An exteriorized loop of mouse intestine was exposed to 147Pm low-energy electrons, where the dose rate decreased by a factor of 5 from the base of the crypt to the top of the proliferative zone. A crypt survival curve was obtained, expressed in terms of exposure time. The shape of the curve was interpreted in terms of survival parameters for colony-forming cells (clonogens) derived using 137Cs gamma rays and the depth-dose curve measured for 147Pm electrons. It is concluded that the shape of the crypt survival curve using 147Pm electrons is inconsistent with the notion of either the presence of a large number of clonogens or a small number near the top of the proliferative zone. A computer fitting procedure showed that the best agreement between predicted and observed curves was achieved with 2.7 +/- 0.5 clonogens at cell position 5.6 +/- 0.6, in the putative stem-cell zone. 相似文献

Physical efficiency of mice and rats at early times following irradiation with doses of 35 to 200 Gy was estimated by the muscle endurance and motor coordination indices. A threshold nature of the effects was exhibited at high doses. For instance, as large as 40 Gy was the dose threshold at which stability in the accomplishment of the known operations was impaired; at 70-100 Gy, the accomplishment decreased down to 50% of the initial level, by the 2nd hour after irradiation, with the subsequent partial restoration; 200 Gy was the threshold dose at which the disturbances were irreversible. Proceeding from an x-ray examination of 143 patients with mandibular fractures, the most informative methods and projections as well as the order of their use with account of the site of a fracture, were determined. The most informative method in all fracture sites was orthopantomography; panoramic roentgenography was indicated only in fractures of the frontal part. If panoramic tomography cannot be done, roentgenography or electroroentgenography in the frontal-nasal projection is recommended. If panoramic roentgenography cannot be done, extraoral roentgenography is indicated in the diagnosis of fractures of the frontal part of the mandible. 相似文献

A new approach to selection of lines of spontaneously transformed cells from the rat embryo "precrisis" cultures is described and their phenotypes at the initial and advanced stages during a long-term cultivation are characterized. The new selective system, referred to as 2T7, differs from the well known 3T3, 2T6 and 3T12 systems (Todaro, Green, 1963; Aaronson, Todaro, 1968). It is based on the maintenance of cultures under maximum cell densities. Such an approach facilitated and accelerated the start of the "crisis" stage (up to 3-8 passages) with the following gradual death of almost the whole normal senescent cell population, the colony formation resulting from the proliferation of single clonogenic cells. The frequency of clonogenic cells was about 6 x 10(-6). Six lines of spontaneously transformed cells from embryos of noninbred white rats (LRec-1--LRec-6) and one line from the Wistar embryos (LRec-7) were established. All the lines are characterized as diploid or near-tetraploid, with 1-4 different marker chromosomes formed from chromosome 7, as was reported elsewhere (Artsybasheva et al., 1988). The values of saturation densities and the time of population doubling for all the 7 lines differed from those for the rat embryo primary cultures cells. LRec-1--LRec-6 cells were unable to form the colonies in soft agar, while LRec-7 cells were able to grow in agar. The lines LRec became oncogenic for 1-2 day old rats after different periods of cultivation in vitro--from 3 to 7 months. The line LRec-7 Wistar appeared to be highly oncogenic from the very beginning after its selection. The histological analysis revealed that the LRec-1 tumors could be classified as polymorphocellular sarcoma. Up to 20 passages the LRec-1 line had numerous clonogenic cells (50-60%) in sparse cultures independently on the serum content in the media. By a 3-step selection of LRec-1 cells, on cultivation in media with lower serum contents (1-0.1-0%), a semisuspension of LRec-1sf subline (serum free) was established. This line was highly oncogenic for 1-2 day old rats, was easily cryopreserved and proliferated in the serum-free media for unlimited time, forming small colonies in agar. Membrane proteins with a molecular weight of 290, 180, and 55 kDa were isolated using immunosorbent attached to sepharose and rabbit antibodies to cytoplasmic tetrodotoxin-sensitive protein from beef brain gray matter. A technique used for research into voltage-dependent sodium channels was applied to reconstruction of these proteins and investigation of toxin-dependent sodium flows through the lipoprotein membrane. Findings are interpreted as evidence of the similarity between cytoplasmic tetrodotoxin-sensitive protein and that of sodium channels at the cell membrane.A. A. Low-energy peptide backbone structures of dermorphin (DM), amide of its N-terminal pentapeptide (DM 1-5) and DM 1-5 analogues with substitutions of Gly4 for Leu, D-Gln, Aal or Tal were determined by energy calculations. The above analogues were shown to possess different affinities toward opiate receptors of mu-type. The comparison of low-energy backbone structures of DM, DM 1-5 and its analogues resulted in development of the dermorphin "biologically active" conformation being characteristic of its binding with mu-type receptors. The specific binding of dermorphin to this receptor apparently depends on the conformation of the whole N-terminal pentapeptide. 相似文献

Modification of 5'-32P-labelled octadecadeoxyribonucleotide d(pC5A8C5) (III) with octathymidylate methylphosphonate derivatives bearing both 3'- and 5'-terminal alkylating 4-(N-2-chloroethyl-N-methylamino)benzylphosphoamide residue has been investigated. Yield in the modification depends on configuration of methylphosphonate fragment, in case of Rp-isomer it may amount to 90%. Tetrahymena cells treated with insulin in mass cultures were separated to single-cell clones or one of the "sister-cells" of dividing Tetrahymena (in single-cell culture) was treated with insulin. In both cases the FITC-insulin binding of sister-cells were compared. The insulin imprinting significantly increased the insulin binding of cells. There was also a significant difference between the imprinted and not imprinted sisters as well as between the not imprinted sisters. This demonstrates the existence of a difference (in hormone binding) between sister-cells and justifies that the information of the first hormone treatment (imprinting) is not equally divided between the sister-cells. 相似文献

The bacterium Xanthomonas campestris, which synthesizes the commercially important polysaccharide xanthan, was grown aseptically in 1.2 L fermenters using semicontinuous cell culture technique (d' = 0.0035 h-1). The effects of carbon-substrate concentration on xanthan production were investigated at three initial glucose concentrations (Go = 15, 20, 25 g/L). Cell biomass synthesis was nitrogen-limited by use of a chemically defined medium that contained NH3-N as the sole nitrogen source at a concentration where it was exhausted before glucose. A linear relationship between biomass synthesis and NH3-N depletion was observed. This relationship remained valid only until NH3-N exhaustion, after which biomass concentration slowly rose another 20 percent before declining. Another linear relationship was found between xanthan synthesis and glucose uptake. This relationship was unaffected by the disappearance of NH3-N and held through glucose exhaustion. The quasi-stoichiometric yield coefficients obtained for each linear relationship were not affected by G0-. Biomass synthesis kinetics showed no variation with G0 before NH3-N exhaustion; afterwards, cell biomass decline was delayed by increasing G0. Xanthan synthesis kinetics displayed no detectable response to depletion of NH3-N and plateauing of biomass concentration; however, there was a marked slow down in the net rate of xanthan synthesis and a drop in xanthan yield after cell biomass decline became noticeable. 相似文献
