Intestinal gene regulation involves mechanisms that direct temporal expression along the vertical and horizontal axes of the alimentary tract. Sucrase-isomaltase (SI), the product of an enterocyte-specific gene, exhibits a complex pattern of expression. Generation of transgenic mice with a mutated SI transgene showed involvement of an overlapping CDP (CCAAT displacement protein)-GATA element in colonic repression of SI throughout postnatal intestinal development. We define this element as CRESIP (colon-repressive element of the SI promoter). Cux/CDP interacts with SI and represses SI promoter activity in a CRESIP-dependent manner. Cux/CDP homozygous mutant mice displayed increased expression of SI mRNA during early postnatal development. Our results demonstrate that an intestinal gene can be repressed in the distal gut and identify Cux/CDP as a regulator of this repression during development. 相似文献

Previous research has shown that administration of either testosterone or estradiol to male quail embryos will demasculinize behavior and morphology. Six experiments in which embryos were treated were conducted to test the hypothesis that this testosterone-induced demasculinization is due to conversion of testosterone to estrogen (aromatization). In Experiment 1, dihydrotestosterone propionate, a nonaromatizable androgen, failed to demasculinize copulatory behavior, but did demasculinize crowing, strutting, and proctodeal glands. In Experiment 2, injection of the aromatizable androgens testosterone propionate (TP), testosterone, or androstenedione demasculinized copulatory behavior, the nonaromatizable androgen androsterone failed to have such an effect, and all androgens demasculinized proctodeal glands. In Experiment 3, Silastic implants of testosterone demasculinized all male characteristics, whereas implants of androsterone demasculinized only proctodeal glands. In Experiment 4, the antiestrogen tamoxifen prevented TP from demasculinizing copulatory behavior, but had no such effect with respect to crowing and strutting. In Experiments 5 and 6, the aromatization inhibitor 1,4,6-androstatrien-3,17-dione (ATD) prevented TP but not estradiol benzoate from demasculinizing copulatory behavior. Thus (1) in quail, testosterone-induced demasculinization of copulatory behavior is due to androgen aromatization, whereas testosterone-induced demasculinization of crowing, strutting, and proctodeal glands is not; (2) the distinct components of normal male reproductive behavior exhibit different patterns of steroid specificity during the organizational period, as was previously shown for the activational period; (3) the steroid specificity of crowing, strutting, and proctodeal glands changes between the organizational and activational periods. During organization, there is little specificity, whereas during activation, these characteristics respond only to androgens, never to estrogens. Introduced species can have profound effects on native species, communities, and ecosystems, and have caused extinctions or declines in native species globally. We examined the evolutionary response of native zooplankton populations to the introduction of non-native salmonids in alpine lakes in the Sierra Nevada of California, USA. Fenamate NSAIDs have several central effects, including anti-epileptic and neuroprotective actions. The underlying mechanism(s) of these actions are not presently understood. In this study, the effects of five members of the fenamate NSAID group were investigated on native ligand-gated ion channels expressed in cultured rat hippocampal neurons. All fenamates tested (1-100 microM) dose-dependently potentiated GABA-evoked currents; mefenamic acid (MFA) was the most potent and efficacious and was found to shift the GABA dose-response curve to the left without effect on the maximum amplitude or the GABA Hill Slope. The modulation of GABA receptors by MFA was not reduced in the presence of the benzodiazepine antagonist, flumazenil (10 microM) and was moderately voltage-dependent. MFA at concentrations >or=10 microM evoked dose-dependent currents in the absence of GABA. These currents were potentiated by diazepam (1 microM) and blocked by bicuculline (10 microM). The MFA (50 microM) current-voltage relationship and reversal potential were similar to that evoked by GABA. MFA (1-100 microM) had no effects on sub-maximal glycine, glutamate or NMDA evoked currents. The Hazard Analysis and Critical Control Point (HACCP) monitoring system has been used traditionally to increase quality control in human food production operations and there is pressure to implement it at the producer and purchaser levels of the food chain. Recently, the concept of HACCP monitoring has extended to food fed to domestic animals. Captive wildlife facilities, such as zoos and aquaria, would benefit from a well‐organized, food safety and nutritional monitoring system. Zoos and aquaria spend significant resources in time and money on maintaining the health of their animals; much of this energy is focused on disease prevention and adequate nutrition. The result of these combined efforts is the implementation of a HACCP program in zoo food management. Although zoo food handling standards have been implemented through the American Zoo and Aquarium Association (AZA) accreditation process, food borne disease outbreaks and malnutrition still exist. By implementing an organized approach to monitoring the quality of food delivered to the animals, the safety and nutritional value of the foods will increase, while decreasing the financial loss due to food waste and time spent caring for ill animals. Michael Hanchard’s the Spectre of Race: How Discrimination Haunts Western Democracy is a sophisticated examination of the disciplinary absence and seething presence of race in the subfield and substance of comparative politics. Hanchard’s analysis reveals a genealogy of how certain concepts, such as political culture, came to be institutionalized in the discipline. Because disciplines discipline, the resultant marginalization of race in comparative politics is itself an act of power. Many of his insights are revelatory, though a more explicit excavation of racial transnationalism is warranted. Such an effort would: first, demonstrate that the transnational “entanglements” Hanchard details are even more knotted than originally presumed; second, challenge the conflation of racialization and colonialism; and third, question whether liberal democratic inclusion is possible or even worth the price of the ticket. 相似文献
