Previous investigations have shown that ammonia oxidation is not inhibited by diesel fuel in a soil with a long history of contamination contrary to a non-contaminated soil. As a consequence, ammonia oxidation does not constitute a limited step in nitrification process (Appl. Environ. Microbiol. 65 (1999) 4008). Moreover, this type of soil also has had the opportunity to develop an abundant microbial population able to metabolise the diesel hydrocarbons. Whether the properties of soil with a long history of diesel fuel contamination may affect the activity of nitrite-oxidising bacteria was investigated. It was observed that re-exposure of soil to diesel fuel apparently stimulated the proliferation of nitrite-oxidising bacteria, as determined by most probable number (MPN) culture technique and MPN-polymerase chain reaction technique. The potential of nitrite-oxidising activity in soil treated with diesel fuel was about 4 times higher than in the control without addition. In the presence of diesel fuel and ammonium, the potential nitrite-oxidising activity was 40% higher than in presence of ammonium only. However, in the presence of hydrocarbon only, low proliferation of Nitrobacter was observed, probably because the heterotrophic bacteria were strongly limited by lack of nitrogen and did not produce sufficient organic metabolites that could be used by the Nitrobacter cells.  相似文献

We describe a novel fully automated high-throughput time-lapse microscopy system and evaluate its performance for precisely tracking the motility of several glioma and osteoblastic cell lines. Use of this system revealed cell motility behavior not discernable with conventional techniques by collecting data (1) from closely spaced time points (minutes), (2) over long periods (hours to days), (3) from multiple areas of interest, (4) in parallel under several different experimental conditions. Quantitation of true individual and average cell velocity and path length was obtained with high spatial and temporal resolution in “scratch” or “wound healing” assays. This revealed unique motility dynamics of drug-treated and adhesion molecule-transfected cells and, thus, this is a considerable improvement over current methods of measurement and analysis. Several fluorescent vital labeling methods commonly used for end-point analyses (GFP expression, DiO lipophilic dye, and Qtracker nanocrystals) were found to be useful for time-lapse studies under specific conditions that are described. To illustrate one application, fluorescently labeled tumor cells were seeded onto cell monolayers expressing ectopic adhesion molecules, and this resulted in consistently reduced tumor cell migration velocities. Multidrug-resistant Plasmodium falciparum parasites have emerged in Cambodia and neighboring countries in Southeast Asia, compromising the efficacy of first-line antimalarial combinations. Dihydroartemisinin + piperaquine (PPQ) treatment failure rates have risen to as high as 50% in some areas in this region. For PPQ, resistance is driven primarily by a series of mutant alleles of the P. falciparum chloroquine resistance transporter (PfCRT). PPQ resistance was reported in China three decades earlier, but the molecular driver remained unknown. Herein, we identify a PPQ-resistant pfcrt allele (China C) from Yunnan Province, China, whose genotypic lineage is distinct from the PPQ-resistant pfcrt alleles currently observed in Cambodia. Combining gene editing and competitive growth assays, we report that PfCRT China C confers moderate PPQ resistance while re-sensitizing parasites to chloroquine (CQ) and incurring a fitness cost that manifests as a reduced rate of parasite growth. PPQ transport assays using purified PfCRT isoforms, combined with molecular dynamics simulations, highlight differences in drug transport kinetics and in this transporter’s central cavity conformation between China C and the current Southeast Asian PPQ-resistant isoforms. We also report a novel computational model that incorporates empirically determined fitness landscapes at varying drug concentrations, combined with antimalarial susceptibility profiles, mutation rates, and drug pharmacokinetics. Our simulations with PPQ-resistant or -sensitive parasite lines predict that a three-day regimen of PPQ combined with CQ can effectively clear infections and prevent the evolution of PfCRT variants. This work suggests that including CQ in combination therapies could be effective in suppressing the evolution of PfCRT-mediated multidrug resistance in regions where PPQ has lost efficacy.  相似文献

Samples from large (100–200 tons) batches of palm kernel cake (PKC, n?=?20) and copra meal (CM, n?=?13) were collected at production facilities of four Indonesian feed mill manufacturers and analysed for aflatoxin B1 (AFB1) by ELISA. Recoveries using spiked samples ranged from 86 to 113 %, with relative standard deviations of <9 % (PKC) and <6 % (CM). All batches were positive for AFB1: in PKC, at levels of 5.8–93.1 μg/kg (mean 49 μg/kg), and in CM, at levels of 1.1–147 μg/kg (mean 38.1 μg/kg). AFB1 levels were, in most batches, below the maximum level (100 μg/kg) recommended by the National Standardisation Agency, Republic of Indonesia. However, about half of the batches exceeded both the European Union and USA regulations for AFB1 in animal feed. In conclusion, serious efforts are necessary to control production, storage and shipment of palm kernel cake and copra meal for feed purposes, and clearly not only for products intended for export but also to reduce AFB1 levels in domestic Indonesian feed.  相似文献

