Krüppel-associated box zinc finger proteins (KRAB-ZFPs) are a huge family of vertebrate-specific repressors that modify gene expression in an epigenetic manner. Despite a well-defined repression mechanism, few biological roles or gene targets of KRAB-ZFP are known. Regulator of sex-limitation 1 (RSL1) is a mouse KRAB-ZFP that enforces male-predominant expression in the liver, affecting body mass and pubertal timing. Here we show that female but not male Rsl1−/− mice gain more weight than wild-type mice on a high-fat diet (HFD) and that key liver and white adipose tissue (WAT) metabolic genes are altered in both Rsl1−/− sexes in response to dietary stress. Expression profiling of Rsl1-sensitive genes in liver and WAT indicates that RSL1 accentuates sex-biased gene expression in liver but greatly diminishes it in WAT. RSL1 expression solely in liver is sufficient to limit diet-induced weight gain and suppress lipogenic genes in WAT, indicating that RSL1 balances metabolism via liver-to-adipose-tissue communication. RSL1''s effects on adult physiology exemplify a significant modulatory capacity of KRAB-ZFPs, in the absence of which there is widespread metabolic dysregulation. This ability to buffer against gene expression noise, coupled with extensive individual genetic variation, highlights the enormous potential of KRAB-Zfp genes as candidate risk factors for complex diseases.  相似文献

Aims Leaf traits of trees exposed to elevated [CO2] in association with other environmental factors are poorly understood in tropical and subtropical regions. Our goal was to investigate the impacts of elevated [CO2] and N fertilization on leaf traits in southern China.Methods Four tree species, Schima superba Gardn. et Champ. (S. superba), Ormosia pinnata (Lour.) Merr (O. pinnata), Castanopsis hystrix AC. DC. (C. hystrix) and Acmena acuminatissima (Blume) Merr. et Perry (A. acuminatissima) were studied in a factorial combination of atmospheric [CO2] (ambient at ~390 μmol mol ? 1 and elevated [CO2] at ~700 μmol mol-1) and N fertilization (ambient and ambient + 100 kg N ha-1 yr-1) in open-top chambers in southern China for 5 years. Leaf mass per unit leaf area (LMA), leaf nutrient concentration and photosynthesis (A sat) were measured.Important findings Results indicated that leaf traits and photosynthesis were affected differently by elevated [CO2] and N fertilization among species. Elevated [CO2] decreased LMA in all species, while N fertilization did not affect LMA. Leaf mass-based N concentration (N M) was significantly greater in O. pinnata and C. hystrix grown in elevated [CO2] but was lower in S. superba. Leaf mass-based P concentration (P M) was significantly greater in C. hystrix and A. acuminatissima exposed to elevated [CO2] but was lower in S. superba. N fertilization significantly increased P M in O. pinnata but decreased P M in S. superba. Photosynthetic stimulation in O. pinnata, C. hystrix and A. acuminatissima was sustained after 5 years of CO2 fumigation. N fertilization did not modify the effects of elevated [CO2] on photosynthesis. Leaf traits (N M, N A, P M, P A) and light-saturated photosynthesis were decreased from the upper to lower canopy. Canopy position did not alter the responses of leaf traits and photosynthesis to elevated [CO2]. Results suggest that photosynthetic stimulation by elevated [CO2] in native species in subtropical regions may be sustained in the long term.  相似文献

