Densely populated, intensively cropped highland areas in the tropics and subtropics are prone to erosion and declining soil fertility, making agriculture unsustainable. Conservation agriculture in its version of permanent raised bed planting with crop residue retention has been proposed as an alternative wheat production system for this agro-ecological zone. A five years field experiment comparing permanent and tilled raised beds with different residue management under rainfed conditions was started at El Batán (Mexico) (2,240 m asl; 19.31°N, 98.50°W; Cumulic Phaeozem) in 1999. The objective of this study was to determine the soil quality status after five years of different management practices. The K concentration was 1.65 times and 1.43 times larger in the 0–5 cm and 5–20 cm profiles, respectively, for permanent raised beds compared to conventionally tilled raised beds. The Na concentration showed the opposite trend. Sodicity was highest for conventionally tilled raised beds and for permanent raised beds it increased with decreasing amounts of residue retained on the surface. Permanent raised beds with full residue retention increased soil organic matter content 1.4 times in the 0–5 cm layer compared to conventionally tilled raised beds with straw incorporated and it increased significantly with increasing amounts of residue retained on the soil surface for permanent raised beds. Soil from permanent raised beds with full residue retention had significantly higher mean weight diameter for wet and dry sieving compared to conventionally tilled raised beds. Permanent raised beds with full residue retention had significantly higher aggregate stability compared to those with residue removal. A lower aggregation resulted in a reduction of infiltration. Principal component analysis (PCA) was performed using these soil physicochemical variables that were significantly influenced by tillage or residue management. The PC1 and PC2 separated the conventionally tilled raised beds from the permanent raised beds and PC3 separated permanent raised beds with at least partial residue retention from permanent raised beds with no residue retention. These clear separations suggest that tillage and residue management have an effect on soil processes. The research indicates that permanent raised bed planting increases the soil quality and can be a sustainable production alternative for the (sub)tropical highlands. Extensive tillage with its associated high costs can be reduced by the use of permanent raised beds while at least partial surface residue retention is needed to insure production sustainability.  相似文献

Induction of assimilatory NO 3 reduction through the application of an easily decomposable substrate in alkaline–saline soils of the former lake Texcoco (Mexico) resulted in a fast immobilization of NO 3 in excess of N required for metabolic activity and the release of large concentrations of NO 2 and smaller amounts of NH 4 + . We postulated that this was regulated by the amounts of NO 3 and glucose added, and affected by the specific characteristics of soil from the former lake Texcoco. This was investigated by spiking soils of different electrolytic conductivity (EC) 56.0 dS m−1 (soil A of Texcoco) and 11.6 dS m−1 (soil B of Texcoco) with different concentrations of NO 3 and glucose while dynamics of CO2, NH 4 + , NO 2 and NO 3 were monitored in an aerobic incubation for 7 days. For comparison reasons (control) an agricultural soil with low EC (0.3 dS m−1) was included as well. In the agricultural soil, 67% of the added glucose mineralized within 7 days, but only 15% in soil A of Texcoco and 20% in soil B of Texcoco. The application of NO 3 to the agricultural soil added with glucose increased cumulative production of CO2 1.2 times, 1.5 times in soil A of Texcoco and 1.8 times in soil B of Texcoco. Concentration of NO 2 increased to > 100 mg NO 2 -N kg−1 when 1000 mg glucose-C kg−1 and 500 mg NO 3 -N kg−1 were added to soil A and B of Texcoco, but remained < 3 mg NO 2 -N kg−1 in the agricultural soil. The ratio between the cumulative production of CO2 and the decrease in concentration of NO 3 was approximately one in soil A and B of Texcoco, but 10 in the agricultural soil after 3 days. It was found that micro-organisms in the alkaline–saline soil of the former lake Texcoco were capable of immobilizing large quantities of NO 3 when an easy decomposable substrate was available in excess of what might be required for metabolic activity while producing large concentrations of NO 2 , but these phenomena were absent in an agricultural soil. In soil of Texcoco, concentrations of NO 2 and NH 4 + increased with increased salinity and availability of NO 3 . This ability to remove large quantities of NO 3 under these conditions and then utilize it at a later time might benefit micro-organisms of the N limited alkaline–saline soils of Texcoco.  相似文献

