Browsing is one of the main factors determining survival, growth rate, woodland structure, and distribution of the high mountain tree Polylepis australis. This species has a substantial regrowth capacity, which may function as a mechanism to tolerate herbivory, but it is unknown to what extent it may compensate for the impact of herbivory. In 15 low-density tree stands subject to exclusion, moderate, and heavy livestock pressure, we selected 12 P. australis individuals <2 m tall, tagged four new shoots per tree and measured shoot length every month during a year. At the stand and at the tree level, we analyzed monthly dynamics of growth and browsing, and the annual output in terms of total browsing and total gross and net growth (not discounting and discounting consumption, respectively). In addition, we assessed the influence of stand, tree and microsite characteristics on growth and browsing patterns. Polylepis australis fully compensated for herbivory in terms of shoot gross growth at moderate, but not at heavy livestock pressure. In terms of net growth, this species did not fully compensate for herbivory at any stocking rate. We found a strong coupling between browsing and growth along the year, suggesting that regrowth attracts browsing, and browsing promotes regrowth. At the stand level, annual gross growth was not affected by habitat characteristics, while at the tree level, annual gross growth decreased on more rocky microsites for browsed but not for unbrowsed trees. We concluded that stocking densities should be limited to allow for a reasonable annual net growth, as its nitrogen rich leaves are a valuable food resource and P. australis forests provide important ecosystem services.  相似文献

In Central Europe, salt-influenced habitats are restricted mainly to the coastlines of the North Sea and the Baltic Sea. The few natural inland salt sites suffer from size reduction and loss in biodiversity, while anthropogenic salt sites around potash mining dumps experienced recent and massive de novo colonization by numerous halophytes. Our study aimed to elucidate the general patterns of gene flow among coastal and inland salt habitats. We used amplified fragment length polymorphisms to assess the genetic diversity and genetic structure of 18 European populations of the halophyte Spergularia media (Caryophyllaceae), sampled from inland and coastal salt sites of Germany, the Netherlands, Denmark, Austria, France and Italy. Estimates of genetic diversity on the population level were generally low, especially within inland salt habitats. Analyses of molecular variance showed comparatively strong differentiation among populations. Multivariate ordination (principal coordinate analysis) and a NeighborNet analysis revealed four distinct groups of populations that showed good correspondence to their geographic origins. A Bayesian analysis performed with the program Structure as well as high pairwise Φ ST values supported the results of the phenetic analyses. The observed patterns of diversity and differentiation among inland populations of S. media are best explained by the isolated nature of suitable salt habitats, with concomitant reduction of gene flow to and among these sites. Our data support the hypothesis that the colonization of anthropogenic salt sites by S. media originated from natural inland habitats.  相似文献

Mediterranean ecosystems are traditionally influenced by human disturbance. In the mountain chains of south-eastern Spain, holm oak forests representing the potential natural vegetation are currently highly fragmented and subjected to extensive grazing by goats and sheep. This grazing activity by large herbivores has important impacts on community structure and dynamics of the remaining remnants because several processes associated with reproduction are affected. The present research analyses the variation of stand structure and regeneration of six forests dominated by Quercus ilex along a gradient of increasing grazing intensity. Stands of higher grazing intensity are characterized by a lower trunk number. Particularly, trees > 1.3 m height and diameter classes < 20 cm are clearly less represented. In addition, more intensively grazed forests show a highly aggregated dispersion of trees, which might be explained by an increase of clonally produced ramets by silvipastoral use. On the other hand, no Quercus ilex seedlings were found in any of the stands examined, although acorn production was high and did not correlate with grazing intensity. However, grazing clearly influences the quality of acorn harvest. We found a significant negative correlation between acorn length, acorn mass and grazing intensity and a positive correlation between the percentage of aborted acorns, acorn predation and herbivore activity. Thus, management of grazing in holm oak forests must take the high influence that livestock exerts on the regeneration of Quercus ilex into account, and maximum sustainable grazing intensity should be significantly lower than 0.8 animals per ha.  相似文献

