把从榛木(Corylusavellana L.)花粉中分离得到的高尔基囊泡与经高度纯化并聚合好的牛脑微管进行体外组合,然后于1.5 m ol/L的蔗糖层上进行超离心,对其沉淀物进行SDS-聚丙烯酰胺凝胶电泳和电镜负染。结果表明,花粉高尔基囊泡可以结合到牛脑微管上,证明植物花粉的高尔基囊泡与动物细胞的某些细胞器一样,也与细胞骨架的主要组成之一——微管具有结构上的紧密联系。花粉高尔基囊泡与牛脑微管的体外结合能力,受10 m m ol/LATP和0.5 m ol/LKCl的影响,但不受5 m m ol/L AMP-PNP的影响,说明两者结合可能是通过高尔基囊泡表面与ATP有关的某种外周膜蛋白来完成的。  相似文献

The Ac/Ds transposon system of maize shows low activity in Arabidopsis. However, fusion of the CaMV 35S promoter to the transposase gene (35S::TPase) increases the abundance of the single Ac mRNA encoded by Ac and increases the frequency of Ds excision. In the experiments reported here it is examined whether this high excision frequency is associated with efficient re-insertion of the transposon. This was measured by using a Ds that carried a hygromycin resistance gene (HPT) and was inserted within a streptomycin resistance gene (SPT). Excision of Ds therefore gives rise to streptomycin resistance, while hygromycin resistance is associated with the presence of a transposed Ds or with retention of the element at its original location. Self-fertilisation of most individuals heterozygous for Ds and 35S::TPase produced many streptomycin-resistant (strepr) progeny, but in many of these families a small proportion of strepr seedlings were also resistant to hygromycin (hygr). Nevertheless, 70% of families tested did give rise to at least one strepr, hygr seedling, and over 90% of these individuals carried a transposed Ds. In contrast, the Ac promoter fusion to the transposase gene (Ac::TPase) produced fewer streprhygr progeny, and only 53% of these carried a transposed Ds. However, a higher proportion of the strepr seedlings were also hygr than after activation by 35S::TPase. We also examined the genotype of strepr, hygr seedlings and demonstrated that after activation by 35S::TPase many of these were homozygous for the transposed Ds, while this did not occur after activation by Ac::TPase. From these and other data we conclude that excisions driven by 35S::TPase usually occur prior to floral development, and that although a low proportion of strepr progeny plants inherit a transposed Ds, those that do can be efficiently selected with an antibiotic resistance gene contained within the element. Our data have important implications for transposon tagging strategies in transgenic plants and these are discussed.  相似文献

The development of nitrogen-fixing nodules is induced on the roots of legume host plants by Rhizobium bacteria. We employed a novel strategy to probe the underlying mechanism of nodule morphogenesis in alfalfa roots using pTZS, a broad host range plasmid carrying a constitutive trans-zeatin secretion (tzs) gene from Agrobacterium tumefaciens T37. This plasmid suppressed the Nod- phenotype of Rhizobium nodulation mutants such that mutants harboring pTZS stimulated the formation of nodulelike structures. Alfalfa roots formed more or fewer of these nodules according to both the nitrogen content of the environment and the position along the root at which the pTZS+ bacteria were applied, which parallels the physiological and developmental regulation of true Rhizobium nodule formation. This plasmid also conferred on Escherichia coli cells the ability to induce root cortical cell mitoses. Both the pattern of induced cell divisions and the spatially restricted expression of an alfalfa nodule-specific marker gene (MsENOD2) in pTZS-induced nodules support the conclusion that localized cytokinin production produces a phenocopy of nodule morphogenesis.  相似文献

The evolutionary position of the yew family, Taxaceae, has been very controversial. Some plant taxonomists strongly advocate excluding Taxaceae from the conifer order and raising its taxonomic status to a new order or even class because of its absence of seed cones, contrary to the case in the majority of conifers. However, other authors believe that the Taxaceae are not fundamentally different from the rest of the conifers except in that they possess the most reduced solitary-ovule cones. To resolve the controversy, we have sequenced the 18S rRNA genes from representative gymnosperms: Taxus mairei (Taxaceae), Podocarpus nakaii (Podocarpaceae), Pinus luchuensis (Pinaceae), and Ginkgo biloba (Ginkgoales). Our phylogenetic analysis of the new sequence data with the published 18S rRNA sequence of Zamia pumila (a cycad) as an outgroup strongly indicates that Taxus, Pinus, and Podocarpus form a monophyletic group with the exclusion of Ginkgo and that Taxus is more closely related to Pinus than to Podocarpus. Therefore, Taxaceae should be classified as a family of Coniferales. Our finding that Taxaceae, Pinaceae, and Podocarpaceae form a clade contradicts both the view that the uniovulate seed of Taxaceae is a primitive character and the view that the Taxaceae are descendants of the Podocarpaceae. Rather, the uniovulate seed of Taxaceae and that of some species of Podocarpus appear to have different origins, probably all reduced from multiovulate cones. Correspondence to: W.-H. Li  相似文献

