Sequence features and phylogenetic analysis of the stress protein hsp90alpha in chinook salmon (Oncorhynchus tshawytscha), a poikilothermic vertebrate.

We cloned and sequenced a chinook salmon Hsp90 cDNA; sequence analysis shows it to be Hsp90alpha. Phylogenetic analysis supports the hypothesis that alpha and beta paralogs of Hsp90 arose as a result of a gene duplication event and that they diverged early in the evolution of vertebrates, before tetrapods separated from the teleost lineage. Among several differences distinguishing poikilothermic Hsp90alpha sequences from their bird and mammal orthologs, the teleost versions specifically lack a characteristic QTQDQP phosphorylation site near the N-terminus. We used the cDNA to develop an RNA (Northern) blot to quantify cellular Hsp90 mRNA levels. Chinook salmon embryonic (CHSE-214) cells responded to heat shock with a rapid rise in Hsp90 mRNA through 4 h, followed by a gradual decline over the next 20 h. Hsp90 mRNA level may be useful as a stress indicator, especially in a laboratory setting or in response to acute heat stress.     

Biochemical changes in tomatoes stored in modified gas atmospheres. I. Sugars and acids

During long-term storage in modified gas atmospheres (MGA), some of the biochemical changes occurring in tomato fruit, cv. Sleaford Abundance, were compared with those in fruit of some Israeli cultivars which had been selected for firmness and slow ripening. The MGA used in the experiments did not succeed in completely restricting changes in sugars and acids during storage, although colour change was prevented. This separation of processes is discussed in relation to the practice of long-term storage and its implications for fruit flavour.     

How reliable are dung counts for estimating elephant numbers?

Dung counts are the most commonly used techniques for estimating elephant numbers in forests, yet there is considerable scepticism concerning their accuracy. Published accounts of dung counts show that they give estimates similar to those from other methods for vertebrates ranging in size from lizards to elephants. For ungulates, macropods and elephants there are strong correlations between estimates from dung counts and other methods. Thus, dung counts are as accurate or inaccurate as other methods for estimating vertebrate numbers, including elephants. Dung counts for elephants give estimates that are as precise as, and sometimes more precise than, those from aerial surveys of elephants. This is because the variance in dung density is usually low and results in a lower than expected variance for the final elephant estimate when combined with the variances of defecation and decay rates. Dung counts may be more appropriate than aerial surveys for monitoring small or declining elephant populations.     

Population cycles can maintain foraging polymorphism

The maintenance of genetic variability in morphological traits that affect fitness is poorly understood. We present a simple Mendelian model of genetic traits affecting foraging efficiency in grazing ungulates, based on a trade-off between rates of energy extraction at low versus high levels of plant abundance. The model suggests that variation in foraging efficiency could be maintained via lottery competition arising as a direct consequence of dynamically unstable interactions between consumers and their food resources. Lottery competition is a plausible mechanism for explaining wide variability in foraging efficiency, such as that documented in unstable Soay sheep populations on the St Kilda archipelago.     

Maximization of pulmonary hygroscopic aerosol deposition

A newly developed computer model is used to predict the aqueous salt solution concentration, breathing pattern, and inhaled droplet size distribution parameters that will maximize pulmonary deposition of hygroscopic medicinal aerosols. The parameter values providing maximum pulmonary deposition include 1) a NaCl concentration in the aerosolized solution of 0.035 g/ml or higher if the subject can tolerate it, 2) as nearly a monodispersed inhaled aerosol size distribution as possible, 3) an aerosol mass median diameter of 2-3 micron, and 4) slow (7 breaths/min) uninterrupted breathing of 1.5-2 liters of aerosol/breath. With these values, the model predicts that pulmonary deposition can be increased by greater than 100% relative to the deposition achieved in conventional inhalation therapy with isotonic saline-based medications.     

Improved assay of coenzyme F420 analogues from methanogenic bacteria

Summary An improved method for separating analogues of coenzyme F₄₂₀ by isocratic reversed-phase high performance liquid chromatography is described. The method offers improved resolution, shorter chromatography runs and requires less complex apparatus.
