Microtubule assembly and disassembly at alkaline pH   总被引:9,自引:4,他引:5       下载免费PDF全文
Although it is now apparent that the intracellular pH may rise considerably above neutrality under physiological conditions, information on the effect of alkaline pH on microtubule assembly and disassembly is still quite fragmentay. We have studied the assembly/disassembly of bovine brain microtubule protein at alkaline pH in vitro. When microtubules are assembled to a new steady state at pH less than 7 and pH is then made more alkaline, they undergo a rapid disassembly to a new steady state. This disassembly is reversed by acidification. The degree of disassembly is determined largely by the pH- dependence of the critical concentration, which increases five to eight times, from pH 7 to 8. A fraction of assembly-incompetent tubulin is identified that increases with pH, but its incompetency is largely reversed with acidification. Measurements of microtubule lengths are used to indicate that disassembly occurs by uniform shortening of microtubules. A comparison of shortening by alkalinization with dilution suggests that the intrinsic rate of disassembly is accelerated by increasing pH. The capacity for initiating assembly is progressively lost with incubation at alkaline pH (although some protection is afforded by sulfhydryl-reducing agents). However, direct assembly from depolymerized mixtures is possible at least up to pH 8.3, and the steady state achieved at these alkaline pH values is stable. Such preparations are readily disassembled by cold and podophyllotoxin (PLN). Disassembly induced by PLN is also markedly enhanced at alkaline pH, suggesting a corresponding enhancement of “treadmilling.” The implications of physiological events leading to alkaline shifts of pH for microtubule assembly/disassembly are discussed, particularly in the light of recent hypotheses regarding treadmilling and its role in controlling the distribution of microtubules in vivo.  相似文献

Plagiochila sect. Vagae is a large pantropical clade that is characterized morphologically by frequent terminal branching, vegetative distribution by propagules on the ventral surface of the leaves and a capsule wall with thickenings in all layers. Plagiochila corrugata from Brazil is characterized by strongly undulate, toothed leaf margins and represents the only known neotropical species of sect. Vagae with unispiral elaters. Plagiochila cambuena from Madagascar is distinguished by the same features. Maximum likelihood and parsimony analyses of 38 nrDNA ITS sequences of Plagiochila reveal P. corrugata and P. cambuena in a weakly (ML) to well (MP) supported monophyletic lineage within P.  sect.  Vagae . As an outcome of the morphological and molecular investigation, P. cambuena is relegated to the synonymy of P. corrugata. Plagiochila corrugata is placed in a Vagae -subclade with 11 further American species. The range of P. corrugata can be ascribed to long-range dispersal from the Neotropics rather than a Gondwanan distribution. Species from tropical Asia and Africa are placed at the base of the Vagae clade. An increasing number of studies demonstrate that plant and animal phenologies such as the timing of bird migration have been advancing over the globe, likely as a result of climate change. Even closely related species differ in their phenological responses, and the sources of this variation are poorly established. We used a large, standardized dataset of first arrival dates (FAD) of migratory birds to test the effects of phylogenetic relationships and various life-history and ecological traits on the degree to which different species adapt to climate change by earlier migration in spring. Using the phylogenetic comparative method, we found that the advancement of FAD was greater in species with more generalized diet, shorter migration distance, more broods per year, and less extensive prebreeding molt. In turn, we found little evidence that FAD trends were influenced by competition for mating (polygamy or extra-pair paternity) and breeding opportunities (cavity nests). Our findings were robust to several potentially confounding effects. These evolutionary correlations, coupled with the low levels of phylogenetic dependence we found, indicate that avian migration phenology adapts to climate change as a species-specific response. Our results suggest that the degree of this response is fundamentally shaped by constraints and selection pressures of the species' life history, and less so by the intensity of sexual selection.  相似文献

Background: A great number of in vitro and in vivo studies have suggested that many pathways or factors can stimulate angiogenesis and lymphangiogenesis, which facilitate tumor progression and metastasis. However, the morphological and immunohistochemical profile of newly formed vasculatures has not been elucidated, making it difficult to differentiate them from the pre-existing ones, and to identify their unique molecular profiles for diagnosis and therapeutic interventions.Experimental findings: As cytokeratin (CK)-19 is a well-recognized stem cell marker and CK-19-positive cells are frequently detected in the peripheral blood of patients with metastatic cancer, our recent studies have assessed the involvement of CK-19 in the formation of new vasculatures in primary colorectal cancer (CRC) tissues. Our studies showed that a subset of lymph node-positive cases harbored some isolated normal epithelial structures with distinct CK-19 immunostaining within an otherwise CK-19-negative background. These structures are exclusively located within or adjacent to lymphoid follicles and are often surrounded by tube-like structures expressing lymphatic endothelial marker D2-40. Similar structures are more frequently seen at the junctions between pre-invasive and invasive CRC with the following features: (1). they consist of a single layer of endothelial cells that express both D2-40 and CD34, (2). their endothelial walls are often incomplete with disseminated cells protruding into the adjacent stroma, and (3). they are exclusively associated with disseminated CK-19-positive cellsHypothesis: Based on these findings, we propose that these tube-like structures represent newly formed vasculatures, which are derived by the convergence of aberrant lymphocyte infiltration and tumor stem cells. Because of their close physical proximity, tumor stem cells within the epithelial and stromal components contribute equally and coordinately to the morphogenesis of new vasculatures, which constitutes the basis for the unique morphologic and immunohistochemical features of newly formed vasculatures. Our hypothesis appears to be applicable to all epithelium-derived cancers.  相似文献

