The fibrillins are a large family of chloroplast proteins that have been linked with stress tolerance and disease resistance. FIBRILLIN4 (FIB4) is found associated with the photosystem II light-harvesting complex, thylakoids, and plastoglobules, which are chloroplast compartments rich in lipophilic antioxidants. For this study, FIB4 expression was knocked down in apple (Malus 3 domestica) using RNA interference. Plastoglobule osmiophilicity was decreased in fib4 knockdown (fib4 KD) tree chloroplasts compared with the wild type, while total plastoglobule number was unchanged. Compared with the wild type, net photosynthetic CO2 fixation in fib4 KD trees was decreased at high light intensity but was increased at low light intensity. Furthermore, fib4 KD trees produced more anthocyanins than the wild type when transferred from low to high light intensity, indicating greater sensitivity to high light stress. Relative to the wild type, fib4 KD apples were more sensitive to methyl viologen and had higher superoxide levels during methyl viologen treatment. Arabidopsis (Arabidopsis thaliana) fib4 mutants and fib4 KD apples were more susceptible than their wild-type counterparts to the bacterial pathogens Pseudomonas syringae pathovar tomato and Erwinia amylovora, respectively, and were more sensitive to ozone-induced tissue damage. Following ozone stress, plastoglobule osmiophilicity decreased in wild-type apple and remained low in fib4 KD trees; total plastoglobule number increased in fib4 KD apples but not in the wild type. These results indicate that FIB4 is required for plastoglobule development and resistance to multiple stresses. This study suggests that FIB4 is involved in regulating plastoglobule content and that defective regulation of plastoglobule content leads to broad stress sensitivity and altered photosynthetic activity.Increased production of reactive oxygen species (ROS) is among the first biochemical responses of plants when challenged by pathogens and harsh environmental conditions (Mehdy, 1994; Lamb and Dixon, 1997; Joo et al., 2005). ROS are implicated in tissue damage during environmental stress and in the promotion of disease development by necrotrophic and hemibiotrophic pathogens (Venisse et al., 2001; Apel and Hirt, 2004; Shetty et al., 2008). For example, ROS production is critical for host colonization and pathogenesis by the bacterium Erwinia amylovora, which causes fire blight disease in rosaceous plants such as apple (Malus 3 domestica) and pear (Pyrus communis; Venisse et al., 2001).The chloroplast is a site of ROS production during biotic and abiotic stress (Joo et al., 2005; Liu et al., 2007). The chloroplast has a battery of enzymes such as superoxide dismutase and ascorbate peroxidase, and antioxidants such as ascorbate, glutathione, and tocopherols, for protection against ROS (Noctor and Foyer, 1998; Asada, 2006). Plastoglobules are lipoprotein bodies attached to the thylakoids (Austin et al., 2006) that store lipids, including antioxidants such as tocopherols, carotenes, and plastoquinones (Steinmüller and Tevini, 1985; Tevini and Steinmüller, 1985). In addition to antioxidants, plastoglobules contain tocopherol cyclase, which is involved in γ-tocopherol synthesis (Austin et al., 2006; Vidi et al., 2006). The antioxidant content of plastoglobules and their apparent involvement in tocopherol biosynthesis imply that they could play a role in plant responses to oxidative stress.Plastoglobules contain fibrillins, which were initially described as protein components of chromoplast fibrils with a molecular mass of approximately 30 kD (Winkenbach et al., 1976; Knoth et al., 1986; Emter et al., 1990; Deruère et al., 1994). Fibrillins are ubiquitous proteins present from cyanobacteria to plants (Laizet et al., 2004). Fibrillins maintain plastoglobule structural integrity (Deruère et al., 1994; Pozueta-Romero et al., 1997; Langenkämper et al., 2001; Vidi et al., 2006; Bréhélin et al., 2007) and stabilize the photosynthetic apparatus during photooxidative stress (Gillet et al., 1998; Yang et al., 2006; Youssef et al., 2010), osmotic stress (Gillet et al., 1998), drought (Pruvot et al., 1996; Rey et al., 2000), and low temperature (Rorat et al., 2001). Fibrillins are involved in abscisic acid-mediated protection from photoinhibition (Yang et al., 2006), and a subfamily of Arabidopsis (Arabidopsis thaliana) fibrillins (FIB1a, -1b, and -2) conditions jasmonate production during low-temperature, photooxidative stress (Youssef et al., 2010). Arabidopsis plants lacking one fibrillin (At4g22240) and tomato (Solanum lycopersicum) plants with suppressed expression of a fibrillin (LeCHRC) are susceptible to Pseudomonas syringae and Botrytis cinerea, respectively (Cooper et al., 2003; Leitner-Dagan et al., 2006), indicating that fibrillins play a role in disease resistance.The Arabidopsis fibrillin encoded by At3g23400 has received various appellations, including FIBRILLIN4 (FIB4; Laizet et al., 2004), Harpin-Binding Protein1 (Song et al., 2002), AtPGL 30.4 (Vidi et al., 2006), and Fibrillin6 (Galetskiy et al., 2008); here, it will be referred to by its earliest published name, FIB4. FIB4 is found associated with the PSII light-harvesting complex (Galetskiy et al., 2008). FIB4 has also been detected in plastoglobules (Vidi et al., 2006; Ytterberg et al., 2006) and thylakoids (Friso et al., 2004; Peltier et al., 2004). However, the specific function of FIB4 is unknown. Several lines of evidence suggest that FIB4 may be involved in plant disease resistance responses: pathogen-associated molecular patterns trigger its phosphorylation (Jones et al., 2006); pathogen-associated molecular patterns stimulate the expression of its ortholog in tobacco (Nicotiana tabacum; Jones et al., 2006; Sanabria and Dubery, 2006); and it can physically interact with the HrpN (harpin) virulence protein of the fire blight pathogen E. amylovora in a yeast two-hybrid assay, suggesting that it could be a receptor or target of HrpN (Song et al., 2002). In addition, it is thought that FIB4 may be involved in the transport of small, hydrophobic molecules because it contains a conserved lipocalin signature (Jones et al., 2006). Here, we report a genetic analysis of FIB4 function in apple and Arabidopsis in terms of its role in plastoglobule development and plant resistance to biotic and abiotic stresses.  相似文献