Patients with relapsing-remitting multiple sclerosis (RR-MS) show a suboptimal CD4(+)CD25(+) regulatory T cell (Treg) function, whereas no Treg alterations are observed in secondary progressive MS (SP-MS) patients. To clarify the difference in Treg activity between early and chronic disease stages in MS, we analyzed the functional capacity and homeostatic parameters of naive CD4(+)CD25(+)CD127(low)CD45RA(+) Tregs (nTregs) and their memory counterparts CD4(+)CD25(+)CD127(low)CD45RO(+) Tregs (mTregs) in untreated MS patients and healthy controls. Interestingly, whereas the suppressive capacity of FACS-sorted nTregs was impaired in both early and chronic MS patients, only the latter group showed a restored mTreg function. Consistent with this observation, chronic MS patients had increased numbers of mTregs as compared with age-matched early MS patients, whereas nTreg frequencies did not differ significantly. TCR excision circle numbers were reduced in nTregs of early MS patients, suggestive of a diminished nTreg thymic output. Moreover, a decreased number of CD31(+) mTregs were observed in early vs chronic MS patients, indicating that inflammatory processes drive the homeostatic turnover of mTregs during the early disease stage. Additionally, early MS patients showed a more restricted nTreg and mTreg TCR BV gene profile as compared with healthy controls and chronic MS patients. Finally, analysis of IFN-beta and glatiramer acetate-treated MS patients showed that these immunomodulatory drugs modify nTreg homeostasis. Taken together, this study provides strong evidence for a disturbed thymic nTreg development and function in MS patients. Moreover, memory Treg but not naive Treg homeostasis recovers during disease progression.  相似文献

The enzyme spermidine/spermine N(1)-acetyltransferase (SSAT) regulates the catabolism and export of intracellular polyamines. We have previously shown that activation of polyamine catabolism by conditional overexpression of SSAT has antiproliferative consequences in LNCaP prostate carcinoma cells. Growth inhibition was causally linked to high metabolic flux arising from a compensatory increase in polyamine biosynthesis. Here we examined the in vivo consequences of SSAT overexpression in a mouse model genetically predisposed to develop prostate cancer. TRAMP (transgenic adenocarcinoma of mouse prostate) female C57BL/6 mice carrying the SV40 early genes (T/t antigens) under an androgen-driven probasin promoter were cross-bred with male C57BL/6 transgenic mice that systemically overexpress SSAT. At 30 weeks of age, the average genitourinary tract weights of TRAMP mice were approximately 4 times greater than those of TRAMP/SSAT bigenic mice, and by 36 weeks, they were approximately 12 times greater indicating sustained suppression of tumor outgrowth. Tumor progression was also affected as indicated by a reduction in the prostate histopathological scores. By immunohistochemistry, SV40 large T antigen expression in the prostate epithelium was the same in TRAMP and TRAMP/SSAT mice. Consistent with the 18-fold increase in SSAT activity in the TRAMP/SSAT bigenic mice, prostatic N(1)-acetylspermidine and putrescine pools were remarkably increased relative to TRAMP mice, while spermidine and spermine pools were minimally decreased due to a compensatory 5-7-fold increase in biosynthetic enzymes activities. The latter led to heightened metabolic flux through the polyamine pathway and an associated approximately 70% reduction in the SSAT cofactor acetyl-CoA and a approximately 40% reduction in the polyamine aminopropyl donor S-adenosylmethionine in TRAMP/SSAT compared with TRAMP prostatic tissue. In addition to elucidating the antiproliferative and metabolic consequences of SSAT overexpression in a prostate cancer model, these findings provide genetic support for the discovery and development of specific small molecule inducers of SSAT as a novel therapeutic strategy targeting prostate cancer.  相似文献