The early steps of symbiotic nodule formation by Rhizobium on plants require coordinate expression of several nod gene operons, which is accomplished by the activating protein NodD. Three different NodD proteins are encoded by Sym plasmid genes in Rhizobium meliloti, the alfalfa symbiont. NodD1 and NodD2 activate nod operons when Rhizobium is exposed to host plant inducers. The third, NodD3, is an inducer-independent activator of nod operons. We previously observed that nodD3 carried on a multicopy plasmid required another closely linked gene, syrM, for constitutive nod operon expression. Here, we show that syrM activates expression of the nodD3 gene, and that nodD3 activates expression of syrM. The two genes constitute a self-amplifying positive regulatory circuit in both cultured Rhizobium and cells within the symbiotic nodule. We find little effect of plant inducers on the circuit or on expression of nodD3 carried on pSyma. This regulatory circuit may be important for regulation of nod genes within the developing nodule.  相似文献

The maximum quantum yields (a,c) for CO2 uptake in low-oxygen atmospheres were determined for 11 species of C3 vascular plants of diverse taxa, habitat and life form using an Ulbricht-sphere leaf chamber. Comparisons were also made between tissues of varied age within species. The species examined were Psilotum nudum (L.) P. Beauv., Davallia bullata Wall. ex Hook., Cycas revoluta Thunb., Araucaria heterophylla (Salisb.) Franco, Picea abies (L.) Karst., Nerium oleander L., Ruellia humilis Nutt., Pilea microphylla (L.) Karst., Beaucarnea stricta Lem., Oplismenus hirtellus (L.) P. Beauv. and Poa annua L. Quantum yields were calculated from the initial slopes of the response of CO2 uptake to the quantity of photons absorbed in conditions of diffuse lighting. Regression analysis of variance of the initial slopes of the response of CO2 uptake to photon absorption failed to show any statistically significant differences between age classes within species or between the mature photosynthetic organs of different species. The constancy of a,c was apparent despite marked variation in the light-saturated rates of CO2 uptake within and between species. The mean a,c was 0.093±0.003 for 11 species. By contrast, surface absorptance varied markedly between species from 0.90 to 0.60, producing proportional variation in the quantum yield calculated on an incidentlight basis. The ratio of variable to maximum fluorescence emission at 695 nm for the same tissues also failed to show any statistically significant variation between species, with a mean of 0.838±0.008. OBJECTIVES: To estimate the magnitude of antituberculous drug resistance and identify the risk factors for its development in tuberculosis patients in Manitoba over a 10-year period. As well, to examine the clinical course of the patients whose initial or subsequent isolates of Mycobacterium tuberculosis were resistant to one or more drugs. DESIGN: Comparison of drug-resistant and non-drug-resistant cases of tuberculosis. SETTING: Manitoba. PATIENTS: All people with tuberculosis reported to the Central Tuberculosis Registry of Manitoba between Jan. 1, 1980, and Dec. 31, 1989. MAIN OUTCOME MEASURES: Of 1478 cases of active tuberculosis 1086 were culture positive, and drug susceptibility testing was performed in these cases. The clinical course, including outcome of treatment, of all drug-resistant cases was described. RESULTS: Of 1086 culture-positive cases of tuberculosis 77 (7.1%) were drug resistant. Odds ratios suggested that the risk of drug resistance was significantly higher among the immigrants than among the other Canadians. Compared with the other Canadians the risk of drug resistance was 9.9 times greater among the immigrants in whom tuberculosis developed within the first year after arrival in Canada and 5.4 times greater among the immigrants in whom it developed 2 to 5 years after arrival in Canada. Of the 71 patients with drug-resistant disease whose type of resistance was known 62% had never taken antituberculous drugs before and 38% had. Most (91%) of the 77 cases of drug-resistant disease were resistant to first-line drugs, especially isoniazid and streptomycin. Thirty-two (42%) of the 77 cases were resistant to two or more first-line drugs. Of patients with drug-resistant disease a subgroup of 10 had disease that became resistant to several drugs over the 10-year period. The outcome of treatment in these individuals was poor, and they presented a particular public health problem. CONCLUSION: Resistance to one or more first-line antituberculous drugs continues to complicate the treatment of tuberculosis and may facilitate the spread of the disease.  相似文献

Compared with the standard method of manual fertilizer broadcasting (MFB), mechanized hill-drilling direct-seeding with deep application of slow-release nitrogen fertilizer (MHDDF) is an efficient method to integrate both fertilization and seeding. However, there are few studies that combine the use of slow-release fertilizer with MHDDF. We sought to explore the combined effect of MHDDF with slow-release fertilizer on rice yield and nitrogen, phosphorus, and potassium utilization, compared to MFB. We compared three different MHDDF methods (D30: 450 kg ha?1, D40: 600 kg ha?1, D50: 750 kg ha?1), with one MFB method (B50: 750 kg ha?1), and one control (CK: 0 kg ha?1). We found that the yield of all MHDDF method was higher than that of both the MFB method. Yield was the highest in the D50 treatment and was 14.14–46.03% higher than that in B50 treatment. Biomass accumulation, nutrient accumulation, and nutrient use efficiency were similarly higher in MHDDF method than both MFB and CK. Compared to B50, the D50 treatment increased nitrogen recovery efficiency by 170.53–231.50%, phosphorus recovery efficiency by 480.00–724.25%, and potassium recovery efficiency by 201.55–169.59%. Overall, we found that combining MHDDF with slow-release fertilizer was an effective method to increase rice yield and nutrient use efficiency compared with MFB.

