Thyroxine transport in choroid plexus   总被引:4,自引:0,他引:4  
The role of the choroid plexus in thyroid hormone transport between body and brain, suggested by strong synthesis and secretion of transthyretin in this tissue, was investigated in in vitro and in vivo systems. Rat choroid plexus pieces incubated in vitro were found to accumulate thyroid hormones from surrounding medium in a non-saturable process. At equilibrium, the ratio of thyroid hormone concentration in choroid plexus pieces to that in medium decreased upon increasing the concentration of transthyretin in the medium. Fluorescence quenching of fluorophores located at different depths in liposome membranes showed maximal hormone accumulation in the middle of the phospholipid bilayer. Partition coefficients of thyroxine and triiodothyronine between lipid and aqueous phase were about 20,000. After intravenous injection of 125I-labeled thyroid hormones, choroid plexus and parts of the brain steadily accumulated 125I-thyroxine, but not [125I]triiodothyronine, for many hours. The accumulation of 125I-thyroxine in choroid plexus preceded that in brain. The amount of 125I-thyroxine in non-brain tissues and the [125I]triiodothyronine content of all tissues decreased steadily beginning immediately after injection. A model is proposed for thyroxine transport from the bloodstream into cerebrospinal fluid based on partitioning of thyroxine between choroid plexus and surrounding fluids and binding of thyroxine to transthyretin newly synthesized and secreted by choroid plexus.  相似文献

Enhanced resistance in perennial ryegrass, Lolium perenne L., to the bluegrass billbug, Sphenophorus parvulus Gyllenhal is associated with the presence of a fungal endophyte, Acremonium loliae Latch et al. Severe billbug infestations occurred in 1981, 1983, and 1984 in two different turf evaluation field trials. Striking differences in cultivar response were observed. Subsequent investigations led to the discovery that billbugresistant ryegrass entries had very high levels of endophyte infection; whereas, susceptible entries had little or no endophyte infection. Further, the resistance was found to be maternally transmitted, and its dramatic prevalence within a maternal line was explained by the high frequency (over 95%) of seed transmission of the endophyte. This newly recognized source of plant resistance to several insect pests warrants additional study.
Résumé Des études récentes ont montré que la présence d'A. loliae à l'intérieur de L. perenne augmentait sa résistance à plusieurs insectes, dont Listronotus (Hyperodes) bonariensis et quelques pyralesnous signalons ici que le même endophyte améliore aussi la résistance à Sphenophorus parvulus (Curculionidae).Des dégâts de S. parvulus ont été observés sur gazon en 1981, 1983 et 1984. Des différences marquées quant à la réaction des cultivars ont été notées; des études ultérieures ont montré que la résistance des cultivars était associée à des niveaux très élevés d'endophytes, ceux-ci étant rares ou absents des cultivars sensibles. La transmission maternelle de la résistance à S. parvulus améliorée par la présence de l'endophyte qui avait été mise en évidence peut s'expliquer par le pourcentage élevé de graines transmettant le champignon. Les cultivars Pennant, Premier, Regal aux niveaux élevés d'endophytes présentent peu de dégâts en gazon.Les avantages dûs à la présence de l'endophyte, exprimés par une meilleure résistance aux insectes, étaient particulièrement spectaculaires lorsque l'entretien de gazon était sommaire. L'amélioration due à l'endophyte n'apparaissait pas avec les gazons bien entretenus où il n'y avait pas de dégâts d'insectes. Ceci ne réduit en rien l'intérêt de cette forme de résistance aux insectes.Des questions importantes restent à résoudre: action de l'endophyte sur l'hôte, identification et influence des toxines éventuellement impliquées, examen d'autres plantes et insectes concernés.

The role of polyamines in the control of phenylalanine hydroxylase phosphorylation state and enzymic activity was investigated. Pre-treatment of liver cells with spermine (1 mM) abolishes the glucagon (1 nM)-stimulated increase in hydroxylase phosphorylation. Concurrently there is a decrease in phenylalanine hydroxylation flux, reflecting decreased enzyme activity; 50% inhibition occurs at approx. 10 microM-spermine. These results are discussed in the context of reports concerning the properties of protein phosphatase 2A.  相似文献

The neural cell adhesion molecule (N-CAM) is believed to be a key regulator of adhesive events in the nervous system and skeletal muscle. The recent isolation of N-CAM cDNAs from different tissues has identified a high degree of diversity in primary amino acid sequence between different isoforms. In this article, we review these recent studies and discuss methods for unravelling the functional consequences of the generation of multiple N-CAM polypeptides using gene transfection approaches.  相似文献

A mitogen derived from supernatants of Mycoplasma arthritidis (MAS) has been shown to induce both T-cell activation and the production of interferon-gamma (IFN-gamma). MAS-induced response required the presence of accessory cells and is under the genetic control associated with the major histocompatibility complex (MHC). We found that recombinant IFN-gamma restored the proliferative response to MAS mitogen in unreactive mice strains, including H-2b and H-2s haplotypes. We postulated that these T-cells fail to respond since they lack part of the I-E molecules on their accessory cells. Our data suggest that interferon-gamma may be able not only to increase the levels of Ia antigens but also to promote the appearance of MHC products that are not usually present on the cell surface. Since Ia antigens have a central role on T-cell activation, we examined the effect of the level of IFN-gamma on MAS-induced T-cell activation. We analyzed the acquisition of responsiveness to IL-2 and IL-2 activity in cells pretreated with IFN-gamma and found that both the steps of T-cell activation were restored to the MAS mitogen in the unreactive mice strains by IFN-gamma.  相似文献

Tissue patterns of gene expression were analyzed by measuring mRNA levels and incorporation of radioactive amino acids for cystatin C and beta 2-microglobulin, the two extracellular proteins in the brain with the highest ratio of concentration in cerebrospinal fluid over that in blood plasma. The primary structure of rat cystatin C mRNA from choroid plexus was determined by nucleotide sequencing of cloned cDNA and the tissue patterns of gene expression were analysed by RNA blot analysis and in situ hybridization. Cystatin C was found to be composed of 120 amino acids and to contain a potential site for N-linked glycosylation. The tissue with the highest cystatin C mRNA level was the choroid plexus of the brain. Cystatin C mRNA was also detected in lower levels in other areas of the brain, testis, epididymis, seminal vesicles, prostate, ovary, submandibular gland, and, in trace amounts, in liver. Choroid plexus pieces in culture secreted radioactive cystatin C when incubated with radioactive leucine. Rat beta 2-microglobulin cDNA was cloned and identified by nucleotide sequencing and comparison of the obtained sequence with that of mouse and human beta 2-microglobulin cDNA. Tissue levels of beta 2-microglobulin mRNA in the rat were measured by hybridization to rat beta 2-microglobulin cDNA. The highest levels of beta 2-microglobulin mRNA were observed in liver and choroid plexus. Other parts of the brain and testis contained lower levels of beta 2-microglobulin mRNA.  相似文献
