The disamed Ti-binary vector pBIN 6 in Agrobacterium tumefaciens has been used in leaf disc transfomations to produce transgenic apple (Malus pumila Mill.) plants with a nomal phenotype except for a somewhat reduced capacity to root. The presence of the genes for nopaline synthase and neomycin phosphotrans ferase (conferring kanamycin resistance), inserted into the host genome by the vector, was confirmed by Southern blot analysis, the detection of nopaline synthase activity and rooting in the presence of the antibiotic.The nopaline synthase gene continued to be expressed in glasshouse-grown plants several months after removal from in vitro growth conditions.  相似文献

In short-term field trials at combinations of ambient temperature (°C) and insolation (W·m−2), larval Colorado potato beetles (Leptinotarsa decemlineata [Say] [Coleoptera: Chrysomelidae]) were observed after their release on the adaxial surface of leaflets on potato plants (Solanum tuberosum L. Solanaceae). The larvae either began feeding or moved under the leaflet; mean interval from release to expression of these behaviors (2.9±0.05 min [n =358]) was independent of air temperature and insolation. Proportion of larvae moving under the leaflet increased logistically with both air temperature and insolation. A 1 W·m−2 change in insolation (P) evoked the same effect on this proportion as a 0.0838 °C change in air temperature (T a ), so the two quantities were combined as T*=T a +P·0.0838 °C/(W·m−2), which has units of °C. The proportion of larvae moving under the leaflet increased logistically with T*. In 1-day field trials we monitored air temperature, insolation and proportion of larvae under the leaflet, and compared the latter to predictions from the logistic regression derived from the short-term trials. Consistently more larvae occurred under leaflets than predicted from the logistic regression; this bias diminished as T* increased until at T*≥40 °C, observed and predicted proportions were equal. This pattern of deviation from the predictions of the logistic regression is consistent with a thermoregulatory strategy in which larvae move away from hostile conditions, rather than seek optimal conditions.  相似文献

The IGFs (IGF-I and IGF-II) are essential for normal mammalian growth and development. Their actions are mediated primarily by their interactions with the type I IGF receptor (IGF-I receptor), a transmembrane tyrosine kinase. The ligands and the IGF-I receptor are structurally related to insulin and to the insulin receptor, respectively. Analysis of evolutionary conservation has often provided insights into essential regions of molecules such as hormones and their receptors. The genes for insulin and IGFs have been partially characterized in a number of vertebrate species extending evolutionarily from humans as far back as fish. The sequences of the exons encoding the mature insulin and IGF peptides are highly conserved among vertebrate species, and IGF-I-Iike molecules are found in species whose origins extend back as much as 550 million years. The insulin receptor is also highly conserved in vertebrate species, and an insulinreceptor-like molecule has been characterized in Drosophila. In contrast, IGF-I receptors have only been characterized in mammalian species and partially studied in Xenopus, in which the tyrosine kinase domain is highly conserved. Studies are presently being undertaken to analyze in more detail the regulation of the genes encoding this important family of growth factors and the structure/function relationships in the gene products themselves. © 1993 Wiley-Liss, Inc.  相似文献

The psaA and psaB genes of the chloroplast genome in oxygenic photosynthetic organisms code for the major peptides of the Photosystem 1 reaction center. A heterodimer of the two polypeptides PsaA and PsaB is thought to bind the reaction center chlorophyll, P700, and the early electron acceptors A0, A1 and Fe-SX. Fe-SX is a 4Fe4S center requiring 4 cysteine residues as ligands from the protein. As PsaA and PsaB have only three and two conserved cysteine residues respectively, it has been proposed by several groups that Fe-SX is an unusual inter-peptide center liganded by two cysteines from each peptide. This hypothesis has been tested by site directed mutagenesis of PsaA residue C575 and the adjacent D576. The C575D mutant does not assemble Photosystem 1. The C575H mutant contains a photoxidisable chlorophyll with EPR properties of P700, but no other Photosystem 1 function has been detected. The D576L mutant assembles a modified Photosystem 1 in which the EPR properties of the Fe-SA/B centers are altered. The results confirm the importance of the conserved cysteine motif region in Photosystem 1 structure.Dedicated to the memory of Daniel I. Arnon.  相似文献

