Two ~4 m vertical sequences of basal ice were collected from tunnels dug into the northern lateral margin of Taylor Glacier, McMurdo Dry Valleys, Antarctica. In both cases the basal sequences exhibit two contrasting ice facies groups; clean (debris-free) and banded dispersed (debris-rich). Debris-rich ices exhibit elevated CO2 and depleted O2 concentrations compared to the clean facies. Bacterial cell numbers, respiration rates, and nutrient concentrations are highest in debris-rich layers. Together, our geochemical and biological data indicate that microbial heterotrophic respiration is likely occurring in situ within the basal ice matrix at ambient temperatures near ?15°C. This implies that the basal ice zone of polar glaciers and larger ice sheets is a viable subglacial microbial habitat and active biome of significant volume that has not previously been considered.  相似文献

Mutations in the Park2 gene, encoding the E3 ubiquitin‐ligase parkin, are responsible for a familial form of Parkinson's disease (PD). Parkin‐mediated ubiquitination is critical for the efficient elimination of depolarized dysfunctional mitochondria by autophagy (mitophagy). As damaged mitochondria are a major source of toxic reactive oxygen species within the cell, this pathway is believed to be highly relevant to the pathogenesis of PD. Little is known about how parkin‐mediated ubiquitination is regulated during mitophagy or about the nature of the ubiquitin conjugates involved. We report here that USP8/UBPY, a deubiquitinating enzyme not previously implicated in mitochondrial quality control, is critical for parkin‐mediated mitophagy. USP8 preferentially removes non‐canonical K6‐linked ubiquitin chains from parkin, a process required for the efficient recruitment of parkin to depolarized mitochondria and for their subsequent elimination by mitophagy. This work uncovers a novel role for USP8‐mediated deubiquitination of K6‐linked ubiquitin conjugates from parkin in mitochondrial quality control.  相似文献

Calcium (Ca2+) is an almost universal second messenger that regulates important activities of all eukaryotic cells. It is of critical importance to neurons, which have developed extensive and intricate pathways to couple the Ca2+ signal to their biochemical machinery. In particular, Ca2+ participates in the transmission of the depolarizing signal and contributes to synaptic activity. During aging and in neurodegenerative disease processes, the ability of neurons to maintain an adequate energy level can be compromised, thus impacting on Ca2+ homeostasis. In Parkinson’s disease (PD), many signs of neurodegeneration result from compromised mitochondrial function attributable to specific effects of toxins on the mitochondrial respiratory chain and/or to genetic mutations. Despite these effects being present in almost all cell types, a distinguishing feature of PD is the extreme selectivity of cell loss, which is restricted to the dopaminergic neurons in the ventral portion of the substantia nigra pars compacta. Many hypotheses have been proposed to explain such selectivity, but only recently it has been convincingly shown that the innate autonomous activity of these neurons, which is sustained by their specific Cav1.3 L-type channel pore-forming subunit, is responsible for the generation of basal metabolic stress that, under physiological conditions, is compensated by mitochondrial buffering. However, when mitochondria function becomes even partially compromised (because of aging, exposure to environmental factors or genetic mutations), the metabolic stress overwhelms the protective mechanisms, and the process of neurodegeneration is engaged. The characteristics of Ca2+ handling in neurons of the substantia nigra pars compacta and the possible involvement of PD-related proteins in the control of Ca2+ homeostasis will be discussed in this review.  相似文献

Multiple factors determine plant reproductive success and their influence may vary spatially. This study addresses several factors influencing female reproductive success in three populations of Ruellia nudiflora, specifically we: (i) determine if fruit set is pollen‐limited and if pollinator visitation rates are related to this condition; (ii) estimate fruit set via autonomous self‐pollination (AS) and relate it to the magnitude of herkogamy; and (iii) evaluate if fruit abortion is a post‐pollination mechanism that determines the magnitude of pollen limitation. At each site we marked 35 plants, grouped as: unmanipulated control (C) plants subjected to open pollination, plants manually cross‐pollinated (MP), and plants excluded from pollinators and only able to self‐pollinate autonomously (AS). Fruit set was greater for MP relative to C plants providing evidence for pollen limitation, while a tendency was observed for lower fruit abortion of MP relative to C plants suggesting that fruit set is influenced not only by pollen delivery per se, but also by subsequent abortion. In addition, although pollinator visits varied significantly among populations, the magnitude of pollen limitation did not, suggesting that pollinator activity was not relevant in determining pollen limitation. Finally, fruit set tended to decrease with the degree of herkogamy for AS plants, but this result was inconclusive. These findings have contributed to identify which factors influence reproductive success in populations of R. nudiflora, with potentially relevant implications for population genetic structure and mating system evolution of this species.  相似文献

