The flight behavior of Anopheles gambiae s.s. Giles and An. stephensi Patton exposed to different odor cues was studied in a wind tunnel. Odors consisted of CO2, CO2 + acetone (at two concentrations), and CO2 + 1-octen-3-ol. Mosquitoes were released singly and their behavior was recorded on video. Parameters studied included flight velocity, percentage of time spent flying, percentage of time spent in plume, and number of turns toward the plume. Large differences in behavior toward the odors tested were observed. An. gambiaedid not respond well to CO 2,whereas An. stephansiwas positively affected by this compound. In contrast, An. gambiaeresponded significantly to CO 2 + acetone (at a low concentration), but the behavior of An. stephensiwas completely suppressed by this combination of odor stimuli. CO 2 + a high concentration of acetone or CO 2 + 1-octen-3-ol did not cause significant effects in An. gambiaecompared to no odor, while these treatments elicited strong behavioral responses in An. stephensi.The latter species responded particularly well to CO 2 + 1-octen-3-ol. The results suggest that the observed differences may be inherent to the known differences in host preferences, where An. gambiaeis highly anthropophilic and An. stephensimore zoophilic. This would explain why the latter species responds well to CO 2 and even better to CO 2 + 1-octen-3-ol, a compound readily emitted by bovine ruminants.  相似文献

Man-made raised sandy soils in the Netherlands are classified as brown or black plaggen soils. When dry, the brown soils are wettable, but the black soils are water repellent. For one growing season, transects were sampled in a maize cropped black plaggen soil at the Heino experimental farm. Due to interception and stemflow, water was concentrated near the roots of the maize. Between the maize rows, higher soil water contents were found in microdepressions, due to rainwater dripping to the ground from overhanging leaves. Redistribution of soil water from wet to dry areas was restricted by the water repellency of the dry sand. As a consequence, there was a distinct variation in soil moisture content. These irregular wetting patterns did not induce preferential downward flow, but widened over time; because the dry, water repellent subsoil impeded and resisted infiltration into the deeper subsoil.  相似文献

In the parallel paper, we developed a property to characterize drug efflux pumps, i.e. the reduced relative resistance (RRR). Using this RRR, we here investigate whether the observed diversity in human multidrug resistance (MDR) phenotypes might be due to variable levels of P-glycoprotein encoded by MDR1. We analyzed resistance phenotypes of various human cell lines in which either one, or both, classical human multidrug resistance genes, MDR1 and MDR3, are overexpressed. In addition, RRR values were calculated for MDR phenotypes presented in the literature. The results suggest that more than a single mechanism is required to account for the observed phenotypic diversity of classical multidrug resistance. This diversity is only partly due to differences in plasma membrane permeabilities between cell line families. It is discussed whether the alternative MDR phenotypes might be MDR1 phenotypes modified by other factors that do not themselves cause MDR. The method we here apply may also be useful for other nonspecific enzymes or pumps.  相似文献

Abstract   The drosophilid fauna is well documented in eastern Australia but is poorly known in other parts of the continent. This paper summarises what is known of this fauna in the Northern Territory (NT), and includes results from banana trapping in the humid and arid zones. The 42 recorded species include species that breed in fruit, fungi and/or flowers, and a larval predator of scale insects. Drosophilids occur in all three major climate zones (humid, semiarid and arid) but predominate in the humid zone. Banana-attracted species in the humid zone (wet-dry tropics) were common in all sampled habitats: urban, rainforest and open woodland. They included predominantly urban and/or rainforest species. Of the species collected in open woodland, some are likely to be breeding there, whereas others may have been intercepted during movement across the area. The semiarid fauna is a depauperate version of that found in the humid region. Only three species have been recorded in the arid region: an endemic arid specialist, and two cosmopolitan species ( D. simulans and D. melanogaster ) in urban Alice Springs. Overall, the NT drosophilid fauna represents a depauperate version of that found in eastern Australia, probably because of climatic factors and natural barriers to range expansion. There is little evidence of regional endemism, with probably only one (and at most three) species endemic to the NT, and no evidence of independent, natural dispersion from nearby Indonesia.  相似文献

Summary The effect of long-term testosterone propionate treatment on the adult female rat and hamster anterior pituitary was investigated by means of combined bioassay and electron microscopy. In the rat, a marked hypertrophy and enhancement of secretory activity of FSH cells occurred which correlated well with a marked increase of follicle stimulating hormone (FSH) stores. A moderate decrease of luteinizing hormone (LH) stores was not accompanied by changes in so-called LH cells. Alterations in acidophilic cell types were also observed. Somatotrophs were atrophic and showed evidence of decreased secretory activity, while lactotrophs appeared to be stimulated by testosterone treatment. Cyanobacteria respond to iron deficiency during growth by expressing the isiA gene, which produces a chlorophyll-carotenoid protein complex known as IsiA or CP43′. Long-term iron deficiency results in the formation of large IsiA aggregates, some of which associate with photosystem I (PSI) while others are not connected to a photosystem. The fluorescence at room temperature of these unconnected aggregates is strongly quenched, which points to a photoprotective function. In this study, we report time-resolved fluorescence measurements of IsiA aggregates at low temperatures. The average fluorescence lifetimes are estimated to be about 600 ps at 5 K and 150 ps at 80 K. Both lifetimes are much shorter than that of the monomeric complex CP47 at 77 K. We conclude that IsiA aggregates quench fluorescence to a significant extent at cryogenic temperatures. We show by low-temperature fluorescence spectroscopy that unconnected IsiA is present already after two days of growth in an iron-deficient medium, when PSI and PSII are still present in significant amounts and that under these conditions the fluorescence quenching is similar to that after 18 days, when PSI is almost completely absent. We conclude that unconnected IsiA provides photoprotection in all stages of iron deficiency.  相似文献

