Fatty-acid-binding-protein isolated from liver of rats receiving normal or high fat diet was studied by three different methods. The effect of high fat diet on the thermal stability of the protein was determined employing differential scanning calorimetry. Fatty acids have a stabilizing effect on the thermal stability of the protein. In order to determine the relative binding affinity of native and delipidated protein a Sephadex G-50 assay was employed using [1-14C] oleate as ligand. The delipidated protein exhibited greater binding of oleate than did the native material. Increases in the transfer of oleic acid from rat liver microsomes to egg lecithin liposomes in vitro were also observed when protein obtained from both sources were delipidated. The results suggest that high corn oil diet would modify the properties of fatty-acid-binding-protein in the uptake and cytosolic transport of long-chain fatty acids.  相似文献

The carbocyclic analogue of (E)-5-(2-bromovinyl)-2'-deoxyuridine, C-BVDU, is a very potent and selective anti-herpes-virus compound. In order to synthesize and study the properties of a DNA that contains C-BVDU, the 5'-triphosphate, C-BVDUTP was prepared and evaluated as a potential substrate of the E. coli Klenow DNA polymerase enzyme. Although C-BVDUTP proved to be a very poor substrate also of this enzyme, it could be incorporated up to 3.6% into the synthetic DNA, poly(dA-dT, C-BVDU). This level of substitution decreased significantly the template activity for DNA and RNA polymerases, as compared to that of poly(dA-dT).  相似文献

We have examined the effect of low molecular weight components of the transport mixture generally used for the import of rat liver pre-ornithine carbamoyltransferase by isolated rat liver mitochondria. These studies revealed that spermidine and spermine, at physiological concentrations, stimulate the transport of the precursor of ornithine carbamoyltransferase into mitochondria. This stimulatory effect of spermidine and spermine is concentration-dependent and is completely inhibited at higher than physiological concentrations (20 mM for spermidine and 4 mM for spermine). Magnesium ions, which also have a stimulatory effect, inhibit the stimulatory effect of spermidine.  相似文献

A scheme is proposed for generating the intact Val-448-Phe-545 polypeptide of human plasminogen which contains the fifth kringle domain of the plasmin heavy chain. The procedure is based on a pepsin fragmentation of miniplasminogen and involves the purification of the kringle 5-containing fragment by gel filtration and ion-exchange chromatography. The final product is characterized by amino acid analysis, N- and C-terminal analyses, and high-resolution 1H-NMR spectroscopy at both 300 MHz and 611 MHz. We detect a (40:60%) Asp/Asn heterogeneity at site 452 of the Glu-plasminogen molecule. In the conventional kringle numbering system, the kringle 5 domain extends from Cys-1 to Cys-80, which corresponds to Cys-461 to Cys-540 in plasminogen. A preliminary 1H-NMR characterization of kringle 5 focuses on the global conformational features of the polypeptide. Assignments are given for a number of resonances, including the Tyr-72, the His imidazoles' and the Trp indoles' spin systems. Comparison with human plasminogen kringles 1 and 4 shows that the kringle 5 conformation is highly structured and very similar to that of the homologous domains. This conservancy is particularly striking in the environment surrounding Leu-46 and in the overall features of the aromatic spectrum. There are some differences, particularly in the buried His-33 imidazole group, whose H2 resonance is shifted to 9.67 ppm. A preliminary study of benzamidine-binding shows that the ligand interacts weakly (Ka approximately equal to 1.7 mM -1) mainly through the amidino functional group. Trp-62 and Tyr-72 are significantly perturbed by benzamidine, suggesting that these residues are part of the ligand-binding site.  相似文献

In vitro culture was established from shoot tips ofDigitalis lanata cotyledonous plants. The propagated plant material was rooted, transplanted into soil and grown under field conditions. Lanatoside C content was determined in a total of 20 clones and statistically evaluated by means of variance analysis of unequal-sized samples.In vitro clonal propagation ofD. lanata was found not to affect lanatoside C content. Five morphological parameters of sister chromatids, viz radius, length, centromeric index, volume and centromere width, were studied by electron microscopy, from 140 human number 1 chromosomes, and 122 human number 2 chromosomes. The degree of variation obtained ranged from 0.277 +/- 0.003% and 0.286 +/- 0.005% for the centromeric index of chromosomes 1 and 2, respectively, to 9.835 +/- 0.933% and 13.472 +/- 1.461% for centromere width.  相似文献

Insulin treatment induced in Tetrahymena pyriformis a positive hormonal imprinting, and in Tetrahymena thermophila a negative imprinting, resulting in increased and decreased binding capacity, respectively, at re-exposure to the hormone. The imprinting, or the information associated with it, is transferred by the nutrient medium of the insulin-treated cells to those not treated. The issue of transfer depends on the nature of the receiver taxon, leading always to a positive imprinting in Tetrahymena pyriformis, and to a negative imprinting in Tetrahymena thermophila, regardless of the nature of the 'imprinted' transmitter taxon. The findings substantiate the transferability of hormonal imprinting by the nutrient medium at the unicellular level, the key role of the postreceptorial mechanism in determining the trend of imprinting and may explain the persistence of imprinting in the progeny generations.  相似文献

Leukocyte-derived inhibitory activity inhibiting the entry of normal progenitor cells of granulocytes and macrophages (CFU-GM) into the S-phase of a cell cycle was investigated in 16 patients with different forms of myelodysplastic syndrome (MDS). The presence of this inhibitory activity was analysed in medium conditioned with low-density cells obtained from peripheral blood of MDS patients. The inhibition rate was measured by 3H-thymidine suicide technique with subsequent cultivation of pretreated cells in semisolid agar medium. Low-density cells from MDS patients of various types were studied: from the twelve patients with refractory anaemia (RA or RAS) only three were positive, one patient with chronic myelomonocytic leukaemia (CMML) was negative while one patient with refractory anaemia with excess of blasts (RAEB) and two patients with RAEB in transformation (RAEB-T) were positive with respect of the described test. In two patients with RA, who underwent a long-term investigation, the production of leukocyte-derived inhibitory activity preceded the development of disease into RAEB or RAEB-T. In five positive cases, supernatants were incubated with antiserum against human placental ferritin; with one exception, the inhibitory activity was neutralized.  相似文献

Insulin gives rise to positive imprinting in Tetrahymena pyriformis, but to negative imprinting in T. thermophila, as revealed by the respective increases and decreases in the insulin-binding capacity of these organisms observed during later interactions with this hormone. We found that changes in insulin-binding capacity exhibited parallelism with fluctuations of the levels of free, intracellular Ca2+ detectable by Quin-2 labeling. An exception was the second interaction of T. thermophila with insulin, which although showing a positive trend, produced a relatively small increase in the level of intracellular Ca2+. These observations suggest an interrelationship between hormone-binding capacity and the fluctuation of intracellular Ca2+ levels. Either hormone binding depends on the availability of Ca2+, or, alternatively, the latter depends on the binding capacity. Further studies are required to elucidate the true nature of this interdependence.  相似文献
