Drug delivery systems are designed to improve therapy efficacy as well as patient compliance. This could be accomplished by specifically targeting a medication intact to its active site, therefore reducing side-effects and enabling high local drug concentrations. Silica nanoparticles have gained ground in the biomedical field for their biocompatibility and biodegradability, being themselves inert and stable, thus enabling a variety of formulation designs for application in the pharmaceutical industry. This paper is a review of the recent patents on the applications of silica nanoparticles for drug delivery and their preparation. The review will focus on the different techniques available to obtain silica nanoparticles with variable morphology and their drug targeting applications, providing an overview of silica particles synthesis described in the literature.  相似文献

Shifts of gaze and shifts of attention are closely linked and it is debated whether they result from the same neural mechanisms. Both processes involve the frontal eye fields (FEF), an area which is also a source of top-down feedback to area V4 during covert attention. To test the relative contributions of oculomotor and attention-related FEF signals to such feedback, we recorded simultaneously from both areas in a covert attention task and in a saccade task. In the attention task, only visual and visuomovement FEF neurons showed enhanced responses, whereas movement cells were unchanged. Importantly, visual, but not movement or visuomovement cells, showed enhanced gamma frequency synchronization with activity in V4 during attention. Within FEF, beta synchronization was increased for movement cells during attention but was suppressed in the saccade task. These findings support the idea that the attentional modulation of visual processing is not mediated by movement neurons.  相似文献

Attention increases the contrast gain of V4 neurons, causing them to respond to an attended stimulus as though its contrast had increased. When multiple stimuli appear within a neuron's receptive field (RF), the neuron responds primarily to the attended stimulus. This suggests that cortical cells may be "hard wired" to respond preferentially to the highest-contrast stimulus in their RF, and neural systems for attention capitalize on this mechanism by dynamically increasing the effective contrast of the stimulus that is task relevant. To test this, we varied the relative contrast of two stimuli within the recorded neurons' RFs, while the monkeys attended away to another location. Increasing the physical contrast of one stimulus caused V4 neurons to respond preferentially to that stimulus and reduced their responses to competing stimuli. When attention was directed to the lower-contrast stimulus, it partially overcame the influence of a competing, higher-contrast stimulus.  相似文献

It is shown that a sigmoid dose vs. response curve will occur in any biological system in which the stimulant compound must diffuse significant distances to reach responding cells, even when the cells themselves respond according to Michaelis-Menten kinetics. Isolated pancreatic islets releasing insulin in response to glucose stimulation have been used as a specific example. Since diffusion and/or other physical processes can produce global effects which could account for the sigmoidal nature of a dose vs. response curve, the existence of complex molecular mechanisms of hormone-receptor interaction can not be inferred solely from the character of a dose vs. response relation.  相似文献

Flow cytometry sex-sorting technology was developed in 1989. However, it is only the bovine species in which offspring of the desired sex are obtained at a commercial level. The aim of the present work was to evaluate efficiency parameters when using fresh sexed semen in a large-scale equine commercial embryo transfer program. During the 2009, 2010 and 2011 breeding seasons, 938 synchronized cycles were artificially inseminated. One hundred (10.6%) mares failed to ovulate, and for the remaining 838 useable cycles, 887 doses of sexed semen were used, representing 1.06 doses per cycle. In general, 435 (51.9%) out of 838 flushing performed resulted in the recovery of at least one embryo and 496 (59.1%) embryos were recovered, including twins and triplets. Pregnancy rate at 25 days achieved 81.5% (one embryo transferred per recipient). Embryo recovery rate was not statistically different either between preovulatory and postovulatory artificially inseminated mares or when increased quantities of sexed sperm per dose were used (15–45 million) (P > 0.05). A broad variation in embryo recovery rate was observed between the different stallions used in this study. Sex accuracy of the sex sorting assessed by ultrasound fetal sex determination was 90.3%. Finally, overall efficiency (female embryo pregnancies per useable cycles) was 39% (325/838), meaning that to obtain a female pregnancy of at least 75 days it was necessary to perform 2.5 flushing.  相似文献