A model is proposed for chromophore optical spectra in solids over a wide range of temperatures and pressures. Inhomogeneous band shapes and their pressure dependence, as well as baric shift coefficients of spectral lines, selected by the frequency, were derived using Lennard-Jones potentials of the ground and excited states. Quadratic electron-phonon coupling constants, describing the thermal shift and broadening of zero-phonon lines, were also calculated. Experimentally, thermal shift and broadening of spectral holes were studied between 5 and 40 K for a synthetic pigment, chlorin, embedded in polymer hosts. The baric effects on holes were determined by applying hydrostatic He gas pressure up to 200 bar, at 6 K. Absorption spectra of pheophytin a, chlorophyll a, and beta-carotene in polymers and plant photosystem II CP47 complex were measured between 5 (or 77) and 300 K, and subject to Voigtian deconvolution. A narrowing of inhomogeneous bandwidth with increasing temperature, predicted on the basis of hole behavior, was observed as the shrinking of Gaussian spectral component. The Lorentzian broadening was ascribed to optical dephasing up to 300 K in transitions with weak to moderate linear electron-phonon coupling strength. The thermal broadening is purely Gaussian in multiphonon transitions (S(2) band of beta-carotene, Soret bands of tetrapyrrolic pigments), and the Lorentz process appears to be suppressed, indicating a lack of exponential dephasing. Density, polarity, polarizability, compressibility, and other local parameters of the pigment binding sites in biologically relevant systems can be deduced from spectroscopic data, provided that sufficient background information is available.  相似文献

A new sutureless technique to successfully anastomose the abdominal aorta of rats (1.3 mm in diameter) by using a fully biodegradable, laser-activated protein solder is presented. A total of 90 rats were divided into two groups randomly. In group one, the anastomoses were performed by using conventional microsuturing technique, whereas in group two, the anastomoses were performed by using a new laser welding technique. In addition, each of the two groups were divided into five subgroups and evaluated at different follow-up periods (10 minutes, 1 hour, 1 day, 1 week, and 6 weeks). At these intervals, the anastomoses were evaluated for patency and tensile strength. Three anastomoses in each subgroup were processed for light and electron microscopy. All anastomoses were found to be patent. The mean clamp time of the anastomoses performed with conventional suturing was 20.6 minutes compared with 7.2 minutes for the laser-activated welded anastomoses (p < 0.001). The strain measurements showed a stronger mechanical bond of the sutured anastomoses in the initial phase. However, at 6 weeks the tensile strength of the laser-welded anastomoses was higher compared with the conventional suture technique. Histologic evaluations revealed a near complete resorption of the solder after 6 weeks. The junction site of the vessel ends cannot be determined on the luminal side of the artery. In conclusion, a resorbable protein used as a solder, activated by a diode laser, can provide a reliable, safe, and rapid arterial anastomosis, which could be performed by any microsurgeon faster than conventional suturing after a short learning curve.  相似文献

Neurofilaments (NFs) are neuron-specific intermediate filaments, and are the major cytoskeletal component in large myelinated axons. Lysine-serine-proline (KSP) repeats in the tail domains of high molecular weight NF proteins (NF-M and NF-H) are extensively phosphorylated in vivo in the axon. This phosphorylation in the tail domain has been postulated to play an important role in mediating neuron-specific properties, including axonal caliber and conduction velocity. Recent studies have shown that the mitogen-activated protein kinases (extracellular signal-regulated kinases, Erk1 and Erk2) phosphorylate KSP motifs in peptide substrates derived from the NF-M and NF-H tail domains in vitro. However, it is not clear whether activation of the mitogen activated protein (MAP) kinase pathway is able to phosphorylate these domains in vivo. To answer this question, a constitutively active form of mitogen-activated Erk activating kinase (MEK1) was cotransfected with an NF-M expression construct into NIH 3T3 cells. The activated mutant, but not the dominant negative mutant, induced phosphorylation of NF-M. In addition, it was shown that epidermal growth factor, which induces the MAP kinase cascade in NIH 3T3 cells, also activated endogenous Erk1 and Erk2 and NF-M tail domain phosphorylation in the transfected cells. These results present direct evidence that in-vivo activation of Erk1 and Erk 2 is sufficient for NF-M tail domain phosphorylation in transfected cells.  相似文献

We report that a long-distance migrating shorebird, the red knot, makes a complete switch from commonly occurring monoester preen waxes to a much rarer class of higher-molecular-weight diester waxes at the time of take-off to the high arctic breeding grounds. The cold arctic climate would have required a lowering of wax-viscosity, and thus, a shift in the reverse direction. We propose that a sexually selected need for a brilliant plumage has lead to this conter-intuitive temporary shift from monoesters to diester waxes. The difficulty of application of the diester preen waxes under cold conditions would ensure the reliability of the quality-signalling function of this most probably sexually selected trait.  相似文献