To investigate the DNA damage, expression of heat shock protein 70 (Hsp70) and cell proliferation of human lens epithelial cells (hLEC) after exposure to the 1.8 GHz radiofrequency field (RF) of a global system for mobile communications (GSM). An Xc-1800 RF exposure system was used to employ a GSM signal at 1.8 GHz (217 Hz amplitude-modulated) with the output power in the specific absorption rate (SAR) of 1, 2 and 3 W/kg. After 2 h exposure to RF, the DNA damage of hLEC was accessed by comet assay at five different incubation times: 0, 30, 60, 120 and 240 min, respectively. Western blot and RT-PCR were used to determine the expression of Hsp70 in hLECs after RF exposure. The proliferation rate of cells was evaluated by bromodeoxyuridine incorporation on days 0, 1 and 4 after exposure. The results show that the difference of DNA-breaks between the exposed and sham-exposed (control) groups induced by 1 and 2 W/kg irradiation were not significant at any incubation time point (P > 0.05). The DNA damage caused by 3 W/kg irradiation was significantly increased at the times of 0 and 30 min after exposure (P < 0.05), a phenomenon that could not be seen at the time points of 60, 120 or 240 min (P > 0.05). Detectable mRNA as well as protein expression of Hsp70 was found in all groups. Exposure at SARs of 2 and 3 W/kg for 2 h exhibited significantly increased Hsp70 protein expression (P < 0.05), while no change in Hsp70 mRNA expression could be found in any of the groups (P > 0.05). No difference of the cell proliferation rate between the sham-exposed and exposed cells was found at any exposure dose tested (P > 0.05). The results indicate that exposure to non-thermal dosages of RF for wireless communications can induce no or repairable DNA damage and the increased Hsp70 protein expression in hLECs occurred without change in the cell proliferation rate. The non-thermal stress response of Hsp70 protein increase to RF exposure might be involved in protecting hLEC from DNA damage and maintaining the cellular capacity for proliferation.  相似文献

鼎湖山南亚热带天然针阔叶混交林臭氧吸收特征   总被引:1,自引:1,他引:0  
针阔叶混交林是我国南亚热带针叶林向地带性常绿阔叶林演替的中间林分类型,为我国南亚地区主要森林类型,发挥着重要的生态系统服务功能。基于树干液流技术和对臭氧浓度的连续监测,评价该森林类型的臭氧吸收特征和能力有着重要的环境生态学意义。对鼎湖山天然针阔叶混交林优势种马尾松(Pinus manssoniana)、锥栗(Castanopsis chinensis)、木荷(Schima superba)和华润楠(Machilus chinensis)在自然环境条件下的臭氧吸收能力进行了分析研究。结果表明:在日尺度上,4个优势树种的冠层气孔对臭氧导度(GO_3)和臭氧吸收通量(FO_3)均呈单峰型曲线,其最大值的时间在干季(10月至竖年3月)比湿季(4月至9月)滞后;季节尺度上,臭氧浓度在湿季达到最大值48.94 n L/L,湿季GO_3、FO_3和年臭氧吸收累积量(accumulative stomatal O_3flux,AFst)均显著高于干季(P 0.01),华润楠的臭氧吸收能力最强,在干季和湿季可分别达1.11 nmol m~(-2)s~(-1)和1.71nmol m~(-2)s~(-1)。随着水汽压亏缺(VPD)增大,优势种GO_3降低。光合有效辐射(PAR)超过1500 umol m~(-2)s~(-1)时,优势树种GO_3和FO_3呈下降趋势。针阔叶混交林的年臭氧吸收累积量超过了保护森林树木所采用的临界阈值,可认为鼎湖山针阔叶混交林受臭氧危害的潜在风险较高。  相似文献

运用连续7a(1993~1999)的水文观测资料,对南亚热带顶级生态系统鼎湖山季风常绿阔叶林集水区水文要素时空规律进行分析,得到如下一些主要结论:(1)鼎湖山多年平均降水量为1910mm,湿季降水量占年降水量80%,干季仅占20%。6月份的降水量最大,1月份最小。(2)季风常绿阔叶林冠层截留率为31.8%,湿季的截留量占全年截留量的66.7%,截留量最大值和最小值所在的月份分别为7和1月份。各月的截留率差异很大,截留量大的月份,截留率较低;截留量小的月份,截留率较高。(3)季风常绿阔叶林集水区多年平均总径流量953.0mm,总径流系数49.9%,其中地表径流量为252.3mm,地表径流系数13.2%;地表径流与降水量之间存在二次抛物线型回归关系,与降水强度的关系不大,这说明季风常绿阔叶林的产流形式是是蓄满产流。(4)季风常绿阔叶林多年平均蒸散948.2mm,占同期降水量的49.7%;蒸散力1031.4mm,年蒸散系数为0.92,蒸散月变化规律较降水量的月变化规律有所滞后。(5)系统贮水量的月变化很大,2~8月份,系统处于蓄水阶段;9月份至翌年1月份,系统处于失水阶段。蓄水和失水的最大值分别出现在湿季和干季的第一个月,即4月份和10月份。(6)集水区多年平均水量总输入2129.9mm,实际输入1910mm(降水量),其中219.9mm的水量输入是由系统贮水量变化而产生。支出的总水量2129.9mm,实际支出1901.3mm(径流和蒸散量),其中228.6mm的水量支出是由系统贮水量变化引起的。  相似文献

