Polydactyly (Po) is a common mutation found in many vertebrates. The UCD-Po.003 congenic chicken line was previously characterized for Po inheritance (autosomal dominant) and the mutation was mapped to chromosome 2p. Here, we describe for the first time the range and variability of the phenotype in this congenic line. Further, we studied the hypothesis that a chromosomal inversion was responsible for the maintenance of a large (6.3 Mb) candidate gene region. Fluorescence in situ hybridization employing BACs encompassing a 10.7-Mb region of GGA2p showed that the Po chromosome was normal, i.e. exhibits the wild-type BAC order. Continued fine-mapping along with a change in breeding strategy reduced the size of the causative region to 1.43 Mb. Recent research indicates that the cause of preaxial Po resides within a 794-bp highly conserved zone of polarizing activity regulatory sequence (ZRS) element located in intron 5 of the LMBR1 gene; however, the ZRS polymorphism of interest is found in some but not all breeds of polydactylous chicken. Therefore, we sequenced the ZRS in 101 heterozygous and 30 unaffected (wild-type) individuals to establish the relevance of this region to the Po condition in the UCD-Po.003 congenic line. A single point mutation (C/A at coordinate GGA2p: 8,414,121) within the ZRS segregated with carrier status. The polydactylous UCD-Silkie line also maintains this SNP in addition to a single base deletion. An inheritance analysis of the phenotypic variation in UCD-Po.003 suggests recessive epistasis as the mode of inheritance for the additional modifying genetic elements, residing outside the ZRS, to impact the preaxial polydactyl phenotype. These results contribute to our understanding of the cause of Po in an important vertebrate model.  相似文献

The SLC26 gene family encodes multifunctional transport proteins in numerous tissues and organs. Some paralogs function as anion exchangers, others as anion channels, and one, prestin (SLC26A5), represents a membrane-bound motor protein in outer hair cells of the inner ear. At present, little is known about the molecular basis of this functional diversity. We studied the subunit stoichiometry of one bacterial, one teleost, and two mammalian SLC26 isoforms expressed in Xenopus laevis oocytes or in mammalian cells using blue native PAGE and chemical cross-linking. All tested SLC26s are assembled as dimers composed of two identical subunits. Co-expression of two mutant prestins with distinct voltage-dependent capacitances results in motor proteins with novel electrical properties, indicating that the two subunits do not function independently. Our results indicate that an evolutionarily conserved dimeric quaternary structure represents the native and functional state of SLC26 transporters.  相似文献

Multispecies cicada communities in neotropical rainforests produce a complex and intense acoustic environment. In a fragment of a Mexican rainforest (Veracruz, Mexico), a cicada community at the end of the dry season consisted of nine species ( Daza montezuma; Pacarina schumanni; Miranha imbellis; Dorisiana sutori; Fidicinoides picea; Fidicinoides pronoe; Quesada gigas; one species of the genus Neocicada and one uncaught canopy species). Seven of the nine species formed dense choruses at dawn and at dusk. Each species showed preferences in the height of calling sites. Males of the species were solitary or gregarious, and followed a 'call-fly' or a 'call-stay' calling strategy. Acoustic signals of each species had particular time and frequency patterns. All these specific features appear to separate the nine species acoustically and lead to a partitioning of the acoustic environment. In an attempt to recognize the possible ecological causes of the decline of a population of Hoopoes Upupa epops in the Swiss Alps, we collected data on resource exploitation. The prey provisioned to nestlings by parents was investigated at four breeding sites using photographs (n = 4353, 80% of which enabled prey identification). Molecrickets Gryllotalpa gryllotalpa and Lepidoptera (larvae and pupae) were dominant in nestling diet (93% frequency; 97% biomass). Although Molecrickets were provisioned less frequently (26%) than Lepidoptera (67%), they represented 68% of the total biomass (vs 29% for Lepidoptera). There was an overall negative relationship between the proportion of Molecricket biomass in the diet and the parents' feeding rate, whereas a comparison between broods showed that a higher provisioning activity did not lead to an increase in the biomass supplied to the chicks. A diet based on Molecrickets therefore appears to be energetically advantageous. As Molecrickets are a traditional prey of Hoopoes in central Europe, this might be relevant to other populations. In the study area, Molecrickets occur only on the intensively cultivated plain, whereas the majority of Hoopoe pairs nest at various altitudes on the foothills adjacent to the plain as the latter provides at present almost no suitable nesting sites. Hoopoes breeding higher up on the foothills seem to experience greater provisioning costs and have, on average, lower breeding success. Providing nest sites on the plain is the main conservation measure proposed for the local Hoopoe population. Further attention should also be paid to Molecrickets as these may be crucial for Hoopoes.  相似文献
