The multitude of terpene carbon skeletons in plants is formed by enzymes known as terpene synthases. This review covers the monoterpene and sesquiterpene synthases presenting an up-to-date list of enzymes reported and evidence for their ability to form multiple products. The reaction mechanisms of these enzyme classes are described, and information on how terpene synthase proteins mediate catalysis is summarized. Correlations between specific amino acid motifs and terpene synthase function are described, including an analysis of the relationships between active site sequence and cyclization type and a discussion of whether specific protein features might facilitate multiple product formation.  相似文献

A municipal solid-waste bottom slag was used to grow maize plants under various abiotic stresses (high pH, high salt and high heavy metal content) and to analyse the structural and chemical adaptations of the cell walls of various root tissues. When compared with roots of control plants, more intensive wall thickenings were detected in the inner tangential wall of the endodermis. In addition, phi thickenings in the rhizodermis in the oldest part of the seminal root were induced when plants were grown in the slag. The role of the phi thickenings may not be a barrier for solutes as an apoplastic dye could freely diffuse through them. The chemical composition of cell walls from endodermis and hypodermis was analysed. Slag-grown plants had higher amounts of lignin in endodermal cell walls when compared to control plants and a higher proportion of H-type lignin in the cell walls of the hypodermis. Finally, the amount of aliphatic suberin in both endo- and hypodermal cell walls was not affected by growing the plants on slag. The role of these changes in relation to the increase in mechanical strengthening of the root is discussed.  相似文献

The aim of this work was to investigate the use of torasemide as a highly sensitive indicator substance and to develop a formulation thereof for establishing quantitative relationships between hot-melt extrusion process conditions and critical quality attributes (CQAs). Using solid-state characterization techniques and a 10 mm lab-scale co-rotating twin-screw extruder, we studied torasemide in a Soluplus® (SOL)-polyethylene glycol 1500 (PEG 1500) matrix, and developed and characterized a formulation which was used as a process indicator to study thermal- and hydrolysis-induced degradation, as well as residual crystallinity. We found that torasemide first dissolved into the matrix and then degraded. Based on this mechanism, extrudates with measurable levels of degradation and residual crystallinity were produced, depending strongly on the main barrel and die temperature and residence time applied. In addition, we found that 10% w/w PEG 1500 as plasticizer resulted in the widest operating space with the widest range of measurable residual crystallinity and degradant levels. Torasemide as an indicator substance behaves like a challenging-to-process API, only with higher sensitivity and more pronounced effects, e.g., degradation and residual crystallinity. Application of a model formulation containing torasemide will enhance the understanding of the dynamic environment inside an extruder and elucidate the cumulative thermal and hydrolysis effects of the extrusion process. The use of such a formulation will also facilitate rational process development and scaling by establishing clear links between process conditions and CQAs.  相似文献

Internal eliminated sequences (IESs) often interrupt ciliate genes in the silent germline nucleus but are exactly excised and eliminated from the developing somatic nucleus from which genes are then expressed. Some long IESs are transposons, supporting the hypothesis that short IESs are ancient transposon relics. In light of that hypothesis and to explore the evolutionary history of a collection of IESs, we have compared various alleles of a particular locus (the 81 locus) of the ciliated protozoa Oxytricha trifallax and O. fallax. Three short IESs that interrupt two genes of the locus are found in alleles from both species, and thus must be relatively ancient, consistent with the hypothesis that short IESs are transposon relics. In contrast, TBE1 transposon interruptions of the locus are allele-specific and probably the results of recent transpositions. These IESs (and the TBE1s) are precisely excised from the DNA of the developing somatic macronucleus. Each IES interrupts a highly conserved sequence. A few nucleotides at the ends of each IES are also conserved, suggesting that they interact critically with IES excision machinery. However, most IES nucleotide positions have evolved at high rates, showing little or no selective constraint for function. Nonetheless, the length of each IES has been maintained (+/- 3 bp). While one IES is approximately 33 bp long, three other IESs have very similar sizes, approximately 70 bp long. Two IESs are surrounded by direct repeats of the sequence TTCTT. No other sequence similarities were found between any of the four IESs. However, the ends of one IES do match the inverted terminal repeat consensus sequence of the "TA" IESs of Paramecium. Three O. trifallax alleles appear to have been recipients in recent conversion events that could have been provoked by double-strand breaks associated with IES ends subsequent to IES transposition. Our findings support the hypothesis that short IESs evolved from ancient transposons that have lost most of their sequences, except those necessary for precise excision during macronuclear development.   相似文献

The ecology and taxonomy of the epilithic diatom flora of the Agnéby River, Ivory Coast were studied in 2012. Ten sites were investigated and diatoms were sampled on glass slides immersed for a period of 30 days during the wet and dry seasons. Physico-chemical parameters were measured at each site while sampling diatoms. Five taxa were largely dominant: Planothidium comperei CE Wetzel, N’Guessan and Tison-Rosebery, Eolimna minima (Grunow) Lange-Bertalot, Planothidium piaficum (JR Carter and Denny) CE Wetzel and Ector, Cocconeis schroederi Foged and Cocconeis scutellum var. parva (Grunow in Van Heurck) Cleve. Electrical conductivity, temperature, dissolved oxygen, nitrite and phosphorus were found to influence the distribution of taxa.  相似文献

