Alzheimer's disease is characterized by the deposits of the 4-kDa amyloid beta peptide (A beta). The A beta protein precursor (APP) is cleaved by beta-secretase to generate a C-terminal fragment, CTF beta, which in turn is cleaved by gamma-secretase to generate A beta. Alternative cleavage of the APP by alpha-secretase at A beta 16/17 generates the C-terminal fragment, CTFalpha. In addition to A beta, endoproteolytic cleavage of CTF alpha and CTF beta by gamma-secretase should yield a C-terminal fragment of 57-59 residues (CTF gamma). However, CTF gamma has not yet been reported in either brain or cell lysates, presumably due to its instability in vivo. We detected the in vitro generation of A beta as well as an approximately 6-kDa fragment from guinea pig brain membranes. We have provided biochemical and pharmacological evidence that this 6-kDa fragment is the elusive CTF gamma, and we describe an in vitro assay for gamma-secretase activity. The fragment migrates with a synthetic peptide corresponding to the 57-residue CTF gamma fragment. Three compounds previously identified as gamma-secretase inhibitors, pepstatin-A, MG132, and a substrate-based difluoroketone (t-butoxycarbonyl-Val-Ile-(S)-4-amino-3-oxo-2, 2-difluoropentanoyl-Val-Ile-OMe), reduced the yield of CTF gamma, providing additional evidence that the fragment arises from gamma-secretase cleavage. Consistent with reports that presenilins are the elusive gamma-secretases, subcellular fractionation studies showed that presenilin-1, CTF alpha, and CTF beta are enriched in the CTF gamma-generating fractions. The in vitro gamma-secretase assay described here will be useful for the detailed characterization of the enzyme and to screen for gamma-secretase inhibitors.  相似文献

Previous studies have reported a protective role for the von Hippel-Lindau (VHL) gene products against pro-apoptotic cellular stresses, but the mechanisms remain unclear. In this study, we examined the role of VHL in renal cells subjected to chemical hypoxia, using four VHL-negative and two VHL-positive cell lines. VHL-negative renal carcinoma cells underwent apoptosis following chemical hypoxia (short-term glucose deprivation and antimycin treatment), as evidenced by morphologic changes and internucleosomal DNA cleavage. Reintroduction of VHL expression prevented this apoptosis. VHL-negative cells displayed a significant (greater than 5-fold) activation of caspase 9 and release of cytochrome c into the cytosol following chemical hypoxia. In contrast, VHL-positive cells showed minimal caspase 9 activation, and absence of cytochrome c release under the same conditions. Caspase 8 was only minimally activated in both VHL-negative and -positive cells. In addition, VHL-positive cells displayed a striking up-regulation of Bcl-2 expression (5-fold) following chemical hypoxia. Antisense oligonucleotides to Bcl-2 significantly down-regulated Bcl-2 protein expression in VHL-positive cells and rendered them sensitive to apoptosis. Overexpression of Bcl-2 in VHL-negative cells conferred resistance to apoptosis. Our results suggest that VHL protects renal cells from apoptosis via Bcl-2-dependent pathways.  相似文献

Photochemical efficiencies of photosystem I (PSI) and photosystem II (PSII) were studied in dry thalli of the lichen Hypogymnia physodes and during their re-hydration. In dry thalli, PSII reaction centers are photochemically inactive, as evidenced by the absence of variable chlorophyll (Chl) fluorescence, whereas the primary electron donor of PSI, P700, exhibits irreversible oxidation under continuous light. Upon application of multiple- and, particularly, single-turnover pulses in dry lichen, P700 oxidation partially reversed, which indicated recombination between P700+ and the reduced acceptor FX of PSI. Re-wetting of air-dried H. physodes initiated the gradual restoration of reversible light-induced redox reactions in both PSII and PSI, but the recovery was faster in PSI. Two slow components of P700+ reduction occurred after irradiation of partially and completely hydrated thalli with strong white light. In contrast, no slow component was found in the kinetics of re-oxidation of QA, the reduced primary acceptor of PSII, after exposure of such thalli to white light. This finding indicated the inability of PSII in H. physodes to provide the reduction of the plastoquinone pool to significant levels. Antibodies specific to recombinant filarial antigens Wb-SXP-1 and Bm-SXP-1 have been used to develop a sandwich ELISA for the detection of circulating filarial antigen (CFA) in sera from patients with lymphatic filariasis caused by Wuchereria bancrofti of Brugia malayi. In patients with W. bancrofti infections, a high proportion of microfilaria (mf) positive (MF) and low proportions of patients with chronic pathology (CP) and endemic normals (EN) showed the presence of CFA. Similarly in patients with brugian infections a high proportion of mf positive individuals contained CFA while none of the patients with chronic pathology or endemic normals showed the presence of CFA. Sera from patients with other parasitic infections (OPI) like O. volvulus, Loa loa, Ascaris lumbricoides and from individuals residing in areas non-endemic to filariasis did not exhibit any reactivity. This assay shows promise for the detection of microfilaremic infections in lymphatic filariasis and its usefulness as a diagnostic tool especially in B. malayi infections, needs to be further evaluated.  相似文献