ABSTRACT.   Megapodes are unique in using only heat from the environment, rather than body heat, to incubate their eggs as well as the precocious independence of their chicks on hatching. Of 22 recognized species of megapodes, 9 are listed as threatened due to factors including habitat loss and fragmentation, and predation on eggs and chicks. Orange-footed Scrubfowl ( Megapodius reinwardt ) are conspicuous components of the Oriental/Austral avifauna that inhabit the monsoon forests of the Lesser Sunda chain of islands in Indonesia. We examined the abundance, patterns of distribution, physical characteristics, seasonal activity, and predation risk of incubation mounds of Orange-footed Scrubfowl on Komodo Island in eastern Indonesia. We surveyed 13 valleys on Komodo Island from April 2002 to January 2005 and located 113 tended and 107 untended incubation mounds. Densities of scrubfowl mounds in our study were similar to that reported by investigators during the 1970s, suggesting little change in the scrubfowl population since then. Most scrubfowl mounds were on sandy or loamy soils in open monsoon forest with little overhead shade, and placement of mounds in such areas may ensure adequate temperatures for egg incubation. Although some mounds were tended during all months, mound use peaked during the late wet season in March. During the dry season (April–November), only a few mounds were tended. Komodo dragons ( Varanus komodoensis ) and wild pigs ( Sus scrofa ) were the primary predators of scrubfowl eggs, with no indication of egg predation by humans. The valley with the largest number of untended mounds in our study also had the largest number of active Komodo dragon nests. This suggests an effect of Komodo dragons on scrubfowl numbers, but additional study is needed.  相似文献

Under section 401 and section 404 of the Clean Water Act, permission to degrade existing natural wetlands in the USA may be conditional on restoring or creating ‘replacement’ wetlands. Success of wetland mitigation efforts in adequately replacing lost wildlife habitats depends on our good understanding of key ecological attributes that affect the structure of wetland faunal communities. We examined the effects of the presence of predatory fish, shallow vegetated littoral zone, emergent vegetation cover, wetland age and size on amphibian diversity in 42 replacement wetlands located in the Ohio’s North Central Tillplain ecoregion. We recorded 13 species of pond-breeding amphibians, and the average local species richness (α-richness) was 4.2 ± 1.7 species per site (range 1–7). There is strong evidence for the positive association between amphibian species richness and presence of a shallow littoral zone, and the negative association with presence of predatory fish. There was no evidence for the association between species richness and age, size, amount of forest cover within 200 m, nor the amount of emergent vegetation cover at the study sites. It is estimated that local species richness in wetlands with shallows was 1.76 species higher on average than in wetlands without shallows (95% CI from 0.75 to 2.76). The presence of predatory fish was associated with an average reduction in species richness by an estimated 1.21 species (95% CI from 0.29 to 2.11). Replacement wetlands were placed in areas with little or no existing forest cover, and amphibian species associated with forested wetlands were either rare (eastern newt, spotted salamander) or not present at all (marbled salamander, wood frog). In addition, we surveyed all replacement wetlands constructed under section 401 in Ohio since 1990, and found that predatory fish were present in 52.4% of the sites and that shallows were absent from 42.7% of the sites. Our results indicate that current wetland replacement practices could have a negative effect on the amphibian diversity within our region.  相似文献

DNA transfer from transgenic plants to native intestinal bacteria and introduced Acinetobacter BD413 was assessed in the gut of the tobacco horn worm (Manduca sexta). The marker was kanamycin resistance gene (nptII), and tobacco carrying the nptII gene in the chloroplasts served as the donor. We detected neither whole gene transfer to native bacteria, nor transfer of fragments of nptII to Acinetobacter, using a marker exchange assay. This negative result was attributed to a heat-labile activity that degraded DNA in the feces, probably DNAase. Nevertheless, a few intact leaf cells survived transit through the gut, and DNA extracted from feces did transform Acinetobacter, albeit at lower frequencies than DNA extracted from leaves.  相似文献

The aim of this study was to investigate neonatal Fc receptor (FcRn) concentration developmental pharmacology in adult and pediatric subjects using minimal physiologically-based pharmacokinetic (mPBPK) modelling. Three types of pharmacokinetic (PK) data for three agents (endogenous/exogenous native IgG, bevacizumab and palivizumab) were used. The adult group contained six subjects with weights from 50 to 100 kg. For pediatric subjects, seven age groups were assumed, with five subjects each having the weight of 95%, 75%, 50%, 25% and 5% percentile of the population. A first evidence-based rating system to evaluate the quality of the source data used to derive pediatric-specific mPBPK model parameter was proposed. A stepwise approach was used to examine the best combination of age/weight effect on the parameters of the mPBPK model in adult and pediatric subjects. IgG synthesis rate (Ksyn), extravasation rate (ER) and FcRn were fitted simultaneously to the PK of bevacizumab and native-IgG in both adult and pediatric. All fitting showed good fits based on the graphs and the coefficient of variation of the fitted parameters (< 50%). Estimated weight-normalized Ksyn increased while weight-normalized FcRn and ER decreased with increasing age. The age and weight effect on FcRn were successfully estimated from the data. The final mPBPK model developed with native IgG and bevacizumab was able to predict the PK of palivizumab in pediatric subjects. Implementation of the mPBPK model enables us to analyze the relationships of age, weight, FcRn, ER and Ksyn in both adult and pediatric subject. This information may benefit the understanding of complex interaction between the FcRn developmental pharmacology and PK parameters, and improve the prediction of the antibody disposition in pediatric subjects.  相似文献

The purpose of the study was to find out differences between treatments of diabetes type 2 after secondary oral antidiabetic drug failure. Three different methods of treatment were compared: lispro insulin in combination with metformin, glimepiride and metformin combination or two daily doses of biphasic insulin 30/70 together with bed-time NPH insulin. The study included 87 patients with diabetes mellitus type 2 randomly distributed into 3 different treatment groups. Fasting and postprandial glucose were analyzed by enzymatic colorimetric method and HbA1c was measured by ion exchange chromatography. HbA1c significantly decreased in all three study groups. The decrease was mostly expressed among patients treated with lispro and metformin. When focused on postprandial glucose control, antihyperglycemic metformin and insulin lispro therapy has greater impact on the overall metabolic control (decrease in level of HbA1c) in comparison with the above mentioned more traditional approaches.  相似文献