The morphological effects of population density in locusts in the production of phases is now well established and in recent years somewhat parallel effects have been observed in the larvae of Lepidoptera. Unlike locusts Lepidoptera pass through a quiescent pupal period but nevertheless the adult may be affected. In both P. gamma and P. brassicae adults from crowded larval cultures were smaller in both size and weight although the effect was counteracted to some extent by approaching pupal diapause in P. brassicae. The effect was similar in many cases to that produced by an increase in temperature except that it disturbed the relationships between the lengths of the wing and hind femur with the live weight. The ratio of these lengths (equivalent to E/F ratio in locusts) was significantly increased but was not a satisfactory index of larval population density. No effects were observed in the wing pattern and colouration in either species.
Zusammenfassung Die Einwirkung der larvalen Populationsdichte auf die Morphologie der jungen Falter wurde bei Plusia gamma und Pieris brassicae durch Vergleich frischgeschlüpfter Falter aus Einzel- und aus Massenzuchten bestimmt.Bei P. gamma wogen Puppen aus Massenkulturen etwas weniger und verloren während des Puppenstadiums mehr an Gewicht. Die entstehenden Falter waren gewichtsmäßig deutlich leichter und hatten etwas kürzere Flügel. Größe und Gewicht der Männchen waren größer als die der Weibchen, und da die Massenzucht die meßbaren Größen bei den Weibchen stärker beeinflußt, verstärkte sie den Unterschied zwischen den Geschlechtern. Jahreszeitliche Unterschiede wurden beobachtet, wobei Gewicht und Größen in der warmen Jahreszeit geringer waren, doch blieb der jahreszeitliche Einfluß geringer als der durch Übervölkerung hervorgerufene.Bei P. brassicae hatte die Massenzucht eine ähnliche Wirkung auf die Puppen- und Erwachsenengewichte und auch auf die Maße, außer daß der Flügel kürzer und schmaler war. Die Unterschiede zwischen den Geschlechtern wurden durch die Massenaufzucht nicht verschärft. Lebendgewicht und Femurlänge waren beim Männchen größer, währen das Weibchen größere Flügel hatte. Bei Herannahen der Diapause wurde die Wirkung der Massenkultur auf Puppen- und Imaginalgewicht kompensiert, aber die Wirkungen auf die Erwachsenenmaße blieben noch erhalten, wenn auch in geringerem Ausmaß als sie bei nichtdiapausierenden Generationen zu beobachten waren. In der wärmeren Jahreszeit wurden Variationen erzielt, die in etwa den durch Übervölkerung bewirkten ähnlich waren, außer daß die Flügel abweichende Form aufwiesen und daß Übervölkerung bei den Männchen die Wirkung stark reduzierte.In Einzelkulturen beider Arten wurden positive, aber nicht stark signifikante Korrelationen in der Beziehung von Costa- und Hinterschenkellänge mit dem Lebendgewicht gefunden. Diese Beziehungen wurden durch Massenkultur gestört.In beiden Arten wurde das Costa-Schenkel-Verhältnis durch Massenkultur signifikant vergrößert (cf. E/F-Rate bei Heuschrecken), ergab aber keinen befriedigenden Index für die larvale Populationsdichte.In der Flügelzeichnung und Färbung beider Arten wurden durch larvale Massenkultur keine Wirkungen beobachtet.

Triple-helix formation of the peptide (Pro-Hyp-Gly)10 was monitored by nmr and CD spectroscopy. The two-dimensional nmr spectra indicated that the Gly C alpha H and Pro C delta H proton resonances shift upfield in going from the nonhelical to helical form, while hydroxy-proline resonances are unchanged. The integrated areas of the helical and nonhelical resonances could be monitored in the one-dimensional nmr spectrum, and indicate that in the (Pro-Hyp-Gly)10 about 90% of the residues are in a defined triple-helical conformation. The introduction of a glycine to alanine substitution or the deletion of a single hydroxyproline residue in the stable triple-helical peptide (Pro-Hyp-Gly)10 still allows trimers to be formed, but the trimers show a substantial loss of triple helix and decreased thermal stability compared with (Pro-Hyp-Gly)10. Two computer models were generated for the Gly----Ala peptide, one with the Ala side chains packed inside the helix and the other with the region containing the alanines forming a beta-bend that loops out from the helix.