MK5, a member of the MAPK-activated protein kinase family, is highly expressed in the heart. Whereas MK2 and MK3 are activated by p38 MAPK, MK5 has also been shown to be activated by ERK3 and ERK4. We studied the regulation of MK5 in mouse heart. mRNA for 5 splice variants (MK5.1–5.5), including the original form (MK5.1), was detected. MK5 comprises 14 exons: exon 12 splicing was modified in MK5.2, MK5.3, and MK5.5. MK5.2 and MK5.5 lacked 6 bases at the 3′-end of exon 12, whereas MK5.3 lacked exon 12, resulting in a frame shift and premature termination of translation at codon 3 of exon 13. MK5.4 and MK5.5 lacked exons 2–6, encoding kinase subdomains I–VI, and were kinase-dead. All 5 MK5 variants were detected at the mRNA level in all mouse tissues examined; however, their relative abundance was tissue-specific. Furthermore, the relative abundance of variant mRNA was altered both during hypertrophy and postnatal cardiac development, suggesting that the generation or the stability of MK5 variant mRNAs is subject to regulation. When expressed in HEK293 cells, MK5.1, MK5.2 and MK5.3 were nuclear whereas MK5.4 and MK5.5 were cytoplasmic. A p38 MAPK activator, anisomycin, induced the redistribution of each variant. In contrast, MK5 co-immunoprecipitated ERK3, but not ERK4 or p38α, in control and hypertrophying hearts. GST pull-down assays revealed unbound ERK4 and p38α but no free MK5 or ERK3 in heart lysates. Hence, 1) in heart MK5 complexes with ERK3 and 2) MK5 splice variants may mediate distinct effects thus increasing the functional diversity of ERK3–MK5 signaling.  相似文献

