The presence of viral antigen in sections from formalin-fixed and paraffin-embedded human tissues was demonstrated by trypsin digestion followed by direct or indirect immunofluorescence. The specimens may be used for retrospective diagnosis. The immunofluorescence technique has to be adapted to the suspected virus infection on the basis of previous histopathologic study. Variations of trypsin concentration time and temperature of incubation, expose different viral antigens and have to be previously tested for each unknown system. For measles virus detection in lung a stronger digestion has to be applied as compared to adenovirus or respiratory disease viruses in the same tissue. Flavivirus in liver tissue needs a weaker digestion. The reproducibility of the method makes it useful as a routine technique in diagnosis of virus infection. 相似文献

Characterization of poliovirus strains by an enzyme immunoassay (EIA). An enzyme immunoassay performed with polyclonal antisera to poliovirus types 1, 2 and 3 absorbed with heterologous strains of the same types was used for the characterization of poliovirus field isolates. The assay allowed the identification of 74 (93.6%) out of 79 isolates as wild or vaccine-like strains. Applications of EIA for the study of polioviruses are discussed. 相似文献

One important limitation of topical photodynamic therapy (PDT) is the limited tissue penetration of precursors. Microneedles (MNs) are minimally invasive devices used to promote intradermal drug delivery. Dissolving MNs contain drug-associated to polymer blends, dissolving after insertion into skin, allowing drug release. This study comprises development and characterization of a pyramidal model of dissolving MNs (500 μm) prepared with 5% wt/wt aminolevulinic acid and 20% wt/wt Gantrez AN-139 in aqueous blend. Protoporphyrin IX formation and distribution were evaluated in tumor mice model by using fluorescence widefield imaging, spectroscopy, and confocal microscopy. MNs demonstrated excellent mechanical resistance penetrating about 250 μm with minor size alteration in vitro, and fluorescence intensity was 5-times higher at 0.5 mm on average compared to cream in vivo (being 10 ± 5 a.u. for MNs and 2.4 ± 0.8 a.u. for cream). Factors influencing the home range of a typical group (ten to seventeen individuals) of mountain gorilla (Gorilla beringei) over a 60-month period are discussed. Three other gorilla groups lived nearby and shared the group's home range. Vegetation zones were defined in terms relevant to the gorillas' needs, and the home range was divided into equal grid squares, each of one sixteenth of a square kilometre. The observer's contacts with the group within each square provide data on the relative utilization of different areas. The physical aspects of the six vegetation zones are discussed, and five types of travel within the various zones are defined. The current expansion of the home range is described. The presence or absence of other gorilla groups and/or individuals appeared to be the strongest determinant of the range patterns of the study group, though food distribution may have played some role in movements between vegetation zones or parts of the range. 相似文献

A dengue outbreak started in March, 1986 in Rio de Janeiro and spread very rapidly to other parts of the country. The great majority of cases presented classical dengue fever but there was one fatal case, confirmed by virus isolation. Dengue type 1 strains were isolated from patients and vectors (Aedes aegypti) in the area by cultivation in A. albopictus C6/36 cell line. The cytopathic effect (CPE) was studied by electron microscopy. An IgM capture test (MAC-ELISA) was applied with clear and reproducible results for diagnosis and evaluation of virus circulation; IgM antibodies appeared soon after start of clinical disease, and persisted for about 90 days in most patients. The test was type-specific in about 50% of the patients but high levels of heterologous response for type 3 were observed. An overall isolation rate of 46.8% (813 virus strains out of 1734 specimens) was recorded. The IgM test increased the number of confirmed cases to 58.2% (1479 out of 2451 suspected cases). The importance of laboratory diagnosis in all regions where the vectors are present is emphasized. 相似文献

The aim of this study was the application of a phenotype-driven N-ethyl-N-nitrosourea (ENU) mutagenesis screen in mice for the identification of dominant mutations involved in the regulation and
modulation of alcohol-drinking behavior. The chemical mutagen ENU was utilized in the generation of 131 male ENU-mutant C57BL/6J
mice (G0). These ENU-treated mice were paired with wild-type C57BL/6J mice to generate G1 and subsequent generations. In total,
3327 mice were generated. Starting with G1, mice were screened for voluntary oral self-administration of 10% (v/v) alcohol
vs. water in a two-bottle paradigm. From these mice, after a total period of 5 weeks of drinking, 43 mutants fulfilled the
criteria of an “alcohol phenotype,” that is, high or low ethanol intake. They were then selected for breeding and tested in
a “confirmation cross” (G2–G4) for inheritance. Although we did not establish stable high or low drinking lines, several results
were obtained in the context of alcohol consumption. First, female mice drank more alcohol than their male counterparts. Second,
the former demonstrated greater infertility. Third, all animals displayed relatively stable alcohol intake, although significantly
different in two different laboratories. Finally, seasonal and monthly variability was observed, with the highest alcohol
consumption occurring in spring and the lowest in autumn. In conclusion, it seems difficult to identify dominant mutations
involved in the modulation or regulation of voluntary alcohol consumption via a phenotype-driven ENU mutagenesis screen. In
accordance with the findings from knockout studies, we suggest that mainly recessive mutations contribute to an alcohol-drinking
or alcohol-avoiding phenotype. 相似文献
