The hydrophobin SC3p of Schizophyllum commune self-assembles into a 10-nm-thick amphipathic membrane at hydrophilic-hydrophobic interfaces. X-ray photoelectron spectroscopy of the hydrophobic membrane side of SC3p, assembled in vitro, showed an atomic composition similar to the calculated composition of SC3p when glycosylation was taken into account. The atomic composition measured at the hydrophilic membrane side deviated from that at the hydrophobic side and indicated the presence of a lower number of peptide bonds. High levels of S and N were detected only on mycelia carrying hydrophobic aerial hyphae, as expected with assembled SC3p present at the surface of these hyphae.  相似文献

Stimulatory GTP-binding Protein (Gs) and adenylate cyclase prepared from bovine brain cortices were co-reconstituted into asolectin vesicles with or without 1000-fold transmembrane Ca2+ gradient. The results showed that both basal activity and Gs-stimulated activity of adenylate cyclase were highest in proteoliposomes with a transmembrane Ca2+ gradient similar to physiological condition (1 M Ca2+ outside and 1 mM Ca2+ inside) and lowest when the transmembrane Ca2+ gradient was in the inverse direction. Such a difference could be diminished following dissipation of the transmembrane Ca2+ gradient by A23187. Comparable conformational changes of Gs in proteoliposomes were also observed when Gs was labeled with the fluorescence probe, acrylodan. These results may indicate that a proper transmembrane Ca2+ gradient is essential not only for higher adenylate cyclase activity but also for its stimulation by Gs.  相似文献

The effect of ABA on the membrane potential of barley (Hordeumvulgare cv. Himalaya) aleurone protoplasts was studied by measuringthe distribution of the lipophilic cation tetraphenylphosphonium(TPP+). The resting membrane potential (Em) according to ourmeasurements with TPP+ is about –53 mV and is in agreementwith membrane potential values as measured with intracellularmicroelectrodes (about –55 mV). The TPP+-measurementscould demonstrate a clear dependence of the resting Em on theexternal pH (pHe). Stimulation of the protoplasts with ABA induced a transienthyperpolarization of the membrane to –62 mV as measuredwith TPP+. The hyperpolarization was ABA-concentration dependent. Inhibition of the H+-ATPases with the specific proton pump inhibitorsdiethylstilbestrol (DES) or Micanozole effectively preventedhyperpolarization. This indicates that the hyperpolarizationis consistent with the activation of plasma membrane H+-ATPases.The K+-inward rectifier inhibitor BaCl2 was able to prolongthe hyperpolarization. This result suggests that the hyperpolarizationcauses the opening of K+-channels. The ABA-induced proton-pump activation may be involved in ABA-inducedgene-expression, as DES was able to inhibit this gene expression.BaCl2 did only show a slight inhibitory effect on ABA-inducedgene-expression. Genes for biosynthesis of a Streptomyces sp. FR-008 heptaene macrolide antibiotic with antifungal and mosquito larvicidal activity were cloned in Escherichia coli using heterologous DNA probes. The cloned genes were implicated in heptaene biosynthiesis by gene replacement. The FR-008 antibiotic contains a 38-membered, poiyketide-derived macrolide ring. Southern hybridization using probes encoding domains of the type i modular erythromycin polyketide synthase (PKS) showed that the Streptomyces sp. FR-008 PKS gene cluster contains repeated sequences spanning c. 105 kb of contiguous DNA; assuming c. 5 kb for each PKS module, this is in striking agreement with the expectation for the 21-step condensation process required for synthesis of the FR-008 carbon chain. The methods developed for transformation and gene replacement in Streptomyces sp. FR-008 make it possible to genetically manipulate polyene macrolide production, and may later lead to the biosynthesis of novel polyene macrolides.  相似文献

A 29-year-old male with Angelman syndrome and an unbalanced reciprocal translocation, 45,XY,-8,-15,+der(8),t(8;15)(p23.3;q11)pat, was evaluated with DNA studies. These showed the underlying mechanism to be paternal uniparental disomy. This is the second case reported of Angelman syndrome that has resulted from a familial unbalanced reciprocal translocation.  相似文献

橐吾属Ligularia Cass.是菊科千里光族款冬亚族的一个大属。在款冬亚族中本属与大吾风草属 Farfugium Lindl.亲缘关系最近,但进化程度较高。本属包括6组,11系129种。所有种类均分布在 亚洲,仅2种扩散至欧洲。在东亚地区有119种,占该属总种数的96%。高度集中在横断山区的有4组、 6系67种,其中61种为特有种,占该属总组数的66%,总系数的54.5%,总种数的52%。这个事实 表明了横断山区是该属的多度中心和多样化中心。通过性状分析,伞房组伞房系Sect.Corymbosae, Ser.Calthifoliae叶肾形,具掌状叶脉,头状花序大而少,排列呈伞房状,总苞半球形,被认为是该属的 原始类群。原始种齿叶橐吾L. dentata和鹿蹄橐吾L.hodgsonii的分布区从我国四川东部经过湖北、湖 南、安徽、福建等省至日本。这个分布格局与近缘属大吴风草属Farfugium一致。 根据共同起源原理,这两个属的祖先极有可能就发生在这一地区。因此我们推测东亚地区从中国四 川东部至日本这一地区是本属的发源地,然而根据地质历史和现代分布,作者认为中国中部(包括四川 东部)是本属的初始起源地。该属起源后,基本上沿亚洲南缘的山地扩散,少数种类向东北至亚洲东北部。本属起源时间至少不晚于中白垩纪。  相似文献

研究了大蒜(Allium sativum L.)中期染色体的超微结构和RNP物质。常规染色表明,大部分染色体内部有低电子密度区,有的染色体中低电子密度区域较大而似孔洞。银染结果也证明了有大小不等的孔洞存在。Bernhard 染色显示,在染色体周边和染色体内部都有RNP分布。用NaOH 处理证明了Bernhard 染色法所显示的深染区确实含有RNA。RNP量的多少与EDTA 的分化时间呈负相关  相似文献
