Enzyme kinetics of a highly purified mitochondrial creatine kinase in comparison with cytosolic forms

Summary Mitochondrial creatine kinase (CK) purified from canine myocardium showed a single protein band on SDS-PAGE and was free of MMCK. Its amino acid composition was different than MMCK or BBCK and did not react to antiserum to MMCK or BBCK. Using purified mitochondrial, MM and BBCK, the velocity of reaction (V) was estimated for creatine phosphate (CP), creatine (C), adenosine triphosphate (ATP) and adenosine diphosphate (ADP) over a wide range of concentrations including those at V_max. The values for K_m (mM/L) derived from Lineweaver-Burke plots are shown: The affinity of mitochondrial CK for C is much greater than MMCK which is compatible with the energy shuttle hypothesis, namely ATP is converted by mitochondrial CK to CP, and then diffuses to the myofibril for conversion to ATP for utilization.     

The Fertility of Melanesian Girls, Laboratory Mice, and Prostitutes: A Comment on the "Bruce Effect"

Greenfield has recently proposed that low fertility in both prostitutes and unmarried Trobriand girls may be explained by the "Bruce effect": the inhibition of pregnancy in female mice that have had intercourse with different males within a short period. An examination of Malinowski's data indicates that a Trobriand girl usually had intercourse with only one man over a fairly long period and her sex life was not at all comparable to that of a prostitute or one of Bruce's mice. Low fertility in unmarried girls has been well documented for several Melanesian societies that differ notably in the behavior permitted to girls. The existence of so-called adolescent sterility still seems to be the best explanation to cover all of these cases. [fertility, sexual behavior, Melanesia]     

A Survey of Infantile Gastroenteritis

In 1967 we admitted 339 cases of infantile gastroenteritis; one-third of these were dehydrated, and in this group the commonest biochemical abnormality found was hypernatraemia, sometimes with metabolic acidosis. A higher incidence of dehydration was found in the patients who had received oral glucose fluids before admission. Enteropathic Escherichia coli were isolated from the faeces of 16% of the cases. Associated infections, especially of the respiratory tract, were common. Treatment was aimed at the restoration of fluid and electrolyte balance. Usually this was achieved with oral fluids, though intravenous fluids were used in the most severely dehydrated cases. Recovery was complete in 320 cases and a further 14 cases were discharged as carriers of enteropathic E. coli. There were five deaths (1·5%) in the series; three occurred immediately after admission.     

Electron Microscope Study of Lens Fibers

Comparative electron microscope investigations on sections of the lens cortex of the normal, mature rat, rabbit, monkey, and the normal calf reveal similar patterns of intracellular organization. The superficial lens fiber contains a nucleus, mitochondria, endoplasmic reticulum, dense granules, Golgi complex, and a quantity of small structures of low opacity which appear as filamentous and spherical configurations. Variations in number, distribution, and spatial arrangement of cytoplasmic elements in lens fibers are described. These changes in the pattern of cytoplasmic organization are concomitant with development of fibers and their displacement towards the center of the lens. Structural details of the various zones of the lens epithelium and the lens fibers are compared.     

Primary structure of rabbit skeletal muscle glycogen synthase deduced from cDNA clones

The complete amino acid sequence of rabbit skeletal muscle glycogen synthase was deduced from cDNA clones with a composite length of 3317 bp. An mRNA of 3.6 kb was identified by Northern blot analysis of rabbit skeletal muscle RNA. The mRNA coded for a protein of 734 residues with a molecular weight of 83,480. The deduced NH2-terminal and COOH-terminal sequences corresponded to those reported for the purified protein, indicating the absence of any proteolytic processing. At the nucleotide level, the 5' untranslated and coding regions were 79 and 90% identical for rabbit and human muscle glycogen synthases, whereas the 3' untranslated regions were significantly less similar. The enzymes had 97% amino acid sequence identity. Interestingly, the NH2 and COOH termini of rabbit and human muscle glycogen synthase, the regions of phosphorylation, showed the greatest sequence variation (15 of 19 mismatches and two insertion/deletion events), which may indicate different evolutionary constraints in the regulatory and catalytic regions of the molecule.     

Embodying illness,embodying cancer

Individuals and societies embody illnesses in different ways, in part determined by the way a person knows and lives his or her diagnosis and prognosis. Based on research in Northern Italy, on the experiences and meanings of cancer and on the practice of nondisclosure of the diagnosis, we find nondisclosure reflects a world divided - life/death, good/bad, mind/body — with the unwanted converted to other. The strong association of cancer with death, suffering, and hopelessness in much of Italy, coupled with the tremendous power attributed to naming and sentencing makes nondisclosure a major mechanism for keeping the condemned in this social world, and keeping death, decay, and suffering in the other. It is the social reality that is dominant here, such that informing a patient of cancer can be tantamount to social death.

Résumé Les individus et les sociétés incorporent la maladie de façon différente, déterminée en partie de comment une personne connait et vit son diagnostic et prognostic. A partir de la recherche des experiences des significations du cancer et de la pratique de ne pas dire la diagnostic au Nord de l'Italie, on a remarqué que l'habitude de ne rien dire reflète un monde séparé entre la vie et la mort, entre le bon et le mal, entre l'esprit et le corps, de sorte que ce qui West pas voulu soit transfomé en l'autre. L'association forte du cancer à la mort, à la souffrance, au désespérance en toute l'Italie, unie au grand pouvoir donne au fait de dénommer et de donner une sentence, rend ne pas dire un méchanisme important pour garder le condamnd dans ce monde social et pour garder la mort, la décadence et la souffrance dans l'autre. C'est la réalité sociale qui est ici dominante, tel que le fait d'informer un patient de cancer soit comme une mort sociale.

     

A new major histocompatibility complex class I b gene expressed in the mouse blastocyst and placenta

 Because of the role major histocompatibility complex (MHC) class I b molecules may play during mouse embryonic development, we thought it would be interesting to search for additional MHC class I b molecules that might be expressed in preimplantation embryos, and in particular in the trophoblastic lineage. We therefore screened a mouse preimplantation blastocyst cDNA library for MHC class I sequences. This search led to the identification and characterization of a new MHC class I b gene, blastocyst MHC. Sequences identical to the exons and 3′ untranslated region of this gene have been found in many laboratory mouse strains, as well as in the related mouse species Mus spreciligus. The presence of this gene in mouse strains of different MHC class I haplotypes argues that blastocyst MHC is a unique, newly-described gene rather than a new allele of a previously described mouse MHC class I gene. Blastocyst MHC has the structure of an MHC class I b gene, with the six exons characteristic of T-region genes. It is linked to H2-D. The amino acid sequence encoded by this gene maintains all the features of a functional antigen-presentation domain. The blastocyst MHC gene, like the human class I b gene HLA-G, is expressed at the blastocyst stage and in the placenta, and may be the mouse analog for HLA-G. Changes to pharmacare programs in British Columbia and Ontario were compared during a recent Toronto conference on the business aspects of Canadian health care. The brunt of the changes in BC are being borne by pharmaceutical companies and prescribers, while in Ontario the new program affects those who receive free drugs from the province and People and groups that rely on private health insurance.