在鼎湖山季风常绿阔叶林设置人工模拟酸雨实验,研究土壤总有机碳含量、微生物量碳含量、土壤p H值和土壤呼吸速率几个指标对不同酸处理梯度(CK:p H值4.5的天然湖水;T1:p H值4.0;T2:p H值3.5;T3:p H值3.0)的响应。结果表明,在模拟酸雨的持续作用下,样地土壤酸化有加剧趋势。2011年的6月(CK:(603.76±46.18)mg/kg,T1:(565.41±44.48)mg/kg,T2:(521.58±30.92)mg/kg,T3:(509.49±19.40)mg/kg)、12月(CK:(488.92±22.71)mg/kg,T1:(379.65±49.46)mg/kg,T2:(346.08±33.81)mg/kg,T3:(318.00±52.35)mg/kg)和2012年6月(CK:(540.48±39.11)mg/kg,T1:(492.30±43.15)mg/kg,T2:(489.65±51.39)mg/kg,T3:(428.53±49.66)mg/kg)3次测定的土壤微生物量碳含量有随模拟酸雨强度增加而显著降低的趋势,高强度的酸处理T3显著低于CK值(P0.05);土壤呼吸速率在各处理中的响应与土壤微生物量碳含量变化一致。由于旱季和湿季的土壤温湿度相差较大,以上各指标在旱湿两季的差异明显,表现为湿季大于旱季。由于土壤总有机碳含量变化缓慢,其在各酸梯度处理下无显著差异(P0.05)。以上结果显示,长期酸雨作用使土壤酸化不断加剧,并降低了土壤微生物量碳的含量,抑制了土壤的呼吸速率,有利于土壤碳的累积,但对土壤总有机碳的影响仍需长期实验研究。  相似文献

The Long-Term Intersite Decomposition Experiment in China (hereafter referred to as LTIDE-China) was established in 2002 to study how substrate quality and macroclimate factors affect leaf litter decomposition. The LTIDE-China includes a wide variety of natural and managed ecosystems, consisting of 12 forest types (eight regional broadleaf forests, three needle-leaf plantations and one broadleaf plantation) at eight locations across China. Samples of mixed leaf litter from the south subtropical evergreen broadleaf forest in Dinghushan (referred to as the DHS sample) were translocated to all 12 forest types. The leaf litter from each of other 11 forest types was placed in its original forest to enable comparison of decomposition rates of DHS and local litters. The experiment lasted for 30 months, involving collection of litterbags from each site every 3 months. Our results show that annual decomposition rate-constants, as represented by regression fitted k-values, ranged from 0.169 to 1.454/year. Climatic factors control the decomposition rate, in which mean annual temperature and annual actual evapotranspiration are dominant and mean annual precipitation is subordinate. Initial C/N and N/P ratios were demonstrated to be important factors of regulating litter decomposition rate. Decomposition process may apparently be divided into two phases controlled by different factors. In our study, 0.75 years is believed to be the dividing line of the two phases. The fact that decomposition rates of DHS litters were slower than those of local litters may have been resulted from the acclimation of local decomposer communities to extraneous substrate.  相似文献
