Aims
A growing body of research supports the feasibility of biocrust rehabilitation. Identifying populations of key species that are amenable to cultivation and that are resilient in rehabilitation contexts would advance the efficacy of these technologies. Here we investigate the growth and stress response of the cosmopolitan biocrust moss, Syntrichia ruralis.Methods
We sampled populations of S. ruralis along a precipitation seasonality gradient from the Colorado Plateau ecoregion of the western United States. We cultivated these populations in an experiment manipulating duration of hydration periods on a weekly cycle. We then treated greenhouse grown materials with brief, stressful watering events, measuring how many events they could survive.Results
All populations grew at an accelerated rate compared to growth in a natural setting, at least doubling biomass in five months. Increasing duration of hydration periods led to more growth in all but one population. Volunteer biocrust algae and cyanobacteria developed during cultivation, and differed among populations. Greenhouse grown mosses differed in their response to stressful watering, with the most susceptible populations dying at half the number events compared to the most tolerant.Conclusions
These findings argue for informed selection and deployment of Syntrichia ruralis populations for soil rehabilitation.213.

A lack of long-term monitoring often impedes the evaluation of translocation used to reinforce populations. Crucial questions regarding the exact timing and place of possible settlement remain unanswered. To examine these issues we radio-tracked during three years 24 tortoises (Testudo hermanni hermanni) released to reinforce a resident population impacted by fire. Individuals from the resident population (N = 20) and from a distant control population (N = 11) were also radio-tracked. More than 11,000 fixes were collected, enabling us to precisely describe movement patterns. Most translocated tortoises first dispersed (> 500 m to > 3000 m away) in a random direction and sometimes crossed unfavorable areas. Later, a marked shift in movement pattern, from a relatively unidirectional course to multidirectional displacements indicated settlement. Movement patterns of translocated and resident individuals became undistinguishable after settlement. Most individuals settled during the first year after release but several settled in the second year. Mean annual survival rate (> 85%) remained within the range of the species but was lower compared to the resident (93%) and control tortoises (100%). Overall, most translocated individuals (~ 63%) settled and adapted well to their novel environment. This result is essential regarding current controversies that are unfounded and that limit conservation translocations. Yet, translocation sites should be large enough and/or surrounded by secondary favorable areas to limit the mortality associated with dispersal in hazardous environments. Large numbers of individuals rescued during urbanization works may easily supply conservation translocations to reinforce fragile populations. 相似文献

Mycoplasma hyopneumoniae is the causative agent of enzootic pneumonia. In our previous work, we reconstructed the metabolic models of this species along with two other mycoplasmas from the respiratory tract of swine: Mycoplasma hyorhinis, considered less pathogenic but which nonetheless causes disease and Mycoplasma flocculare, a commensal bacterium. We identified metabolic differences that partially explained their different levels of pathogenicity. One important trait was the production of hydrogen peroxide from the glycerol metabolism only in the pathogenic species. Another important feature was a pathway for the metabolism of myo‐inositol in M. hyopneumoniae. Here, we tested these traits to understand their relation to the different levels of pathogenicity, comparing not only the species but also pathogenic and attenuated strains of M. hyopneumoniae. Regarding the myo‐inositol metabolism, we show that only M. hyopneumoniae assimilated this carbohydrate and remained viable when myo‐inositol was the primary energy source. Strikingly, only the two pathogenic strains of M. hyopneumoniae produced hydrogen peroxide in complex medium. We also show that this production was dependent on the presence of glycerol. Although further functional tests are needed, we present in this work two interesting metabolic traits of M. hyopneumoniae that might be directly related to its enhanced virulence. 相似文献

