Temperature is a determinant environmental variable in metabolic rates of organisms ultimately influencing important physiological and behavioural features. Stressful conditions such as increasing temperature, particularly within high ranges occurring in the summer, have been suggested to induce flotation behaviour in Corbicula fluminea which may be important in dispersal of this invasive species. However, there has been no experimental evidence supporting this hypothesis. It was already proven that C. fluminea drift is supported by a mucilaginous drogue line produced by mucocytes present in the ctenidia. Detailed microscopic examination of changes in these cells and quantification of clam flotation following one, two and three weeks of exposure to 22, 25 and 30°C was carried out so that the effects of increasing water temperatures in dispersal patterns could be discussed. Results show that changes in temperature triggered an acceleration of the mucocytes production and stimulated flotation behaviour, especially following one week of exposure. Dilution of these effects occurred following longer exposure periods. It is possible that these bivalves perceive changing temperature as a stress and respond accordingly in the short-term, and then acclimate to the new environmental conditions. The response patterns suggest that increasing water temperatures could stimulate C. fluminea population expansion. 相似文献

It has been shown that 1-hour acute immobilization of rats resulted in a decrease of total protein content (per 1 cell) in neurones (by 17% in cytoplasm and 22% in nucleus) of the supraoptic nucleus of the hypothalamus and in their glial satellite cells (by 10%), mainly at the expense of the nuclei of the latter. These changes remained at the same level in the neuronal cytoplasm and satellite cells within 6 hours after cessation of acute immobilization, being abolished in the neuronal nuclei. Deprivation of REM sleep for 24 hours affected these parameters to the same extent as acute immobilization. However, 6-hour rest was sufficient for normalisation in satellite cells and for decrease in protein deficiency in the neuronal nuclei; yet protein deficiency still sustained in the neuronal cytoplasm. REM sleep deprivation was also accompanied by a decrease in tritium incorporation into satellite cell proteins (by 18%); after 3-hour rest, incorporation of the label into satellite cells was restored to the initial level being increased by 20% in neuronal nuclei; another 3-hour rest resulted in normalisation of labeling in the neuronal nuclei and in 27% increase of labeling in the satellite cells. 相似文献

Summary In southwestern Québec, non-harvested moose populations stabilize at a density of 0.40 animal·km-2. In an attempt to test whether or not moose were regulated by predators, we investigated wolf predation near this equilibrium density (0.37) and at 2 lower densities (0.23, 0.17). Scat analysis in summer and feeding observations in winter indicated a greater use of alternative food resources by wolves at lower moose densities. Each wolf pack killed on average 5.3, 1.8, 1.1 moose·100 days in the area of 0.37, 0.23, and 0.17 moose·km-2, respectively. Consumption of moose per wolf was 2.8, 1.7, and 1.6 kg/day, respectively. January wolf densities were estimated at 1.38, 0.82, and 0.36 animals·100 km-2, respectively. Year-long predation rates proved to be density-dependent, increasing with moose density from 6.1 to 19.3% of the postnatal populations. We conclude that moose populations in southwestern Québec are regulated largely by predators (wolves and maybe black bears) at a density where competition for forage produces no detrimental effect. We support the concept that wolf predation can have an important regulatory effect at low moose densities but also a depensatory (inversely density-dependent) effect at higher densities. 相似文献

The solution conformation of a synthetic peptide of 20 amino acids (P235-254) derived from the calmodulin-binding domain of Bordetella pertussis adenylate cyclase was studied by proton two-dimensional NMR spectroscopy and circular dichroism. Based on the standard techniques we have assigned all the resonances in the NMR spectrum to the corresponding protons of the peptide. Analysis of the secondary chemical shift distribution and of the nuclear Overhauser effect connectivities showed no evidence for a highly populated regular conformation but suggested the tendency to form an alpha-helix around the unique Trp residue. The propensity for a helical structure is in agreement with the results of circular dichroic spectroscopy showing a slight negative band at 222 nm which was cancelled by 6 M guanidine hydrochloride. Increasing amounts of 2,2,2-trifluoroethanol (up to 40%) increase considerably the helical population of the peptide as reflected in the circular dichroic spectra. Analysis of the present results shows that the free peptide P235-254 has the tendency to form a basic amphiphilic helix. The presence of two acid residues, Glu236 and Asp239, on the hydrophilic side of the alpha-helix, which is mainly composed by basic residues, may explain the lower affinity of this peptide for calmodulin as compared with other peptides derived from calmodulin-activated enzymes. 相似文献