The whitefly and thrips predator Typhlodromips swirskii (Athias-Henriot) (Acari: Phytoseiidae) can be reared on the factitious astigmatid mite Suidasia medanensis (Oudemans) (Acari: Suidasiidae). The predator–prey relationship allows the system to be incorporated into a breeding sachet which releases predators into a crop over several weeks ensuring predator presence on arrival of the target pests and increased predator numerical response on the crop through immigration from the breeding sachet. This study investigated whether the prey preference and functional and numerical response of T. swirskii to different development stages of S. medanensis can provide understanding of the predator–prey interactions sustaining such a breeding sachet. T. swirskii elicited a strong preference to egg stages of S. medanensis, exhibited a Type II functional response and increased oviposition rate with increasing prey density. The relevance of these attributes to a balanced breeding sachet is discussed.  相似文献

New agricultural pest species attacking introduced crops may evolve from pre‐existing local herbivores by ecological speciation, thereby becoming a species by becoming a pest. We compare the evolutionary pathways by which two maize pests (the Asian and the European corn borers, ACB and ECB) in the genus Ostrinia (Lepidoptera, Crambidae) probably diverged from an ancestral species close to the current Adzuki bean borer (ABB). We typed larval Ostrinia populations collected on maize and dicotyledons across China and eastern Siberia, at microsatellite and mitochondrial loci. We found only two clusters: one on maize (as expected) and a single one on dicotyledons despite differences in male mid‐tibia morphology, suggesting that all individuals from dicotyledons belonged to the ABB. We found evidence for migrants and hybrids on both host plant types. Hybrids suggest that field reproductive isolation is incomplete between ACB and ABB. Interestingly, a few individuals with an ‘ABB‐like’ microsatellite profile collected on dicotyledons had ‘ACB’ mtDNA rather than ‘ABB‐like’ mtDNA, whereas the reverse was never found on maize. This suggests asymmetrical gene flow directed from the ACB towards the ABB. Hybrids and backcrosses in all directions were obtained in no‐choice tests. In laboratory conditions, they survived as well as parental strain individuals. In Xinjiang, we found ACB and ECB in sympatry, but no hybrids. Altogether, our results suggest that reproductive isolation between ACB and ABB is incomplete and mostly prezygotic. This points to ecological speciation as a possible evolutionary scenario, as previously found for ECB and ABB in Europe.  相似文献

The trajectory of an animal''s growth in early development has been shown to have long-term effects on a range of life-history traits. Although it is known that individual differences in behaviour may also be related to certain life-history traits, the linkage between early growth or development and individual variation in behaviour has received little attention. We used brief temperature manipulations, independent of food availability, to stimulate compensatory growth in juvenile three-spined sticklebacks Gasterosteus aculeatus. Here, we examine how these manipulated growth trajectories affected the sexual responsiveness of the male fish at the time of sexual maturation, explore associations between reproductive behaviour and investment and lifespan and test whether the perceived time stress (until the onset of the breeding season) influenced such trade-offs. We found a negative impact of growth rate on sexual responsiveness: fish induced (by temperature manipulation) to grow slowest prior to the breeding season were consistently quickest to respond to the presence of a gravid female. This speed of sexual responsiveness was also positively correlated with the rate of development of sexual ornaments and time taken to build a nest. However, after controlling for effects of growth rate, those males that had the greatest sexual responsiveness to females had the shortest lifespan. Moreover, the time available to compensate in size before the onset of the breeding season (time stress) affected the magnitude of these effects. Our results demonstrate that developmental perturbations in early life can influence mating behaviour, with long-term effects on longevity.  相似文献