A high-performance thin-layer chromatographic (HPTLC) method for the assay of flurbiprofen in plasma is reported. The drug was extracted from acidified plasma with hexane–diethyl ether (80:20). The mobile phase composition was n-hexane–ethyl acetate–glacial acetic acid (60:30:10). Densitometric analysis of flurbiprofen was carried out at 247 nm. The calibration curves of flurbiprofen in methanol and in plasma were linear in the range 40–400 ng. The mean values of correlation coefficient, slope and intercept were 0.995±0.003, 0.075±0.002 and 4.39±0.05 for standard curves in methanol and 0.992±0.002, 0.066±0.007 and 3.40±0.72 for standard curves in plasma, respectively. The limit of quantitation for flurbiprofen in human plasma was 40 ng, and no interference was found from endogenous compounds. The recovery of flurbiprofen from human plasma using the described extraction procedure was about 87%. The coefficient of variation for within-day and between-day analyses was 2.53% and 3.96% for 200 ng and 1.76% and 2.30% for 400 ng flurbiprofen concentration, respectively. The method was utilized to monitor plasma concentration of flurbiprofen post administration of sustained release capsules in human patient volunteers.  相似文献

We have examined the conformational properties of poly(dG-m5dC) under a variety of low salt conditions and sample preparations. Extensive dialysis against 0.5 mM Na-cacodylate resulted in a left-handed polynucleotide conformation as determined by circular dichroism, in agreement with recently reported results. Similarly, extensive dialysis against Tris-EGTA also led to a left-handed conformation. Dilution of these samples led to a transition to the right-handed conformation. More stringent treatments such as dialysis followed by passage over an ion exchange column also resulted in a right-handed conformation. When these various solutions were examined using atomic absorption spectroscopy, significant levels of Mg+2 were observed (greater than or equal to 190 per 1000 nucleotides) in all samples showing a left-handed form, while much lower levels (less than or equal to 45 per 1000 nucleotides) were found in the low salt samples displaying a right-handed conformation. Addition of MgCl2 to samples in which divalent cations had been almost completely removed led to the reformation of the left-handed form. These results indicate that the left-handed form seen under certain low salt conditions is due to the presence of Mg+2 ions that remain bound to the polynucleotide, even in the presence of EDTA.  相似文献

Berberine (BBR), a traditional Chinese phytomedicine extracted from various parts of Berberis plants, is an isoquinoline alkaloid used for centuries to treat diabetes, hypercholesterolemia, hypertension, and so forth. It has recently received immense attention worldwide to treat cancer due to its potent pro-apoptotic, antiproliferative, and anti-inflammatory properties. BBR efficiently induces tumor apoptosis, replicative quiescence and abrogates cell proliferation, epithelial–mesenchymal transition, tumor neovascularization, and metastasis by modulating diverse molecular and cell signaling pathways. Furthermore, BBR could also reverse drug resistance, make tumor cells sensitive to current cancer treatment and significantly minimize the harmful side effects of cytotoxic therapies. This review comprehensively analyzed the pharmacological effects of BBR against the development, growth, progression, metastasis, and therapy resistance in wide varieties of cancer. Also, it critically discusses the significant limitations behind the development of BBR into pharmaceuticals to treat cancer and the future research directions to overcome these limitations.  相似文献

Successful vaginal and bicornual cannulations were performed with a tabular cannulator shaped like a giant hypodermic needle in corssbred (Bos indicus x Bos taurus ) heifers (n=7). A total of 58 embryos from a possible 73 were flushed out. The embryo recovery rate was 79.45% and average recovery time was 50 minutes. The wound healing at cannulation sites was rapid. All heifers except one conceived due to insemination during subsequent cycles. One heifer developed pyometra and did not settle.  相似文献

Antiserum to cell wall proteins of S. typhi showed immunoprecipitin lines of identity with homologous antigen, normal human serum and human transferrin. Also, anti-transferrin antibody showed immunoprecipitin band with S. typhi cell wall proteins and agglutinated S. typhi whole cells, but not Eschericia coli. It is speculated that transferrin or a transferrin-like protein present in the cell wall may play a role in iron-uptake by S. typhi.  相似文献