The recovery of free purine and pyrimidine bases and their degradation products represent alternative pathways in plant cells either to synthesize nucleotides (salvage pathways) by low energy consumption or to reuse organic nitrogen. Such recycling of metabolites often requires their uptake into the cell by specialized transport systems residing in the plasma membrane. In plants, it has been suggested that several protein families are involved in this process, but only a few transporters have so far been characterized. In this work, gene expression, substrate specificities, and transport mechanisms of members of the Ureide Permease family in Arabidopsis (AtUPS) were analyzed and compared. Promoter-GUS studies indicated that the members of the family have distinct and partially overlapping expression patterns with regard to developmental stages or tissue specific localization. In addition, two alternative splice variants of AtUPS5, a novel member of the transporter family, were identified and investigated. The abundance of both alternative mRNAs varied in different organs, while the relative amounts were comparable. AtUPS5l (longer isoform) shares similar structural prediction with AtUPS1 and AtUPS2. In contrast, AtUPS5s (shorter isoform) lacks two transmembrane domains as structural consequence of the additional splice event. When expressed in yeast, AtUPS5l mediates cellular import of cyclic purine degradation products and pyrimidines similarly to AtUPS1 and AtUPS2, but differences in transport efficiencies were observed. AtUPS5s, however, could not be shown to mediate uptake of these compounds into yeast cells and might therefore be defective or have a different function.  相似文献

PACSIN2 regulates cell adhesion during gastrulation in Xenopus laevis   总被引:1,自引:0,他引:1  
We previously identified the adaptor protein PACSIN2 as a negative regulator of ADAM13 proteolytic function. In Xenopus embryos, PACSIN2 is ubiquitously expressed, suggesting that PACSIN2 may control other proteins during development. To investigate this possibility, we studied PACSIN2 function during Xenopus gastrulation and in XTC cells. Our results show that PACSIN2 is localized to the plasma membrane via its coiled-coil domain. We also show that increased levels of PACSIN2 in embryos inhibit gastrulation, fibronectin (FN) fibrillogenesis and the ability of ectodermal cells to spread on a FN substrate. These effects require PACSIN2 coiled-coil domain and are not due to a reduction of FN or integrin expression and/or trafficking. The expression of a Mitochondria Anchored PACSIN2 (PACSIN2-MA) sequesters wild type PACSIN2 to mitochondria, and blocks gastrulation without interfering with cell spreading or FN fibrillogenesis but perturbs both epiboly and convergence/extension. In XTC cells, the over-expression of PACSIN2 but not PACSIN2-MA prevents the localization of integrin β1 to focal adhesions (FA) and filamin to stress fiber. PACSIN2-MA prevents filamin localization to membrane ruffles but not to stress fiber. We propose that PACSIN2 may regulate gastrulation by controlling the population of activated α5β1 integrin and cytoskeleton strength during cell movement.  相似文献

A specialized database (DB) for Arabidopsis membrane proteins, ARAMEMNON, was designed that facilitates the interpretation of gene and protein sequence data by integrating features that are presently only available from individual sources. Using several publicly available prediction programs, putative integral membrane proteins were identified among the approximately 25,500 proteins in the Arabidopsis genome DBs. By averaging the predictions from seven programs, approximately 6,500 proteins were classified as transmembrane (TM) candidate proteins. Some 1,800 of these contain at least four TM spans and are possibly linked to transport functions. The ARAMEMNON DB enables direct comparison of the predictions of seven different TM span computation programs and the predictions of subcellular localization by eight signal peptide recognition programs. A special function displays the proteins related to the query and dynamically generates a protein family structure. As a first set of proteins from other organisms, all of the approximately 700 putative membrane proteins were extracted from the genome of the cyanobacterium Synechocystis sp. and incorporated in the ARAMEMNON DB.
