The hypothesis, derived from experience in Nova Scotia, that artificial pneumothorax, requiring repeated fluoroscopy of the chest, is associated with an increased risk of subsequent breast cancer was tested in Ontario by means of a retrospective cancer patient/control study using record linkage. Sanatorium patients treated by artificial pneumothorax were estimated to have incurred a risk of breast cancer between two and three times as high as that of matched, contemporary sanatorium patients treated by other means. There was a tendency for the malignant disease to develop on the side of collapse, and there was evidence of a characteristic latent interval between exposure and onset.  相似文献

SUMMARY 1. The changes in the vertical distributions of red coloured Planktothrix rubescens and green P. agardhii filaments in Blelham Tarn, English Lake District, were related to vertical profiles of temperature and light attenuation and to continuous records of the surface irradiance and windspeed, from August 1999 to October 2000. 2. The potential growth rate of each organism was calculated from the irradiance and temperature at 0.5 m depths and hourly intervals throughout the year, using algorithms determined from growth rates in culture. The analyses indicated that there was sufficient irradiance for growth, integrated over the 24‐h cycle, at depths down to the metalimnion where the Planktothrix populations stratified in summer. The compensation depth for growth by P. rubescens reached a maximum of 9.3 m in spring and midsummer, and fell to a minimum of 1.6 m in midwinter; the corresponding values for P. agardhii were 7.9 and 0.5 m. 3. The mixed depth (zm) exceeded the critical depth for growth (zb) by P. rubescens (the condition preventing population increase) on only 3 days of the year; for P. agardhii, however, zm exceeded zb on 31 days, contributing to its faster decline. The stratified population of P. rubescens was the major cause of light attenuation during the summer of 2000, and resulted in competitive exclusion of P. agardhii. 4. The calculated growth rates integrated over the depth of the water column in Blelham Tarn equalled, or exceeded, the measured changes of the populations during periods when they were increasing, during summer and autumn. Close agreement between the two values was found for much of the year when allowance was made for dilution of the lake population by rainfall over the watershed. During periods of rapid decline, of P. agardhii in September 1999, P. rubescens in December 1999 and both in July–August 2000, additional losses (e.g. by chytrid parasitism and grazing) are invoked.  相似文献

Among the members of the superfamily of cys-loop ligand-gated ion channels (LGICs) are receptors distinguished by the presence of two cys-loops in the ligand-binding domain, for example, the glycine receptor. Such receptors have thus far been cloned only from vertebrates and from ecdysozoa (arthropods and nematodes). We have now cloned and expressed two 2-cys-loop receptors from Aplysia californica, a lophotrocozoan, and have shown that they form homomeric glutamate receptors. We have also built up a database including the two receptors cloned here, previously cloned vertebrate and ecdysozoan 2-cys-loop receptors taken from GenBank, and the same type of receptors obtained by a search of recently cloned genomes, including two non-vertebrate chordates, an echinoderm, a crustacean, an annelid, and another mollusk. We subjected these receptors to phylogenetic analysis, alone and in combination with GABA-A receptors from the same phyla and from a recently cloned cnidarian. The phylogenetic analysis revealed the presence of two independent clades of glutamate receptors: one from lophotrocozoa and other from ecdysozoa, and suggests that the ancestors of the current 2-cys-loop receptor types diverged from the GABA-A receptors and from each other before the bilateria-cnidaria split. Finally, combining the results from the phylogenetic analysis with those obtained from an analysis of the 2-cys-loop receptors in light of recently published hypotheses concerning the glycine binding pocket, we predict that glycine receptors are not exclusively a vertebrate-receptor type. The effects of sympathetic and parasympathetic agonists and antagonists on discharge of secretory product by the granular and mucous glands were examined in the red-spotted newt, Notopthalmus viridescens viridescens. Observations were made also on the South African clawed toad. Xenopus laevis, the grass frog, Rana pipiens, and the crested newt, Triturus cristatus. In contrast to the granular glands of the South African clawed toad and the grass frog, which were stimulated by alpha-adrenergic agents, those of the red-spotted newt discharge in response to acetylcholine, either in vitro when added to the Holtfreter's solution in which explants were incubated, or in vivo when injected subcutaneously. Granular glands of the crested newt were also dischared in response to subcutaneous injection of acetylcholine. Stimulation of the granular glands by acetylcholine was blocked by atopine but not by tubocurarie, indicating that the cholinergic receptors are muscarinic rather than nicotinic. The mucous glands of the red-spotted newt, on the other hand, did not discharge in response to either acetylcholine or to adrenergic agents.  相似文献

The c-myc oncogene codes for a DNA binding protein that functions in a cell cycle-related manner. A useful model for studying the relationship of c-myc expression with cell cycle kinetics is the HL60 cell line. HL60 cells constitutively express high levels of c-myc mRNA; however, the level can be down-regulated as the cells are induced to differentiate. We have developed a flow cytometric assay for correlating c-myc oncoprotein levels with DNA content. C-myc oncoprotein levels were additionally correlated with c-myc mRNA levels as determined by slot blot hybridization. Dimethylsulphoxide (DMSO) and cytosine arabinoside were used to induce granulocytic and monocytic maturation respectively. Treatment of HL60 cells with DMSO leads to an increase in the per cent of cells in G1/G0 and a decrease in mean c-myc mRNA and oncoprotein levels. The cells with G1 DNA content show the greatest decrease in c-myc protein. ARA-c treatment of HL60 cells leads to a slowing and an accumulation of cells in S phase with a moderate decrease in mean mRNA and only a slight decrease in mean c-myc protein levels. These data support the hypothesis that c-myc is involved in the switch from G1 to G0.  相似文献

In continuing study of gene and genome interaction as a possible etiological mechanism in normal and abnormal growth and malformation, the ventral spinous processes (VSPs, crural insertions of the diaphragm) were used as additional epigenetic variants to portray differences in the basic gradient growth pattern. Over 700 comparisons of mean differences in number (range or magnitude) and peak (position) of the VSP gradient, in the same populations of strains III, DA, and IIIDa (into which the Da gene had been introduced from strain DA), provided populations of +/+, Da/+ and Da/Da on two different genome backgrounds. They reveal the individual effects on the VSPs of underlying growth processes associated with the Da gene dosage, vertebral border shifts and stillbirths. Both Da and stillbirths demonstrate growth influences which interact additively to reduce the range and shift the peak anteriorly in opposition to those of the border shifts. The growth effects induced by border shifts are in some cases significantly so much greater as to seemingly inhibit or even reverse either the specific gene Da or stillbirth effects. The way that interaction of such growth influences can enhance, suppress or cancel each other and the relation to specific growth gradients and functions is of particular importance to understanding the etiology and growth mechanisms of spontaneous and unexpected exogenously or endogenously induced malformations in non-isogenic stocks.  相似文献