Photodynamic therapy combines three non-toxic components: light, oxygen and a photosensitizer to generate singlet oxygen and/or other ROS molecules in order to target destruction of cancer cells. The damage induced in the targeted cells can furthermore propagate to non-exposed bystander cells thereby exacerbating the damage. Ca2+ signaling is strongly intertwined with ROS signaling and both play crucial roles in cell death. In this review we aimed to review current knowledge on the role of Ca2+ and ROS signaling, their effect on cell-cell propagation via connexin-linked mechanisms and the outcome in terms of cell death. In general, photodynamic therapy results in an increased cytosolic Ca2+ concentration originating from Ca2+ entry or Ca2+ release from internal stores. While photodynamic therapy can certainly induce cell death, the outcome depends on the cell type and the photosensitizer used. Connexin channels propagating the Ca2+ signal, and presumably regenerating ROS at distance, may play a role in spreading the effect to neighboring non-exposed bystander cells. Given the various cell types and photosensitizers used, there is currently no unified signaling scheme to explain the role of Ca2+ and connexins in the responses following photodynamic therapy. In the Maremma Natural Park (Grosseto, Italy), the Collelungo beach exhibits a clear morphodynamics gradient, resulting in a severe erosion to a gradual accretion along the shoreline. Here, three sub-populations of the sandhopper Talitrus saltator (Montagu) (Crustacea, Amphipoda) were studied to highlight eventual intra- and inter-population variation of the endogenous locomotor rhythm. Individual activity recordings were carried out for 21 days in constant darkness and controlled temperature and humidity conditions. The variation of the free-running period was analysed at individual and sub-population levels, and the dependence on intrinsic (individual features) and extrinsic (habitat of origin and season) factors was analysed. A seasonal variation was highlighted, related to the seasonal spontaneous activity of the species. In summer and autumn, a significant correlation between the behavioural variation in the sub-populations (period and definition of the period) and beach physical parameters (slope and width) defined the dependence of the behavioural gradient on the environmental gradient. The observed relationship may represent an adaptation to the local coastline dynamics, suggesting the importance of a plastic behaviour as adaptive response to a variable environment. 相似文献

Kin selection theory predicts that costly cooperative behaviors evolve most readily when directed toward kin. Dispersal plays a controversial role in the evolution of cooperation: dispersal decreases local population relatedness and thus opposes the evolution of cooperation, but limited dispersal increases kin competition and can negate the benefits of cooperation. Theoretical work has suggested that plasticity of dispersal, where individuals can adjust their dispersal decisions according to the social context, might help resolve this paradox and promote the evolution of cooperation. Here, we experimentally tested the hypothesis that conditional dispersal decisions are mediated by a cooperative strategy: we quantified the density‐dependent dispersal decisions and subsequent colonization efficiency from single cells or groups of cells among six genetic strains of the unicellular Tetrahymena thermophila that differ in their aggregation level (high, medium, and low), a behavior associated with cooperation strategy. We found that the plastic reaction norms of dispersal rate relative to density differed according to aggregation level: highly aggregative genotypes showed negative density‐dependent dispersal, whereas low‐aggregation genotypes showed maximum dispersal rates at intermediate density, and medium‐aggregation genotypes showed density‐independent dispersal with intermediate dispersal rate. Dispersers from highly aggregative genotypes had specialized long‐distance dispersal phenotypes, contrary to low‐aggregation genotypes; medium‐aggregation genotypes showing intermediate dispersal phenotype. Moreover, highly aggregation genotypes showed evidence for beneficial kin‐cooperation during dispersal. Our experimental results should help to resolve the evolutionary conflict between cooperation and dispersal: cooperative individuals are expected to avoid kin‐competition by dispersing long distances, but maintain the benefits of cooperation by dispersing in small groups. 相似文献

A putative CLC voltage-gated anion channel gene from Aspergillus nidulans (AnCLCA) is characterised. The expression of the AnCLCA cDNA restored the iron-limited growth of the Saccharomyces cerevisiae CLC null mutant strain (gef1) suggesting that AnCLCA functions as a chloride channel. An AnCLCA conditional mutant was created and exhibited a strong and specific growth inhibition in the presence of extracellular copper concentrations >18 microM. This sensitivity was shown to be the result of a hyper-accumulation of copper by the conditional mutant, which generates superoxide to toxic levels inhibiting the growth. Further analysis revealed that copper dependent enzymes were disrupted in the AnCLCA conditional null mutant, specifically, a reduced activity of the copper-zinc superoxide dismutase (CuZn-SOD) and enhanced activity of the cytochrome oxidase (COX). These results suggest that AnCLCA plays a key role in copper homeostasis in A. nidulans and that a malfunction of this chloride channel results in disrupted intracellular copper trafficking. 相似文献

A simple and sensitive high-performance liquid chromatographic method is described for the determination of paclitaxel (Taxol®) at 230 nm using a Nucleosil C18 (5 μm) column and a methanol–water (70:30, v/v) mobile phase following a single-step extraction from serum with dichloromethane. The assay was validated against the classical criteria and was applied to a toxicokinetic study in rats after one or five, one per week) intraperitoneal administrations of 16 mg/kg Taxol®. 相似文献
