A high amount of intravarietal variation in satellites and C-banded chromosomes was observed in the hexaploid wheatgrass synthetic cultivar 'Greenleaf' (Agropyron intermedium ssp. trichophorum (Link) A. &Gr., 2n = 6x = 42, genome E1E1E2E2SS). The cultivar is an open-pollinated perennial that shows extensive interplant polymorphism for many biological characters. Maximum number of satellites detected varied among plants from zero to six. In 61% of the plants, we observed two large satellites in association with zero, one, or two small ones. Chromosome constitution differed significantly among plants as revealed by analysis of variance based on the total number of banded chromosomes and the number of banded chromosomes with telomeric bands at either one or both ends. Heteromorphism in C-banding patterns between homologues was found in most of the chromosomes and was classified into four types: (i) difference in band size, (ii) difference in presence/absence of one or two bands, (iii) completely different banding patterns, and (iv) banded versus unbanded. Homologous chromosomes having types iii and iv heteromorphism could only be matched by their relative length and arm ratio instead of C-banding patterns. Deletions were detected in two chromosomes. Overall, C-banded chromosomes of this cultivar were characterized by the presence of large telomeric bands and were quite different from the previously reported karyotypes of the supposed diploid ancestor Agropyron elongatum (Host) P. Beauv. (genome EE) and an Ag. intermedium (Host) P. Beauv. accession (E1E1E2E2SS) The results suggest that dramatic chromosome modifications have occurred in this species during the course of evolution. The study sheds light on the extent of intrapopulation polymorphism present in the karyotypes of outcrossing polyploids and synthetic cultivars and has implications regarding strategies for chromosomal manipulation involving open-pollinated species.  相似文献

The effects of progesterone, 20α-hydroxy-pregn-4-en-3-one, (20α-OH-P) 17 α hydroxyprogesterone (17α-OH-P) and 5α-pregnan-3, 20-dione (5α-P) on endometrium of ovariectomized rabbits have been examined. Progesterone, at 5 mg/kg/day and at 1.7 mg/kg/day increased the number of mitotic figures observed in luminal and glandular epithelium after 5 days of treatment and induced considerable arborization. None of the other compounds induced significant arborization and only 10 mg/kg/day of 20α-OH-P increased the mitotic index in luminal and glandular epithelium. Administrations of 20α-OH-P, 17α-OH-P or 5α-P resulted in the appearance of very large, pale cells in the luminal epithelium which were not present in progesterone treated rabbits. Cytochemical techniques revealed no specific staining in these cells for polysaccharides (PAS, Alcian blue), lipid (Oil Red O) or for blastokinin, a progesterone-inducible, uterine protein, examined by the fluoroscein isothiocyanate labeled antibody technique. The possibility of an apparent dissociation between histodifferentiation and cytodifferentiation is discussed.  相似文献

BrdU and BrdC have been employed as DNA labeling agents for differentiation of sister chromatids and for extension of sister chromatid exchange (SCE) methods to regenerating murine liver cells in vivo. Comparisons were made between bone marrow and liver cells isolated simultaneously from mice following DNA labeling with either BrdC or BrdU. Although the total mitotic yield of bone marrow cells was considerably greater than in liver, a higher percentage of second division metaphases was observed in liver cell preparations. The percentages of second division c-metaphase cells observed were 31.5% in bone marrow and 73% in liver cell preparations. Utilizing either BrdU or BrdC, no significant difference in percentage of second division metaphases was discerned. The number of spontaneous SCEs per cell was distributed according to the Poisson probability function. No significant differences in mean numbers of SCEs per cell were found in comparisons of bone marrow (1.40) and liver cells (1.65) or of cells which had incorporated BrdU or BrdC.  相似文献

Effects of pH on proton transport by vacuolar pumps from maize roots   总被引:1,自引:0,他引:1  
Protons pumps of the tonoplast may be involved in the regulation of cytosolic pH, but the effects of pH on the coupled activities of these transporters are poorly understood. The effects of pH on the activities of the H+-translocating pyrophosphatase (PPiase) and vacuolar-type H+-translocating adenosine triphosphatase (H+-ATPase) from maize ( Zea mays L. cv. FRB 73) root membranes were assessed by model that simultaneously considers proton transport by the pump and those processes that reduce net transport. The addition of either pyrophosphate or ATP to either microsomal or tonoplast membranes generated a pH gradient. The pH gradient generated in the presence of both substrates was not the sum of the gradients produced by the two substrates added separately. When membranes were separated by sucrose density gradient centrifugation, pyrophosphate (PPi)-dependent proton transport was associated with light density membranes having tonoplast H+-ATPase activity. These results indicate that some portion of the PPiase was located on the same membrane system as the tonoplast ATPase; however, tonoplast vesicles may be heterogeneous, differing slightly in the ratio of ATP- to PPi-dependent transport. Proton transport by both the PPiase and ATPase had maximal activity at pH 7.0 to 8.0 Decreases in proton transport by the ATPase at pH above the optimum were associated with increases in the processes that reduce net transport. Such an association was not observed at pH values below the optimum. These results are discussed in terms of in situ regulation of cytoplasmic pH by the two pumps.  相似文献