Abstract. A qualitative bioassay was adapted and used to determine the ability of the early post-implantation mouse embryo to synthesise interferon. Interferon production was not seen in any embryo tissue in the absence of an inducer and could only be detected in virus-induced tissue from the early 7th day of development. This induced interferon synthesis was initially confined to the trophoblast of the early 7th day embryo. It was then found in tissues of both trophoblast and inner cell mass origin in the early 8th day, and subsequently, in derivatives of the embryonic ectoderm in the 13th-day embryo.  相似文献

The possibility thatChlamydia trachomatis contains peptidoglycan was examined by three methods. Preincubation of chlamydia with enzymes known to cleave peptidoglycan had no adverse effect on the subsequent development. Immunofluorescence studies with antistreptococcal peptidoglycan antisera failed to show any cross reactions with chlamydial antigens. The antichlamydial activity of anti-cell-wall antimicrobials was examined; lactams proved the most active, and cycloserine and bacitracin also showed antichlamydial activity. Alaphosphin, phosphomycin, and vancomycin showed no antichlamydial activity at the concentrations tested.  相似文献

Summary Seventee white-rot and brown-rot fungi were screened for their ability to fractionate the lignocellulose structure of oat straw through the preferential attack of lignin or cellulose. Fermentations were carried out under solid-state conditions with 25 g quantities of straw. The fermented straw was analyzed for weight loss, Klason lignin loss and cellulase digestion. All the fungi attacked both lignin and carbohydrate fractions causing 3–28% weight losses and 26–34 g/100 g enzymatic digestibility. Polyporus tulipiferae, Phanerochaete chrysosporium and Polyporus sp. were tested for the effects of various nitrogen, phosphate and carbon levels, incubation temperatures and incubation time. There is presently a debate concerning the number of phases in fluorescence induction and on the identification of the several possible heterogeneities in PS II centres. However, the usual methods of analysis present numerical problems, including a lack of robustness (robustness being defined as the ability to give the correct answer in the presence of distortions or artefacts). We present here the adaptation of the method of moments, which was developed for robustness, to the analysis of fluorescence induction. We were thus able to identify three phases in the fluorescence induction in the presence of DCMU. The slowest phase was attributed to the centres inactive in plastoquinone reduction by using duroquinone as electron acceptor. In order to compare fluorescence with and without DCMU, we introduced the rate of photochemistry, defined as the product of the area times the rate constant of an exponential. This quantity is invariant for a given centre no matter what the size of the electron acceptor pool is. The two fastest phases in the presence of DCMU were attributed to active centres because their rate of photochemistry was the same as that of the plastoquinone-reducing phases in the absence of DCMU. Summary Reproductive effort was compared in Littorina rudis (ovoviviparous), L. nigrolineata (benthic eggs with direct development), L. neritoides and L. littorea (planktonic eggs and larvae). Three indices of reproductive effort were used: the proportion of total production committed to reproduction per unit time, the cumulative proportion of total production committed to reproduction up to a given age, and the ratio of reproductive production per unit time to somatic biomass at the beginning of the unit time interval. The indices were plotted against age and the snails ranked in order of their reproductive efforts at equivalent ages. When plotted on axes of absolute time, all three indices ranked the snails in the order L. neritoides. The rank order of reproductive efforts on a time axis standardized for growth rate was L. littorea, and on a time axis standardized for generation time was L. neritoidesL. rudis. The implications and relative merits of the different expressions of reproductive effort are discussed. No general relationship was found between the magnitude of reproductive effort and either reproductive type or population ecology among the Littorina spp., or among other intertidal prosobranchs reviewed from the literature. Semelparous species had among the highest reproductive efforts, but there was no clear separation in this respect from certain iteroparous species. Parental investment, measured as the energy content of an egg plus any accompanying jelly or capsular material, was one to two orders of magnitude greater in the species with direct development than in those with indirect development. There were significant differences between egg and hatchling sizes of L. rudis from three contrasted types of shore and these differences were thought to be correlated with the intensity of desiccation and predation on hatchlings rather than with demographic factors.  相似文献
