Childhood acute lymphoblastic leukaemia (ALL), a malignant transformation of the lymphoblasts, is highly responsive to chemotherapy. However, due to certain inadequacy in detection of minimal residual disease (MRD), relapse is a common phenomenon. To address this question, the present review deals with the induction of an unique O-acetyl derivative of sialic acid on a few disease-associated glycoproteins and glycolipids at the onset of childhood ALL, a finding of our group in the last decade. This information has been successfully utilized for diagnosis and prognosis of the disease. Existing literature is included for comparison. Additionally, cell surface overexpression of 9-O-acetylated sialoglycoproteins and antibodies against them present in patients' sera aid the survival of the malignant lymphoblasts and suggest a multifaceted role played by these molecules. Taken together, monitoring these molecules helps not only in unravelling the biology of this paediatric malignancy but also in personalizing the treatment strategies for the betterment of the patient population. 相似文献

Different forms of C-reactive proteins have been purified to electrophoretic homogeneity by calcium dependent affinity chromatography on a phosphorylcholine (PC)-Sepharose column from the sera of Labeo rohita confined in fresh water (CRP(N)) and water polluted with sublethal doses of cadmium (CRP(Cd)), mercury (CRP(Hg)), phenol (CRP(Ph)), and hexachlorocyclohexane (CRP(Hx)), which elevate serum CRP levels by three- to fivefold. On native PAGE, induced forms of CRP show remarkable differences in their electrophoteric mobility indicating differences in molecular mass, charge, and/or shape. Kinetic studies reveal the appearance of a pollutant specific molecular variant, which replaces the normal form at the peak of induction. Studies on amino acid and carbohydrate compositions, isoelectric focusing, binding to PC, C-polysaccharide (CPS) & lectins, and secondary structures of the purified CRPs, indicate, that, they differ significantly from each other, but grossly share the common properties of a CRP, including pentraxin, structure revealed by electron microscopy. 相似文献

The newly emerged adult male and female Schizodactylus monstrosus D. were treated with an insecticide, pyrethrum or with petroleum ether in the case of controls. At selected intervals of 6 h, 12 h and 18 h after treatment, the brain and ventral nerve cord and ganglia were homogenized to estimate the activity of acetyl-cholinesterase, and acid and alkaline phosphatase. The resultant activities of these three enzymes showed that: acetylcholinesterase decreased rapidly, and acid and alkaline phosphatase increased significantly after pyrethrum treatment in both brain and ventral nerve cord with ganglia compared with the controls. The activities of alkaline phosphatase showed a marked fluctuation at different post-treatment periods in both the tissues. The results have been discussed in relation to the impact caused by the treated insecticide and its metabolism. 相似文献

