements, and extensive agricultural areas are assumed to have a high impact on these species. The European Habitat’s Directive aims to conserve and restore habitat networks to lower these impacts. We propose that the idea of securing habitat networks for protected species should be incorporated within large scale landscape planning e.g. by modelling and improving corridors for umbrella species. Within a Danish-German project we developed a model demonstrating potential connecting corridors for Muscardinus avellanarius, a specialised forest dwelling rodent species with low dispersal ability. We used presence data and eco-geographical variables to find a data based time-efficient procedure which may be applied also in other species for future landscape planning. The habitat suitability model shows that the hazel dormouse occupies a narrow niche with highly suitable habitats comprising edge habitats. It indicates the preference of forest patches, linear structures and networks of the mentioned habitats. Along with connectivity the diversity of suitable habitats diversity is the major factor predicting hazel dormouse presence. For conservation management, we calculated habitat corridors and highlight sections with missing connections. This allowed us to accentuate regions of high management interest. The results represent the foundation not only for this cross-border conservation project, but also for long-term dormouse conservation on a federal-state level. 相似文献

Upon high light excitation in photosynthetic bacteria, various triplet states of pigments can accumulate leading to harmful effects. Here, the generation and lifetime of flash-induced carotenoid triplets (3Car) have been studied by observation of the quenching of bacteriochlorophyll (BChl) fluorescence in different strains of photosynthetic bacteria including Rvx. gelatinosus (anaerobic and semianaerobic), Rsp. rubrum, Thio. roseopersicina, Rba. sphaeroides 2.4.1 and carotenoid- and cytochrome-deficient mutants Rba. sphaeroides Ga, R-26, and cycA, respectively. The following results were obtained: (1) 3Car quenching is observed during and not exclusively after the photochemical rise of the fluorescence yield of BChl indicating that the charge separation in the reaction center (RC) and the carotenoid triplet formation are not consecutive but parallel processes. (2) The photoprotective function of 3Car is not limited to the RC only and can be described by a model in which the carotenoids are distributed in the lake of the BChl pigments. (3) The observed lifetime of 3Car in intact cells is the weighted average of the lifetimes of the carotenoids with various numbers of conjugated double bonds in the bacterial strain. (4) The lifetime of 3Car measured in the light is significantly shorter (1–2 μs) than that measured in the dark (2–10 μs). The difference reveals the importance of the dynamics of 3Car before relaxation. The results will be discussed not only in terms of energy levels of the 3Car but also in terms of the kinetics of transitions among different sublevels in the excited triplet state of the carotenoid. 相似文献

Sortilin was first identified based on its activity as part of intracellular protein sorting machinery. Recently, it was discovered that sortilin also acts as a cell surface receptor for the propeptide form of nerve growth factor (proNGF), progranulin, and neurotensin. The interaction of sortilin to these neurotrophic ligands is linked to diseases of the nervous system that lead to neurodegeneration and neuropathic pain. Blocking of the interaction of sortilin to these ligands may prevent or slow the progress of these nervous system disorders. In vitro screening assays for blocking compounds or peptides are part of the standard set of tools for drug discovery. However, assays for sortilin biology are not readily available to determine if the selected blocking agent inhibits sortilin activity on the surface of cells. We have developed a sortilin specific cell based assay to identify compounds that specifically block interaction between sortilin and proNGF prodomain. The assay system records both the presence of sortilin on the cell surface and the interaction with the pro domain of NGF. Fluorescent images of the sortilin expressing cells are analyzed for the presence of pro domain of NGF. Sortilin-positive and sortilin-negative cells within one well are concomitantly and automatically analyzed. Sortilin—pro domain interaction can be blocked dose dependently by neurotensin and synthetic compounds. The assay will facilitate the discovery of entities interfering with the binding of sortilin to the NGF pro domain. This assay can be modified to screen for inhibitors of the binding of ligands to other complex cell surface receptors. 相似文献