We previously demonstrated that angiotensin II (Ang II) receptor signaling is involved in azoxymethane-induced mouse colon tumorigenesis. In order to clarify the role of Ang II in COX-2 expression in the intestinal epithelium, the receptor subtype-specific effect on COX-2 expression in a rat intestinal epithelial cell line (RIE-1) has been investigated. Ang II dose- and time-dependently increased the expression of COX-2, but not COX-1 mRNA and protein. This stimulation was completely blocked by the AT(1) receptor antagonist but not the AT(2) receptor antagonist. Ang II and lipopolysaccharide (LPS) additively induced COX-2 protein in RIE-1 cells, whereas the LPS-induced COX-2 expression was significantly attenuated by low concentrations of Ang II or the AT(2) agonistic peptide CGP-42112A only in AT(2) over-expressed cells. These data indicate that Ang II bi-directionally regulates COX-2 expression via both AT(1) and AT(2) receptors. Control of COX-2 expression through Ang II signaling may have significance in cytokine-induced COX-2 induction and colon tumorigenesis.  相似文献

Fish‐mint (Houttuynia cordataThunb .), belonging to family Saururaceae, has long been used as food and traditional herbal medicine. The present study was framed to assess the changes occurring in the essential‐oil composition of H. cordata during annual growth and to evaluate allelopathic, antibacterial, antifungal, and antiacetylcholinesterase activities. The essential‐oil content ranged from 0.06 – 0.14% and 0.08 – 0.16% in aerial parts and underground stem, respectively. The essential oils were analysed by GC‐FID, GC/MS, and NMR (1H and 13C). Major constituents of aerial‐parts oil was 2‐undecanone (19.4 – 56.3%), myrcene (2.6 – 44.3%), ethyl decanoate (0.0 – 10.6%), ethyl dodecanoate (1.1 – 8.6%), 2‐tridecanone (0.5 – 8.3%), and decanal (1.1 – 6.9%). However, major constituents of underground‐stem oil were 2‐undecanone (29.5 – 42.3%), myrcene (14.4 – 20.8%), sabinene (6.0 – 11.1%), 2‐tridecanone (1.8 – 10.5%), β‐pinene (5.3 – 10.0%), and ethyl dodecanoate (0.8 – 7.3%). Cluster analysis revealed that essential‐oil composition varied substantially due to the plant parts and season of collection. The oils exhibited significant allelopathic (inhibition: 77.8 – 88.8%; LD50: 2.45 – 3.05 μl/plate), antibacterial (MIC: 0.52 – 2.08 μl/ml; MBC: bacteriostatic) and antifungal (MIC: 2.08 – 33.33 μl/ml; MFC: 4.16 – 33.33 μl/ml) activities. The results indicate that the essential oil from Hcordata has a significant potential to allow future exploration and exploitation as a natural antimicrobial and allelopathic agent.  相似文献

A series of 1,4-naphthoquinone derivatives were synthesized and evaluated for antibacterial and antiviral activities. The structure-activity relationships of these compounds were also studied. The results suggest that compounds 9-22 showed in vitro marked antibacterial activity. Compounds 4c and 7a showed inhibitory effect against RNA dependent RNA polymerase induced poliovirus type 2 infected HeLa cells.  相似文献