The biogeochemistry of Ca, Mg, K, and Nawere investigated in two forested catchments in theCzech Republic, one underlain by leucogranite, theother by serpentinite. High weathering rates at theserpentinite site at Pluhv Bor resultedin Mg2+ as the dominant cation on the soilexchange complex and in drainage water. Other basiccations (Ca2+, K+, Na+) showedrelatively low concentrations and outflow instreamwater. The catchment exhibited high basesaturation in mineral soils (>70%), and nearneutral soil and stream pH, despite elevated inputsof acidic deposition. Slow growth of Norway spruceat Pluhv Bor may be caused by K deficiency, Mgoversupply and/or Ni toxicity. In contrast, thegranitic site at Lysina showed low concentrations ofbasic cations on the soil exchange complex and instreamwater. Soil and drainage water at Lysina werehighly impacted by acidic deposition. Soil pH wasextremely acidic (<4.5) throughout the soilprofile, and the base saturation of the mineral soilwas very low (<5%). Supplies of basic cationsfrom atmospheric deposition and soil processes wereless than inputs of SO2- 4 on anequivalence basis, resulting in low pH and highconcentrations of total Al in drainage water. Needle yellowing in Norway spruce was possibly theresult of Mg deficiency at Lysina. Because of theirextremely different lithologies, these catchmentsserve as valuable end-members of ecosystemsensitivity to elevated levels of acidicdeposition.  相似文献

Substance use is a major cause of disability globally. This has been recognized in the recent United Nations Sustainable Development Goals (SDGs), in which treatment coverage for substance use disorders is identified as one of the indicators. There have been no estimates of this treatment coverage cross‐nationally, making it difficult to know what is the baseline for that SDG target. Here we report data from the World Health Organization (WHO)'s World Mental Health Surveys (WMHS), based on representative community household surveys in 26 countries. We assessed the 12‐month prevalence of substance use disorders (alcohol or drug abuse/dependence); the proportion of people with these disorders who were aware that they needed treatment and who wished to receive care; the proportion of those seeking care who received it; and the proportion of such treatment that met minimal standards for treatment quality (“minimally adequate treatment”). Among the 70,880 participants, 2.6% met 12‐month criteria for substance use disorders; the prevalence was higher in upper‐middle income (3.3%) than in high‐income (2.6%) and low/lower‐middle income (2.0%) countries. Overall, 39.1% of those with 12‐month substance use disorders recognized a treatment need; this recognition was more common in high‐income (43.1%) than in upper‐middle (35.6%) and low/lower‐middle income (31.5%) countries. Among those who recognized treatment need, 61.3% made at least one visit to a service provider, and 29.5% of the latter received minimally adequate treatment exposure (35.3% in high, 20.3% in upper‐middle, and 8.6% in low/lower‐middle income countries). Overall, only 7.1% of those with past‐year substance use disorders received minimally adequate treatment: 10.3% in high income, 4.3% in upper‐middle income and 1.0% in low/lower‐middle income countries. These data suggest that only a small minority of people with substance use disorders receive even minimally adequate treatment. At least three barriers are involved: awareness/perceived treatment need, accessing treatment once a need is recognized, and compliance (on the part of both provider and client) to obtain adequate treatment. Various factors are likely to be involved in each of these three barriers, all of which need to be addressed to improve treatment coverage of substance use disorders. These data provide a baseline for the global monitoring of progress of treatment coverage for these disorders as an indicator within the SDGs.  相似文献

The distribution of the phytohormone abscisic acid (ABA) between plant and soil and within plants growing on an alkaline substrate has been studied in order to separate the true effect of high soil pH from any effects that might be a result of the high salinity normally observed in alkaline soils. Leaves of a range of plants grown in an alkaline and saline solid substrate (municipal solid waste incinerator bottom slag) exhibited higher ABA levels than leaves of control plants. In contrast, roots of most plants grown on alkaline and saline substrates, particularly those without an exodermis (various species of Fabaceae), had slightly lower than or comparable ABA contents to control roots. However, in corn roots (Zea maysL. cv. Garant FAO 240) which possess a well-developed exodermis, alkaline and saline conditions in the rhizosphere did not reduce the endogenous ABA concentration, because the leaching of ABA from corn roots into the rhizosphere was lower than that from Vicia faba (variety Dreifache Weisse) roots. ABA efflux from corn and Vicia roots into the soil solution was observed only during the first days of the experiments and thereafter became substantially decreased. Because the leaching of ABA from Vicia faba roots into the rhizosphere was higher than that from corn roots, the leaves of Vicia plants grown in alkaline soil at low salinity no longer exhibited an elevated ABA concentration. However, whilst the roots of corn plants grown on desalted slag retained ABA levels that were higher than those of the control, the ABA content of leaves was not significant higher than the controls. For this reason, root ABA retention must be enough to induce tolerance to alkalinity in corn plants and there is no need to implicate changes in ABA concentrations in the aerial parts of the plant as having a role in this tolerance. In alkaline soil substrates, considerable portions of the ABA synthesised in the roots leached out into the soil solution of the rhizosphere according to the anion trap concept. An exodermis substantially reduces this leakage. The transient nature of ABA efflux into the rhizosphere was a result of the fact that the salt stress itself was only a transient phenomenon due to a washout of salt by irrigation. The results match predictions of mathematical models describing the effect of alkaline pH on the distribution of abscisic acid within plants and between roots and the rhizosphere. Species that can retain root ABA in the face of its tendency to leach into the more alkaline compartment are able to tolerate these normally harmful sites.