It is shown that the group of -conjugated, nonclassical (non-Kekulé) homonuclear, alternative organic polyradicals and polymers with degenerate NBMOs can be essentially extended to a large class of heterocyclic analogues having a set of degenerate MOs. The presence of a set of degenerate MOs (DMOs) results from the molecular topology of the system. The conditions of occurrence of DMOs are determined by the generalized Coulson-Rushbrooke-Longuet-Higgins theorem. The character of spin-exchange interaction of -electrons in the half-filled band (HFB) of a large group of model polymers, analogues of poly(meta-anilines), has been investigated. It is shown that the main component of the ferromagnetic exchange interaction is the potential (Coulomb) exchange and a smaller contribution of the indirect exchange among the HFB electrons via the delocalized -electrons in the occupied bands. The theoretical method used, which predicts the existence of a set of DMOs, may serve as a guiding principle in the design of narrow-band, high-spin organic polymers in which cooperative magnetic phenomena can arise.Part IX:Theoret. Abstract. Phenological development of the colonizer Bunias orientalis L. was investigated in permanent plots in herbaceous vegetation, using photography and image analysis. In all habitats but one, B. orientalis showed a two-phased rosette growth behaviour, with a first peak cover reached in spring, roughly coinciding with an early peak cover of the matrix vegetation, and a second autumn growth flush occurring in a phase of reduced matrix vegetation cover. This seasonally bimodal growth pattern of B. orientalis appears to partly compensate for its competitive inferiority in crowded herbaceous vegetation. Small or overshadowed individuals of this species particularly profit from higher above-ground performance or release from overlap in autumn. The significance of temporal niche separation for survival and growth of B. orientalis individuals varies with habitat conditions, being most apparent in occasionally disturbed habitats with a relatively low frequency of gap formation. Despite some limitations, image analysis proved to be useful for phenological investigations within herbaceous plant stands. 相似文献

Wealso determined whether either vasodilating 2-adrenergic receptors ornitric oxide (NO) contributes to this dilation. Forearm blood flow wasmeasured bilaterally in healthy volunteers(n = 10) by using plethysmographyduring two bouts of graded lower body negative pressure (LBNP) tosyncope. In one forearm, drugs were infused via a brachial arterycatheter while the other forearm served as a control. In the controlarm, forearm vascular resistance (FVR) increased from 77 ± 7 unitsat baseline to 191 ± 36 units with 40 mmHg of LBNP(P < 0.05). Mean arterial pressurefell from 94 ± 2 to 47 ± 4 mmHg just before syncope, and allsubjects demonstrated sudden bradycardia at the time of syncope. At theonset of syncope, there was sudden vasodilation and FVR fell to 26 ± 6 units (P < 0.05 vs. baseline). When the experimental forearm was treated withbretylium, phentolamine, and propranolol, baseline FVR fell to 26 ± 2 units, the vasoconstriction during LBNP was absent, and FVR fellfurther to 16 ± 1 units at syncope(P < 0.05 vs. baseline). During thesecond trial of LBNP, mean arterial pressure again fell to 47 ± 4 mmHg and bradycardia was again observed. Treatment of the experimentalforearm with the NO synthase inhibitorNG-monomethyl-L-arginine in additionto bretylium, phentolamine, and propranolol significantly increasedbaseline FVR to 65 ± 5 units but did not prevent the marked forearmvasodilation during syncope (FVR = 24 ± 4 vs. 29 ± 8 units inthe control forearm). These data suggest that the profound vasodilationobserved in the human forearm during syncope is not mediated solely bysympathetic withdrawal and also suggest that neither2-adrenergic-receptor-mediated vasodilation nor NO is essential to observe this response. 相似文献

The faunas living in the vast deep sea regions around the Antarctic are very poorly known. This is especially true for the biodiversity of polychaetes inhabiting these remote areas. Therefore, we report new morphological data of Glyceriformia from the ANDEEP cruises to the South Atlantic Ocean and the Southern Ocean. Based on benthos samples from three expeditions aboard R/V POLARSTERN, two species of Glyceridae (Glycera capitata, G. diva) and four species of Goniadidae (Bathyglycinde sibogana, B. stepaniantsae, Goniada maculata, Progoniada regularis) were studied. Furthermore, new morphological details (especially for the previously unknown tail) for the rarely found taxon B. stepaniantsae are given. The distribution patterns of the different taxa demonstrated that some species have a high dispersal capability and show an extended level of eurybathy, whereas other species are restricted to the deep sea. 相似文献