The thermophilic, sulfur metabolizing Archaeoglobus fulgidus contains two genes, AF0473 and AF0152, encoding for PIB-type heavy metal transport ATPases. In this study, we describe the cloning, heterologous expression, purification, and functional characterization of one of these ATPases, CopA (NCB accession number AAB90763), encoded by AF0473. CopA is active at high temperatures (75 degrees C; E(a) = 103 kJ/mol) and inactive at 37 degrees C. It is activated by Ag+ (ATPase V(max) = 14.82 micromol/mg/h) and to a lesser extent by Cu+ (ATPase V(max) = 3.66 micromol/mg/h). However, Cu+ interacts with the enzyme with higher apparent affinity (ATPase stimulation, Ag+ K(12) = 29.4 microm; Cu+ K(12) = 2.1 microm). This activation by Ag+ or Cu+ is dependent on the presence of millimolar amounts of cysteine. In the presence of ATP, these metals drive the formation of an acid-stable phosphoenzyme with apparent affinities similar to those observed in the ATPase activity determinations (Ag+, K(12) = 23.0 microm; Cu+, K(12) = 3.9 microm). However, comparable levels of phosphoenzyme are reached in the presence of both cations (Ag+, 1.40 nmol/mg; Cu+, 1.08 nmol/mg). The stimulation of phosphorylation by the cations suggests that CopA drives the outward movement of the metal. CopA presents additional functional characteristics similar to other P-type ATPases. ATP interacts with the enzyme with two apparent affinities (ATPase K(m) = 0.25 mm; phosphorylation K(m) = 4.81 microm), and the presence of vanadate leads to enzyme inactivation (IC(50) = 24 microm). This is the first Ag+/Cu+ -ATPase expressed and purified in a functional form. Thus, it provides a model for structure-functional studies of these transporters. Fms-like tyrosine kinase-3 (FLT3) is a growth factor receptor normally expressed on hematopoietic progenitor cells. Approximately one third of all patients with AML carry an activating mutation in FLT3 that drives proliferation and survival of the leukemic cells. The most common activating mutation is the so-called internal tandem duplication (ITD), which involves an in-frame duplication of a segment of varying length in the region of the FLT3 gene that encodes the juxtamembrane domain. The pathways downstream of FLT3-ITD are partially known but further knowledge regarding the downstream signal transduction molecules is important in order to develop alternative strategies for pharmacological intervention.In this paper we have studied the role of MEK/ERK5 in FLT3-ITD mediated transformation. We have found that both wild-type FLT3 and FLT3-ITD activate MEK5 leading to the activation of ERK5. By use of the selective inhibitor of MEK5, BIX02188, we have shown that activation of AKT downstream of FLT3 is partially dependent on ERK5. Furthermore, inhibition of MEK5/ERK5 induces apoptosis of both FLT3-ITD transfected Ba/F3 cells as well as the FLT3-ITD carrying leukemic cell lines MV4-11 and MOLM-13. These results suggest that MEK5/ERK5 is important for FLT3-ITD induced hematopoietic transformation and may thus represent an alternative therapeutic target in the treatment of FLT3-ITD positive leukemia. 相似文献

We report here the crystal structure of the partially self-complementary decameric sequence d(CGGCGGCCGC), which self assembles to form a four-way junction with sticky ends. Each junction binds to four others through Watson–Crick base pairing at the sticky ends to form a rhombic structure. The rhombuses bind to each other and form two dimensional tiles. The tiles stack to form the crystal. The crystal diffracted in the space group P1 to a resolution of 2.5 Å. The junction has the anti-parallel stacked-X conformation like other junction structures, though the formation of the rhombic net noticeably alters the details of the junction geometry. 相似文献

Extracellular polymeric substances (EPS) produced by a toxic dinoflagellate Amphidinium carterae Hulburt 1957 was isolated and characterized. Molecular masses of the EPS were about 233 and 1,354 kDa. Spectral analyses
by 1H nuclear magnetic resonance and Fourier Transformed–Infrared Spectroscopy revealed the characteristic of the functional groups

The present study describes preparation and characterization of a thermally stable and biodegradable biopolymer using collagen and a natural polymer, alginic acid (AA). Required concentration of alginic acid and collagen was optimized and the resulting biopolymer was characterized for, degree of cross-linking, mechanical strength, thermal stability, biocompatibility (toxicity) and biodegradability. Results reveal, the degree of cross-linking of alginic acid (at 1.5% concentration) with collagen was calculated as 75%, whereas it was 83% with standard cross-linking agent, glutaraldehyde (at 1.5% concentration). The AA cross-linked biopolymer was stable up to 245°C and Exhibits 5-6-fold increase in mechanical (tensile) strength compared to plain collagen (native) materials. However, glutaraldehyde cross-linked material exhibits comparatively less thermal stability and brittle in nature (low tensile strength). With regard to cell toxicity, no cytotoxicity was observed for AA cross-linked material when tested with mesenchymal cells and found degradable when treated with collagenase enzyme. The nature of bonding pattern and the reason for thermal stability of AA cross-linked collagen biopolymer was discussed in detail with the help of bioinformatics. A supplementary file on efficacy of AACC as a wound dressing material is demonstrated in detail with animal model studies. 相似文献
