Antigen presentation by Hodgkin's disease cells   总被引:3,自引:0,他引:3  
The L428 tumor cell line is a long-term tissue culture of Reed-Sternberg cells which was derived from the pleural effusion of a patient with Hodgkin's disease. The L428 cells express all known cell surface antigens, cytochemical staining, and cytologic features of freshly explanted Reed-Sternberg cells. In addition to the previously described HLA-DR cell surface antigens, the L428 cells are now demonstrated to express both DS and SB alloantigens. Thus, the L428 cells express all of the known subclasses of the human immune response genes that are located in the major histocompatibility complex. Furthermore, the L428 cells are capable of presenting soluble antigen to T cells in a genetically restricted fashion. T cell lines were established from normal donors previously immunized with tetanus toxoid. The T cells utilized were incapable of tetanus toxoid-induced proliferation unless antigen-presenting cells were added to the cultures. However, T cells from the two normal donors, which like the L428 cells expressed HLA-DR 5, demonstrated significant proliferative responses when cultured with tetanus toxoid and L428 cells. No proliferative response was observed when the L428 cells were used as antigen-presenting cells for a DR (4,-), DR (2,-) or DR (1,7) T cell line. The tetanus toxoid dose-response curve was similar regardless of whether autologous mononuclear leukocytes or L428 cells were used as antigen-presenting cells. The T cell proliferation induced by soluble antigen was also blocked by anti-HLA-DR antibody. Thus, functionally, Hodgkin's disease may be classified as a tumor of antigen-presenting cells.  相似文献

Twenty crossbred gilts with at least 2 consecutive estrous cycles of 18 to 21 days in length were used to study the effects of prostaglandins E2 and F2α (PGE2 and PGF2α) on luteal function in indomethacin (INDO) treated cycling gilts. Intrauterine and jugular vein catheters were surgically palced before day 7 of the treatment estrous cycle and gilts were randomly assigned to 1 of 5 treatment groups (4/groups). With exception of the controls (Group I) all gilts received 3.3 mg/kg INDO every 8 h, Groups III, IV and V received 2.5 mg PGF2; 2.5 mg PGF2α + 400 μg PGE2 every 4 hr, or 400μg PGE2 every 4 h, respectively. All treatments were initiated on day 7 and continued until estrus or day 23. Jugular blood for progesterone analysis was collected twice daily from day 7 to 30. Estradiol-17β (E2-17β) concentrations were dtermined in samples collected twice daily, from 2 d before until 2 d following the day of estrus onset. When compared to pretreatment values, estrous cycle length was unaffected (P>0.05) in Group I, prolonged (P<0.05) in Groups II, IV and V; and shortened (P<0.05) in Group III. The decline in plasma progesterone concentration that normally occurs around day 15 was unaffected (P>.05) in Group I; delayed (P<0.05) in Groups II, IV and V; and occurred early (P<0.05) in Group III. Mean E2-17β remained high (31.2 ± 4.9 to 49.3 ± 3.1 pg/ml) in Groups III and IV, while the mean concentrations in Groups III and V varied considerably (17.0 ± 2.0 to 52.2 ± 3.5 pg/ml). The results of this study have shown that PGE2 will counteract the effects of PGF2α in INDO treated cycling gilts. The inclusion of PGF2α appeared to either stimulate E2-17β secretion or maintain it at a higher level than other treatments.  相似文献

Zusammenfassung Die Herzfrequenz (HF) von 4 Amsel- wurde mit Hilfe radiotelemetrischer Techniken aufgezeichnet, während die Vögel Subsong oder Vollgesang äußerten. Die 3 Versuchsbedingungen waren: (1) Käfighaltung mit akustischem Kontakt zu anderen Amsel- und ; (2) Isolierte Käfighaltung in einem reflexionsarmen Raum, in dem Tonbandvorspielexperimente mit den Amseln durchgeführt wurden; (3) Haltung in einer Freivoliere. Die Äußerung von Subsong konnte von 3 verschiedenen HF-Verläufen begleitet sein: a) einer mit langfristigen Schwankungen einhergehenden deutlichen Akzeleration; b) einem gleichmäßig ruhigen und niedrigen HF-Verlauf; c) einer Dezeleration, die mit Beginn des Gesangsabschnittes abrupt einsetzte und mit Beendigung des Singens ebenso abrupt wieder endete. Vollgesang-Strophen konnten von 4 verschiedenen HF-Verläufen begleitet sein: a) einer reinen Akzeleration; b) einer abrupten Dezeleration; c) einer gleichmäßig ruhigen HF auf dem Niveau der Ruhewerte; d) einem biphasischen Verlauf. In den unterschiedlichen HF-Kurventypen drücken sich möglicherweise unterschiedliche Grade der physischen und/oder psychischen Belastung der Amseln während des Singens aus. Niedrigste Belastung, d. h. eine entspannte Situation, wird angenommen, wenn die HF niedrig und gleichmäßig ist. Erhöhte Anspannung scheint zu bestehen, wenn Dezelerationen auftreten. Bei noch höherer Belastung wird der Gesang von HF-Erhöhungen begleitet. Der Grad der physischen und/oder psychischen Belastung des singenden Vogels hängt wohl mit dem sozialen Kontext und den Umgebungsbedingungen zusammen. Abhängig von dieser Belastung ist möglicherweise die Funktion der Lautäußerungen auch jedesmal eine andere.
Radiotelemetric measurements of heart rate in singing Blackbirds (Turdus merula)
Summary The heart rate (HR) of 4 male Blackbirds was monitored by means of radiotelemetry in subsong and full song phases of singing birds. Results were obtained under 3 experimental conditions: (1) caged birds in the presence of conspecific and ; (2) isolated caged birds in an anachoic chamber where playback experiments were conducted; (3) birds in an outdoor aviary. The singing of subsong could be related to 3 different types of HR curves: a) a marked acceleration combined with oscillations in longer cycles; b) a low and steady heart rate; c) a deceleration beginning abruptly with the onset of singing and ending abruptly when singing terminated. Strophes of full song were related to 4 kinds of HR curves: a) an acceleration; b) an abrupt deceleration; c) a steady HR on the level of the resting HR values; d) a biphasic course. The rather heterogeneous pattern of HR curves during singing suggests a hypothetic hierarchy related to motivation and function of certain song types. The low and steady curves express the lowest level, the decelerations a medium level and the accelerations the highest level of physiological strain or psychical tension. Strain and tension probably depend on the social context and the conditions in the birds' environment. The functions of the sounds may also differ and find expression in different modes of HR alterations.