Coyotes (Canis latrans) may affect adult and neonate white-tailed deer (Odocoileus virginianus) survival and have been implicated as a contributor to the decline of deer populations. Additionally, coyote diet composition is influenced by prey availability, season, and region. Because coyote movement and diet vary by region, local data are important to understand coyote population dynamics and their impact on prey species. In southeast Minnesota, we investigated the effect of coyotes on white-tailed deer populations by documenting movement rates, distances moved, and habitats searched by coyotes during fawning and nonfawning periods. Additionally, we determined survival, cause-specific mortality, and seasonal diet composition of coyotes. From 2001 to 2003, we captured and radiocollared 30 coyotes. Per-hour rate of movement averaged 0.87 km and was greater (P = 0.046) during the fawning (1.07 km) than the nonfawning period (0.80 km); areas searched were similar (P = 0.175) between seasons. Coyote habitat use differed during both seasons; habitats were not used in proportion to their availability (P < 0.001). Croplands were used more (P < 0.001) than their proportional availability during both seasons. Use of grasslands was greater during the fawning period (P = 0.030), whereas use of cropland was greater in the nonfawning period (P < 0.001). We collected 66 fecal samples during the nonfawning period; coyote diets were primarily composed of Microtus spp. (65.2%), and consumption of deer was 9.1%. During the study, 19 coyotes died; annual survival rate range was 0.33–0.41, which was low compared with other studies. Consumption of deer was low and coyotes searched open areas (i.e., cropland) more than fawning areas with dense cover. These factors in addition to high coyote mortality suggested that coyote predation was not likely limiting white-tailed deer populations in southeast Minnesota. © 2011 The Wildlife Society.  相似文献

Feral pigs (Sus scrofa) survive in many climates, reproduce year-round, and are dietary generalists. In the United States, the size and range of the feral pig population has expanded, resulting in greater interaction with humans and domestic swine and increased potential for disease transmission. We conducted a serosurvey in feral pigs from eastern North Carolina to determine exposure to the zoonotic parasites, Toxoplasma gondii and Trichinella spp. Between September 2007 and March 2009, blood serum was collected from 83 feral pigs harvested at Howell Woods Environmental Learning Center, Four Oaks, North Carolina, USA. We used a modified agglutination test to test for T. gondii antibodies and an enzyme-linked immunosorbent assay to test for Trichinella spp. antibodies. The prevalences of antibodies to T. gondii and Trichinella spp. were 27.7% and 13.3%, respectively and 4% (n=3) had antibodies to both agents. We detected an increased risk of T. gondii antibodies with age, whereas the risk of exposure to T. gondii across years and between sexes was similar. In eastern North Carolina, feral pigs have been exposed to T. gondii and Trichinella spp. and may pose a health risk to domestic swine and humans.  相似文献

Previously we described lectin-like molecules in the ciliate Tetrahymena pyriformis; by application of synthetic neoglycoconjugates it is now shown that T. pyriformis contains considerable amounts of both a β-d-glucose- and a lactose-specific lectin. No evidence for the presence of α-d-mannose-, α-d-galactose- or of α-l-fucose-specific lectins could be obtained. The two lectins, identified in T. pyriformis, are associated with the kinetids. During cell division the lectins disappear or become masked in the fission furrow. Therefore, we assume that these lectins are involved in the organization of the distribution pattern of the kinetids during cell division perhaps due to lectin—glycoprotein interactions.  相似文献

Lizards in seleroglossan families of actively foraging carnivores and the herbivorous iguanids use the tongue to gather chemical samples to detect, identify, and locate food prior to attack, and to relocate lost food. In contrast, previously studied iguanian families other than Iguanidae lack lingually mediated prey chemical discrimination (PCD) and do not exhibit lingually mediated chemosensory searching behavior for bitten prey that has escaped or been lost (SICS = strike-induced chemosensory searching). In the present study, experimental tests showed that PCD and SICS are absent in Crotaphytus collaris, a member of the previously unstudied family of iguanian ambush foragers, Crotaphytidae. Available data suggest that in active foragers natural selection favors use of chemical cues to locate hidden prey, whereas in ambush foragers natural selection favors immobility to avoid detection by predators and SICS precludes simultaneous ambush. In most families the states of PCD and SICS are retained from the ancestors, but when foraging mode shifts, a change in chemosensory behavior appears to be induced. The inhibitory effect of 23N-alkyl-4-piperidylesters (alkyl = ethyl-butyl) (APEA) and 8N-ethyl-2-pyrrolidinylmethylesters (EPMEA) of 2- and 3-substituted alkoxyphenylcarbamic acids (alkoxy = butoxy-heptyloxy-) on photosynthetic Hill reaction activity of spinach chloroplasts and on chlorophyll (Chl) synthesis in green algaeChlorella vulgaris was investigated. Inhibitory activities of these compounds were strongly connected with the lipophilicity of the whole molecule. A lower inhibitory activity of 2-alkoxy-substituted derivatives in relation to the corresponding 3-substituted ones was confirmed. Electron spin resonance (ESR) spectra of spinach chloroplasts demonstrated that the studied compounds affected the structure of photosystem (PS) 2 with the release of Mn2+ ions into interior of thylakoid membranes.  相似文献