Conservation tillage in its version of permanent bed planting under zero-tillage with crop residue retention has been proposed as an alternative wheat production system for northwest Mexico. However, little is known about the dynamics of C and N in soils under wheat/maize on permanent beds (PB) where straw was burned, removed, partly removed or retained, as opposed to conventionally tilled beds (CTB) where straw was incorporated. We investigated the dynamics of soil C and N and normalized difference vegetative index (NDVI) crop values in zero-tilled PB and CTB after 26 successive maize and wheat crops. Organic C and total N were respectively, 1.15 and 1.17 times greater in PB with straw partly removed and with straw retained on the surface, than in CTB with straw incorporated. Organic C and total N were 1.10 times greater in soils with 300 kg N ha−1 added than in unfertilized soil. Cumulative production of CO2 was lower under CTB with straw incorporated than under PB treatments, and CO2 production increased with increments in inorganic fertilizer. The N-mineralization rate was 1.18 times greater than in unamended soils when 150 kg inorganic N ha−1 was applied, and 1.48 times greater when 300 kg inorganic N ha−1 was added. The N-mineralization rate was significantly (1.66 times) greater in PB where the straw was burned or retained on the surface than in CTB where the straw was incorporated, but significantly (1.25 times) lower than in PB with straw partly removed. The NDVI values reached a maximum 56 days after planting and decreased thereafter. The NDVI for unfertilized soil were similar for CTB with straw incorporated, PB with straw partly removed, and PB with straw retained on the surface, but significantly lower for PB with straw burned and PB with straw removed. In soils to which 150 or 300 kg N ha−1 was added, NDVI was significantly lower for PB with straw burned than for other treatments. Among other things, this suggests the utility of rotating maize or wheat with crops whose residues have lower C–N ratios, thus avoiding immobilization of large amounts of N for extended periods. PB with residue burning, however, is an unsustainable practice leading to low crop performance and soil and environmental degradation.  相似文献

The effect of cell density on xylanolytic activity and productivity of Cellulomonas flavigena was evaluated under two different culturing conditions: fed-batch culture with discontinuous feed of sugar cane bagasse (SCB; condition 1) and glycerol fed-batch culture followed by addition of SBC as xylanases inducer (condition 2). The enzymatic profile of xylanases was similar in both systems, regardless of the initial cell density at time of induction. However, the xylanolytic activity changed with initial cell density at the time of induction (condition 2). The maximum volumetric xylanase activity increased with increased initial cell density from 4 to 34 g l−1 but decreased above this value. The largest total volumetric xylanase productivity under condition 2 (1.3 IU ml−1 h−1) was significantly greater compared to condition 1 (maximum 0.6 IU ml−1 h−1). Consequently, induction of xylanase activity by SCB after growing of C. flavigena on glycerol at intermediate cell density can be a feasible alternative to improve activity and productivity of xylanolytic enzymes.  相似文献

The dynamics of inorganic N are important in soil, and this applies particularly to the saline–alkaline soils of the former lake Texcoco in Mexico with high pH and salinity where a forestation program was started in the 1970s. In soils of lake Texcoco, in Mexico, more than 50% of applied N could not be accounted for one day after application of 200 mg kg–1 soil along with glucose amendment. It was not clear whether this was due to abiotic or biotic processes, the form of inorganic N applied or the result of applying an easily decomposable substrate. We investigated this by adding glucose and 200 mg kg–1 soil as (NH4)2SO4-N or KNO3-N to sterilized and unsterilized soil. The changes in inorganic and ninhydrin N, microbial biomass C and production of CO2 were then monitored. Between the time of applying N and extraction with 0.5 M K2SO4, i.e., after ca 2 h, approximately 110 mg NH4 +-N kg–1 dry soil could not be accounted for in the unsterilized and sterilized soil and that remained so for the entire incubation in the sterilized soil. After 1 day this increased to 140 mg NH4 +-N kg–1 dry soil in the unsterilized control and 170 mg NH4 +-N kg–1 dry soil in C amended soil. Volatilization of NH3 accounted for 56 mg NH4 +-N kg–1 so the rest appeared to be adsorbed on the soil matrix. The NH3 volatilization and NH4 + fixed in the soil matrix remained constant over time and no oxidation to NO2 or NO3 had occurred, so unaccounted N in unsterilized soil was probably incorporated into the microbial biomass in excess of what was required for metabolic activity. The unaccounted N was ca 70 mg NO3 –N in nitrate amended soil after 3 days and 138 NO3 –N when glucose was additionally added. Losses through abiotic processes were absent as inferred from changes in sterilized soil and the aerobic incubation inhibited possible losses through denitrification. It was inferred that NO3 that could not be accounted for was taken up by micro-organisms in excess of what was required for metabolic activity.  相似文献