This paper analyzes nonmetric trait variation in 11 late Late Woodland (ca. AD 700-1000) and one Mississippian (AD 1000-1300) skeletal samples from west-central Illinois from a population-structure perspective. Most of the sites are of the Bluff phase of Late Woodland in the lower Illinois River valley; others are from a nearby, contemporary archaeological phase. Late Woodland as a whole era (ca. AD 250-1000) was a period of marked population growth and expansion into new regional environments, trends that accompanied horticultural intensification in the area. Overall variation between sites was low, but males, females, and the total sample exhibited a significant geographic component to variation due to interregional morphological differences. The Bluff sites tended to group together relative to the non-Bluff sites. However, there was no significant geographic component to variation among the Bluff sites. The results are only partially consistent with archaeological data suggesting population growth and expansion through fissioning. Previous studies have demonstrated significant heterogeneity for nonmetric trait frequencies among Middle Woodland (ca. 100 BC to AD 250) sites, suggesting a Middle to Late Woodland change in population structure that lowered levels of morphological variation. This supports a model of increased intra- and interregional interaction from Middle to Late Woodland times developed from ceramic data by Braun and by Braun and Plog.  相似文献

Cortical granule exocytosis is important for the block to polyspermy at fertilization in the eggs of most vertebrates and many invertebrates. Cortical granules are poised at the cell surface and exocytose in response to sperm stimulation. Following exocytosis, the cortical granule contents modify the extracellular environment of the egg, the major result of which is to block additional sperm binding. Here we show that proteins homologous to members of the SNARE hypothesis—a molecular model designed to explain the trafficking, docking, and exocytosis of vesicles in the secretory compartment—are present in eggs at the right time and place to be involved in the regulation of cortical granule exocytosis. Using polymerase chain reaction (PCR) screens we have found homologues of synaptobrevin/VAMP, syntaxin, synaptotagmin, and rab3. Antibodies generated to fusion proteins or to synthetic peptides encoded by the cloned cDNAs were used in an immunofluorescence assay to show that each of the cognate proteins are present in the cortex of the egg. A synaptobrevin/VAMP homologue appears to be specifically associated with the membrane of cortical granules before fertilization and, following cortical granule exocytosis, is incorporated into the plasma membrane of the zygote. A rab3 homologue is also associated with cortical granules specifically but, following fertilization, the protein reassociates with different, yet undefined, vesicles throughout the cytoplasm of the zygote. Homologues of synaptotagmin and syntaxin are also present at the egg cortex but, in contrast to rab3 and VAMP, appear to be associated with the plasma membrane. Following fertilization, syntaxin and tagmin remain associated with the plasma membrane and are more readily immunolabeled, presumably due to an increased accessibility of the antibodies to the target protein domains. We also show by immunoblotting experiments that the cognate proteins are of the sizes predicted for these homologues. These results suggest that at least some steps in the biology of cortical granules may be mediated by SNARE homologues, and this finding, along with the unique biology of cortical granules, should facilitate examination of specific events of the fertilization reaction and the mechanism of stimulus-dependent exocytosis.  As global climate changes, sea level rise and increased frequency of hurricanes will expose coastal forests to increased flooding and salinity. Quercus species are frequently dominant in these forest, yet little is known about their salinity tolerance, especially in combination with flooding. In this study, 1-year-old seedlings of Quercus lyrata Walt. (overcup oak), Q. michauxii Nutt. (swamp chestnut oak), Q. nigra L. (water oak), and Q. nuttallii Palmer (Nuttall oak) were chronically (simulating sea level rise) and acutely (simulating hurricane storm surge) exposed to increased flooding and salinity, individually and in combination. The four species demonstrated two response patterns of photosynthesis (A), conductance, and leaf water potential, apparently related to their relative flood tolerance. In Q. lyrata, Q. nuttallii, and Q. nigra (moderately flood-tolerant), A was not immediately reduced after the initiation of the freshwater flooding, but was reduced as the duration of flooding increased. In the second pattern, demonstrated by the weakly flood-tolerant Q. michauxii, A was immediately reduced by freshwater flooding with an increasing impact over time. Watering with 2 parts per thousand (ppt) saline water did not consistently reduce A, but flooding with 2 ppt reduced A of all species, similar to the response with freshwater flooding. Photosynthesis of all species was reduced by 6 ppt watering or flooding, with the latter treatment killing all species within 8 weeks. When acutely exposed to 30 ppt salinity, A was quickly and severely reduced regardless of whether the seedlings were watered or flooded. Acutely flooded seedlings exposed to high salinity died within 2 weeks, but seedlings watered with 30 ppt saline water recovered and A was not reduced the following spring. As saline flooding of coastal areas increases due to sea level rise, photosynthesis of these species will be differentially affected based primarily on their flood tolerance. This suggests that increased flooding associated with sea level rise will impact these tree species to a greater extent than small increases in soil salinity. Historically, CGRP receptors have been classified as CGRP(1) or CGRP(2) subtypes, chiefly depending on their affinity for the antagonist CGRP(8-37). It has been shown that the complex between calcitonin receptor-like receptor (CRLR or CL) and receptor activity modifying protein (RAMP) 1 provides a molecular correlate for the CGRP(1) receptor; however, this does not explain the range of affinities seen for CGRP(8-37) in isolated tissues. It is suggested that these may largely be explained by a combination of methodological factors and CGRP-responsive receptors generated by CL and RAMP2 or RAMP3 and complexes of RAMPs with the calcitonin receptor.  相似文献