The thymus supports differentiation of T cell precursors. This process requires relocation of developing thymocytes throughout multiple microenvironments of the organ, mainly with thymic epithelial cells (TEC), which control intrathymic T cell differentiation influencing the formation and maintenance of the immunological synapse. In addition to the proteins of the major histocompatibility complex (MHC), this structure is supported by several adhesion molecules. During the process of thymopoiesis, we previously showed that laminin-mediated interactions are involved in the entrance of T-cell precursors into the thymus, as well as migration of differentiating thymocytes within the organ. Using small interference RNA strategy, we knocked-down the ITGA6 gene (which encodes the CD49f integrin α-chain) in cultured human TEC, generating a decrease in the expression of the corresponding CD49f subunit, in addition to modulation in several other genes related to cell adhesion and migration. Thymocyte adhesion to TEC was significantly impaired, comprising both immature and mature thymocyte subsets. Moreover, we found a modulation of the MHC, with a decrease in membrane expression of HLA-ABC, in contrast with increase in the expression of HLA-DR. Interestingly, the knockdown of the B2M gene (encoding the β-2 microglobulin of the HLA-ABC complex) increased CD49f expression levels, thus unraveling the existence of a cross-talk event in the reciprocal control of CD49f and HLA-ABC. Our data suggest that the expression levels of CD49f may be relevant in the general control of MHC expression by TEC and consequently the corresponding synapse with developing thymocytes mediated by the T-cell receptor. 相似文献

While the xylem hydraulic properties, such as vulnerability to cavitation (VC), are of paramount importance in drought resistance, their genetic determinants remain unexplored. There is evidence that pectins and their methylation pattern are involved, but the detail of their involvement and the corresponding genes need to be clarified. We analyzed the hydraulic properties of the 35S::PME1 transgenic aspen that ectopically under‐ or over‐express a xylem‐abundant pectin methyl esterase, PtxtPME1. We also produced and analyzed 4CL1::PGII transgenic poplars expressing a fungal polygalacturonase, AnPGII, under the control of the Ptxa4CL1 promoter that is active in the developing xylem after xylem cell expansion. Both the 35S::PME1 under‐ and over‐expressing aspen lines developed xylem with lower‐specific hydraulic conductivity and lower VC, while the 4CL1::PGII plants developed xylem with a higher VC. These xylem hydraulic changes were associated with modifications in xylem structure or in intervessel pit structure that can result in changes in mechanical behavior of the pit membrane. This study shows that homogalacturonans and their methylation pattern influence xylem hydraulic properties, through its effect on xylem cell expansion and on intervessel pit properties and it show a role for PtxtPME1 in the xylem hydraulic properties. 相似文献

The radiation of neotropical echimyid rodents resulted in the diversification of spiny rats, hutias, and the coypu, grouped into four major clades. Different echimyid lineages specialized for terrestrial, semifossorial, semiaquatic, and arboreal locomotion, and for living in bamboo thickets. Comparative phylogenetic methods were herein used for evaluating the relative effect of historical and ecological factors in shaping morphometric traits of external morphology (length of head and body, tail, ear, and hindfeet). Additionally, we investigated whether the adoption of different locomotory habits was associated with changes in their macroevolutionary dynamics. Our findings showed that variation of all traits was phylogenetically structured, although size was less structured than other traits. Tail, ear, and mainly hindfeet were allometrically correlated with head and body length, indicating the occurrence of evolutionary structural constraints. In addition to phylogenetic and allometric components, morphometric variation was strongly associated with locomotory specializations, except for body size. Disparification of body size and tail length took place gradually, fitting the Brownian motion model albeit with some punctual shifts in evolutionary rates. Some of these shifts were associated with changes in locomotory habits, notably with adoption of a semiaquatic habit by the large‐sized Myocastor lineage, which occurred with an accelerated rate of size evolution. Evolutionary changes in ear and hindfeet length were concentrated during early echimyid diversification, concomitantly with the emergence of most locomotory habits, with subsequent deceleration of evolutionary rates. Fusarium species have emerged as responsible for a broad spectrum of infections, including superficial, locally invasive and disseminated ones, especially in the hospital environment. Since there are few reports of invasive and disseminated fusariosis in children, the aim of this study was to report four cases of nosocomial infection caused by this microorganism in children with cancer hospitalized in a public children’s hospital located in Brazil. Two of these patients were female and two were male. All patients presented febrile neutropenia, while three patients had acute lymphocytic leukemia and one patient had Wilms’ tumor as underlying disease. In two cases, fungi were isolated from blood and identified as Fusarium oxysporum species complex after phenotypic and genotypic studies, while in two other cases fungi were isolated from skin biopsies and identified as Fusarium solani species complex. One patient died 12 days after the onset of cutaneous lesions. All isolates, after susceptibility testing, presented high levels of minimum inhibitory concentration for itraconazole, voriconazole and amphotericin B. Considering the emergence of filamentous fungi as etiologic agents of nosocomial infections, health professionals should be aware of the problems these infections, especially fungal ones, may cause to debilitated patients. 相似文献