The alkaline α-amylase produced by Texcoconibacillus texcoconensis 13CCT strain was identified by random mutagenesis and confirmed by directed mutagenesis. A transposon mutagenesis approach was taken to identify the gene responsible for the degradation of starch in T. texcoconensis 13CCT strain. The deduced amino acids of the amy gene had a 99 % similarity with those of Bacillus selenitireducens MLS10 and 97 % with those of Paenibacillus curdlanolyticus YK9. The enzyme showed a maximum activity of 131.1 U/mL at 37 °C and pH 9.5 to 10.5. In situ activity of the enzyme determined by polyacrylamide gel electrophoresis showed only one band with amylolytic activity. This is the first report of a bacterium isolated from the extreme alkaline–saline soil of the former Lake Texcoco (Mexico) with amylolytic activity in alkaline conditions while its potential as a source of amylases for the industry is discussed.  相似文献

Early retinal vascular changes in the development of diabetic retinopathy (DR) include capillary basal lamina (BL) thickening, pericyte loss and the development of acellular capillaries. Expression of the CCN (connective tissue growth factor/cysteine-rich 61/nephroblastoma overexpressed) family member CCN2 or connective tissue growth factor (CTGF), a potent inducer of the expression of BL components, is upregulated early in diabetes. Diabetic mice lacking one functional CTGF allele (CTGF+/−) do not show this BL thickening. As early events in DR may be interrelated, we hypothesized that CTGF plays a role in the pathological changes of retinal capillaries other than BL thickening. We studied the effects of long-term (6-8 months) streptozotocin-induced diabetes on retinal capillary BL thickness, numbers of pericytes and the development of acellular capillaries in wild type and CTGF+/− mice. Our results show that an absence of BL thickening of retinal capillaries in long-term diabetic CTGF+/− mice is associated with reduced pericyte dropout and reduced formation of acellular capillaries. We conclude that CTGF is involved in structural retinal vascular changes in diabetic rodents. Inhibition of CTGF in the eye may therefore be protective against the development of DR.  相似文献

The diversity of the dissimilatory and respiratory nitrate-reducing communities was studied in two soils of the former lake Texcoco (Mexico). Genes encoding the membrane-bound nitrate reductase (narG) and the periplasmic nitrate reductase (napA) were used as functional markers. To investigate bacterial communities containing napA and narG in saline alkaline soils of the former lake Texcoco, libraries of the two sites were constructed (soil T3 with pH 11 and electrolytic conductivity in saturated extract (ECSE) 160 dS m−1 and soil T1 with pH 8.5 and ECSE 0.8 dS m−1). Phylogenetic analysis of napA sequences separated the clone families into two main groups: dependent or independent of NapB. Most of napA sequences from site T1 were grouped in the NapB-dependent clade, meanwhile most of the napA sequences from the extreme soil T3 were affiliated to the NapB-independent group. For both sites, partial narG sequences were associated with representatives of the Proteobacteria, Firmicutes and Actinobacteria phyla, but the proportions of the clones were different. Our results support the concept of a specific and complex nitrate-reducing community for each soil of the former lake Texcoco.  相似文献

This paper reports on the optimum concentrations of naphthalene acetic acid (NAA) and 6-benzyladenine (BA) to stimulate callus growth and NAA; kinetin and silver nitrate (AgNO3) for callus redifferentiation in Dianthus caryophyllus L. Meristems were excised and placed in MS medium with 30 g l−1 sucrose and 9.0 μM 2,4-d. Callus clusters were transferred to MS medium containing NAA (0, 1.7, 3.3, and 5.0 μM) and BA (0, 1.7, 3.3, and 5.0 μM) for proliferation and to MS medium with 30 g l−1 sucrose, 2.5 g l−1 phytagel, kinetin (0, 33, and 66 μM); NAA (0, 7.95, and 15.9 μM) and AgNO3 (0, 23.54 and 47.08 μM) for shoot and root induction. Treatments were applied according to a Box–Behnken design. After callus growth and redifferentiation, plants were incubated in the greenhouse at 18 ± 2°C for 4 wk and at 20–26°C for 4 wk. Finally, plants were changed to near-commercial greenhouse conditions with different day (30–35°C) and night (16–24°C) temperatures. Results showed better callus growth at higher NAA concentrations. A maximum callus weight was found with 5.0 μM NAA but without BA. A maximum of 78% calluses with shoots was obtained with 15.9 μM NAA, 47.08 μM AgNO3, and 0.74 μM kinetin and 58% with roots with 15.7 μM NAA and 47.08 μM AgNO3, but without kinetin. The shoots obtained showed little hyperhydricity. Vigorous plants were obtained after gradual acclimatization with an 80% survival rate under nursery conditions.  相似文献