Tumor cell lines derived from malignant schwannomas removed from patients with neurofibromatosis type 1 (NF1) have been examined for the level of expression of NF1 protein. All three NF1 lines examined expressed lower levels of NF1 protein than control cells, and the level in one line was barely detectable. The tumor lines expressed normal levels of p120GAP and p21ras. Although the p21ras proteins isolated from the tumor cells had normal (nonmutant) biochemical properties in vitro, they displayed elevated levels of bound GTP in vivo. The level of total cellular GAP-like activity was reduced in extracts from the tumor line that expresses very little NF1 protein. Introduction of the catalytic region of GAP into this line resulted in morphological reversion and lower in vivo GTP binding by endogenous p21ras. These data implicate NF1 protein as a tumor suppressor gene product that negatively regulates p21ras and define a "positive" growth role for ras activity in NF1 malignancies.  相似文献

Flows of energy and matter across habitat boundaries can be major determinants of the functioning of recipient ecosystems. It is currently debated whether terrestrial dissolved organic matter (tDOM) is a resource subsidy or a resource subtraction in recipient lakes. We present data from a long-term field experiment in which pelagic phosphorus concentration and whole-ecosystem primary production increased with increasing tDOM input, suggesting that tDOM acted primarily as a direct nutrient subsidy. Piecewise structural equation modeling supports, however, a substantial contribution of a second mechanism: colored tDOM acted also as a resource subtraction by shading benthic algae, preventing them from intercepting nutrients released across the sediment–water interface. Inhibition of benthic algae by colored tDOM thus indirectly promoted pelagic algae and whole-ecosystem primary production. We conclude that cross-ecosystem terrestrial DOM inputs can modify light and nutrient flows between aquatic habitats and alter the relative contributions of benthic and pelagic habitats to total primary production. These results are particularly relevant for shallow northern lakes, which are projected to receive increased tDOM runoff.  相似文献

This paper reports on ant and termite species inhabiting the mounds (murundus) found in three wetland sites in Santo Antonio da Patrulha. Ants and termites were found in 100% of the mounds of two sites and in 20% of those in the third site. Colonies of Camponotus fastigatus were found inhabiting all the mounds, while colonies of Brachymyrmex sp., Linepithema sp., Pheidole sp., and/or Solenopsis sp. were collected in less than 30% of the mounds. In the mounds of the three sites, colonies of Anoplotermes sp. and/or Aparatermes sp. termites were found together with the ant colonies. Another cohabiting termite species, Cortaritermes sp., was found only in the mounds of one site. The results suggest that C. fastigatus is the species building the mounds, with the other species, whether ants or termites, being the inquilines.  相似文献

Retinopathy of prematurity (ROP) is a vasoproliferative disorder that occurs in premature infants and may lead to permanent visual impairment. We investigated both the possible protective role of N-acetyl cysteine (NAC) for preventing ROP and the role of IGF-1 in the disorder. Forty-five newborn rats were divided into three groups. Group 1 was raised in room air as controls. Group 2 was exposed to 60% oxygen for 14 days after birth, then transferred to room air. Group 3 was exposed to the same conditions as group 2, but received intraperitoneal injections of NAC on postnatal days 7–17. After 35 days, both eyes of all rats were processed for histology. Some sections were stained with hematoxylin and eosin to assess structural changes and other sections were immunostained to determine the location of IGF-1. Frozen sections also were prepared and stained for adenosine triphosphatase to detect retinal blood vessels. Compared to the controls, more blood vessels, many of which were abnormal, and increased IGF-1 expression were observed in group 2. In group 3, abnormal blood vessels and IGF-1 expression were less evident. NAC appeared to be an effective vascular-protective agent for ROP by decreasing IGF-1 expression.  相似文献

In this paper we show that a 211-base pair segment of CEN3 DNA is sufficient to confer wild-type centromere function in the yeast Saccharomyces cerevisiae. We used site-directed mutagenesis of the 211-base pair fragment to examine the sequence-specific functional requirements of a conserved 11-base pair segment of centromere DNA, element III (5'-TGATTTATCCGAA-3'). Element III is the most highly conserved of the centromeric DNA sequences, differing by only a single adenine X thymine base pair among the four centromere DNAs sequenced thus far. All of the element III sequences contain specific cytosine X guanine base pairs, including a 5'-CCG-3' arrangement, which we targeted for single cytosine-to-thymine mutations by using sodium bisulfite. The effects of element III mutations on plasmid and chromosome segregation were determined by mitotic stability assays. Conversion of CCG to CTG completely abolished centromere function both in plasmids and in chromosome III, whereas conversion of CCG to TCG decreased plasmid and chromosome stability moderately. The other two guanine X cytosine base pairs in element III could be independently converted to adenine X thymine base pairs without affecting plasmid or chromosome stability. We concluded that while some specific nucleotides within the conserved element III sequence are essential for proper centromere function, other conserved nucleotides can be changed.  相似文献