Studies on the structure-function relationship of transporters require the availability of sufficient amounts of the protein in a functional state. In this paper, we report the functional expression, purification, and reconstitution of the human sodium/d-glucose cotransporter1 (hSGLT1) in Pichia pastoris and ligand-induced conformational changes of hSGLT1 in solution as studied by intrinsic tryptophan fluorescence. hSGLT1 gene containing FLAG tag at position 574 was cloned into pPICZB plasmid, and the resulting expression vector pPICZB-hSGLT1 was introduced into P. pastoris strain GS115 by electroporation. Purification of recombinant hSGLT1 by nickel-affinity chromatography yields about 3 mg of purified recombinant hSGLT1 per 1-liter of cultured Pichia cells. Purified hSGLT1 migrates on SDS-PAGE with an apparent mass of 55 kDa. Kinetic analysis of hSGLT1 in proteoliposomes revealed sodium-dependent, secondary active, phlorizin-sensitive, and stereospecific alpha-methyl-d-glucopyranoside transport, demonstrating its full catalytic activity. The position of the maximum intrinsic tryptophan fluorescence and titration with hydrophilic collisional quenchers KI, acrylamide, and trichloroethanol suggested that most of Trps in hSGLT1 in solution are in a hydrophobic environment. In the presence of sodium, sugars that have been identified earlier as substrate for the transporter increase intrinsic fluorescence in a saturable manner by a maximum of 15%. alpha-Methyl-d-glucopyranoside had the highest affinity (K(d) = 0.71 mM), followed by d-glucose, d-galactose, d-mannose, and d-allose which showed a much lower affinity. l-Glucose was without effect. d-Glucose also increased the accessibility of the Trps to hydrophilic collisional quenchers. On the contrary phlorizin, the well-established inhibitor of SGLT1, decreased intrinsic fluorescence by a maximum of 50%, and induced a blue shift of maximum (5 nm). Again, the effects were sodium-dependent and saturable and a high affinity K(d) of 5 muM was observed. In addition the surface of hSGLT1 was labeled with 1-anilinonaphthalene-8-sulfonic acid, a reporter molecule for the surface hydrophobicity. In the presence of sodium, addition of d-glucose decreased ANS fluorescence whereas phlorizin increased ANS fluorescence. Thus three conformational states of SGLT1 could be defined which differ in their packing density and hydrophobicity of their surface. They reflect properties of the empty carrier, the d-glucose loaded carrier facing the outside of membrane and the complex of the outside-orientated carrier with phlorizin.  相似文献

Fermentation processes have been developed for producing most of the essential amino acids. Methionine is one exception. Although microbial production of methionine has been attempted, no commercial bioproduction exists. Here, we discuss the prospects of producing methionine by fermentation. A detailed account is given of methionine biosynthesis and its regulation in some potential producer microorganisms. Problems associated with isolation of methionine overproducing strains are discussed. Approaches to selecting microorganism having relaxed and complex regulatory control mechanisms for methionine biosynthesis are examined. The importance of fermentation media composition and culture conditions for methionine production is assessed and methods for recovering methionine from fermentation broth are considered.  相似文献

Coal combustion produces carbon dioxides, SOx, NOx and a variety of byproducts, including fly-ash, flue gas and scrubber sludge. Fly-ash consists of minute glass-like particles and its deposition on leaves inhibits the normal transpiration and photosynthesis of plants. Fly-ash also affects the physicochemical characteristics of soil because it is generally very basic, rich in various essential and non-essential elements, but poor in both nitrogen and available phosphorus. The massive fly-ash materials have been a potential resource for the agricultural activities as well as the other industrial purposes. Practical value of fly-ash in agriculture as an 'effective and safe' fertiliser or soil amendment can be established after repeated field experiments. Here remains to be disclosed the biological processes and interactions due to 'lack and excess' of the fly-ash exposures along with abiotic and biotic factors. These may involve the symbiotic fixation of nitrogen and the biological extraction of metals following immobilisation of toxic heavy metal ions, as well as other neutralisation and equilibration processes during weathering. Nitrogen-fixing plants with an apparent heavy metal-tolerance can be helpful as the early colonisers of fly-ash dumps and nearby areas. Electronic Publication  相似文献