The beta-adrenergic receptor responsiveness of isolated guinea pig tracheal spirals can be negatively affected by intraperitoneal administration of the Gram-negative bacterium Haemophilus influenzae, four days prior to the experiment. The reduction in tracheal relaxation is accompanied by a decrease in beta-adrenergic receptor binding sites in splenic lymphocyte membranes and by a decrease in the fluidity of these membranes. The H. influenzae-induced dysfunction of both the respiratory airway and lymphocyte beta-adrenergic systems can be mimicked by modulating the amount of linoleic acid in the diet. This linoleic acid induced dysfunction of the beta-adrenergic system is also accompanied by a decrease in the plasma membrane fluidity of the splenic lymphocyte membranes of the guinea pigs. The role for plasma membrane fluidity in asthma is discussed in relation to current concepts for atopy.  相似文献

Experimental evidence about how generalist consumers affect exotic plant invasions is equivocal, but most tests have been limited to few plant species, single herbivore guilds, and single locations. Using a seed‐addition experiment, we studied effects of gastropods and rodents on recruitment success of 37 exotic and 37 native plant species affiliated to three different functional groups (i.e. grasses, legumes and non‐legume herbs). We replicated our seed addition x herbivore exclusion experiment at multiple grassland sites, located within a few km of each other in two regions, coastal central California (USA) and southern Saxony–Anhalt (Germany). The two study regions differed in climate, land‐use, invasion history and species pools which allowed us to disentangle general from context‐specific effects. In both regions, herbivory by gastropods had a stronger impact on the proportion of recruited seedlings and the proportion of recruited species than rodent herbivory, but this effect was much more pronounced in California than in Germany. Especially, seedling recruitment of non‐legume herbs and legumes suffered from gastropod herbivory. Contrastingly, the effect of rodents was negative at the German sites and positive at the Californian sites, likely driven by context‐specific differences in the rodent assemblages. Across both study regions, exotics had higher seedling recruitment than natives, indicating that higher recruitment success constitutes an inherent feature of exotic species. After two years, more exotic than native species established at grassland sites in California while the opposite was true for the German grassland sites. Consistently across regions, native and exotic species did, however, not differ in their response to herbivory, suggesting that generalist consumers suppress recruitment and colonization of plant species irrespective of their origin. Our results demonstrate the importance of a multi‐species, multi‐site approach to separate general responses of exotic and native plants to generalist herbivory from local, regional or species‐specific peculiarities.  相似文献

Although VP1 region 140 to 160 of foot-and-mouth disease virus (FMDV) is able to elicit neutralizing antibody in cattle, the protection against virus challenge that is conferred by peptide immunization is often poor. Here, we show that bovine T cells primed with peptides derived from this region generally show no reactivity to intact FMDV. In contrast, T-cell epitope VP4[20-34] is able to prime for a virus-specific response.  相似文献

At least six epizootics of simian hemorrhagic fever have occurred at four different primate centers. Although these diseases could easily be transmitted to other monkeys of the Macaca species, difficulty has been encountered in isolating the causative virus in cell culture. The results of this study have shown that the isolation of simian hemorrhagic fever virus strains in cell culture is dependent upon the use of a susceptible MA-104 cell strain and that the ability of such strains to support the replication of these viral agents may vary. By using this information we have been able to isolate a viral agent in cell culture from materials derived from the Sussex/69 epizootic.  相似文献

The mechanism of internalization of the vasopressin-receptor (V2-subtype) of LLC-PK1-cells, a pig renal tubular cell line, is unknown. We studied internalization utilizing a novel, highly specific vasopressin analogue ((125I)-[8-p(OH)-phenylpropionyl]-LVP, 2000 Ci/mmol). Scatchard analysis performed with membranes of LLC-PK1-cells revealed a Kd of 0.8 +/- 0.2 nM and a Bmax of 366 +/- 41 fmol/mg of protein. Degradation of the ligand was excluded by RP-HPLC-analysis. Internalization was proven by the acid-wash technique, quantitative light-microscopic autoradiography and electron microscopy. The ligand was internalized in a time- and temperature-dependent manner. At 4 degrees C, no uptake was found; at 22 degrees C, after 30 min of incubation, more than 50% of the radioligand was found inside the cell. Electron microscopy demonstrated that plasma-membrane bound vasopressin receptors are internalized by receptor-mediated endocytosis via coated pits.  相似文献
