为分析2013~2014流感监测年度中国大陆地区H3N2亚型流感病毒的抗原性和基因变异情况,本文选择了本监测年度中国分离的H3N2亚型流感病毒,利用标准雪貂抗血清进行抗原性分析,利用Sanger测序法进行病毒基因测序,采用邻位相临法(Neighbor-Joining,N-J)方法进行种系进化分析,分析我国H3N2亚型流感病毒的变异情况,进一步比较其与疫苗株的匹配性。抗原分析显示,本监测年度H3N2亚型流感病毒大部分为疫苗株A/Victoria/361/2011细胞株的类似株(99.6%),但以A/Texas/50/2012鸡胚株为参考抗原,只有15.1%为类似株,仅有11.9%与中国流行株的鸡胚分离株A/Shanghai-Changning/1507/2012抗原性类似。HA基因特性分析显示我国毒株均位于同一大分支,NA基因没有发现与耐药性相关的氨基酸位点突变。总之,2013~2014流感监测年度我国H3N2亚型流感病毒在流行过程中未发生明显变异,但病毒在鸡胚中传代会导致关键氨基酸位点变异。应及时分析并发现我国病毒的抗原性和基因特性变异情况,推选出更适合我国的疫苗株。  相似文献

The effects of heme on the synthesis of subunits V and VII of cytochrome c oxidase were examined in the heme-deficient yeast mutant, GL1. In vitro translation and immunodetection with subunit-specific antisera indicated a 50% decrease in both proteins, with RNA obtained from hemeless cells. Unsupplemented, pulse-labeled cells contained both V and VII polypeptides, but at extremely low levels as compared with those found in delta-aminolevulinic acid-supplemented cells. The data suggest that heme controls the formation of mRNAs for the two subunits, and may also have a regulatory role in translation and in the stability of the polypeptides.  相似文献

When performing a skill such as throwing a dart, many different combinations of joint motions suffice to hit the target. The motor system adapts rapidly to reduce bias in the desired outcome (i.e., the first-order moment of the error); however, the essence of skill is to produce movements with less variability (i.e., to reduce the second-order moment). It is easy to see how feedback about success or failure could sculpt performance to achieve this aim. However, it is unclear whether the dimensions responsible for success or failure need to be known explicitly by the subjects, or whether learning can proceed without explicit awareness of the movement parameters that need to change. Here, we designed a redundant, two-dimensional reaching task in which we could selectively manipulate task success and the variability of action outcomes, whilst also manipulating awareness of the dimension along which performance could be improved. Variability was manipulated either by amplifying natural errors, leaving the correlation between the executed movement and the visual feedback intact, or by adding extrinsic noise, decorrelating movement and feedback. We found that explicit, binary, feedback about success or failure was only sufficient for learning when participants were aware of the dimension along which motor behavior had to change. Without such awareness, learning was only present when extrinsic noise was added to the feedback, but not when task success or variability was manipulated in isolation; learning was also much slower. Our results highlight the importance of conscious awareness of the relevant dimension during motor learning, and suggest that higher-order moments of outcome signals are likely to play a significant role in skill learning in complex tasks.  相似文献

Immunofluorescence has been used to study visceral organs from a case of subacute sclerosing panencephalitis. Immune complexes were shown as granular deposits of IgG, complement, and measles antigens in renal glomeruli. Measles antigens were detected in the spleen, liver, and lymph nodes from many parts of the body.Immune-complex formation may be important in the aetiology of this disease and perhaps in causing some of its tissue damage. The rarity of subacute sclerosing panencephalitis may be due to an unusual pattern of immunological reactivity required in a patient before a measles infection can produce a subacute encephalitis.  相似文献

The little fire ant, Wasmannia auropunctata, probably arrived in Israel in ca. 1998 and was identified in 2005; this is the first record of this species from open areas outside the tropics and subtropics. It survives harsher conditions than in its native habitats, with minimal annual temperatures as low as 6°C, and 5–12 consecutive rainless months (under 15 mm rainfall per month). It is now known from 26 localities in Israel, mostly in irrigated gardens. As in other regions where they have invaded, these ants pose a serious threat to local biodiversity. At high densities they displaced almost all the local ant species sampled, affecting population abundances, species richness, and community structure. W. auropunctata seems to have a detrimental effect also on other ground arthropods, judging from the observed decline in spider and beetle abundances. We show here that this tropical species can pose a critical threat to local arthropods at a wider range of climatic conditions than was previously known.  相似文献

Aims/Hypothesis

We aimed to understand early alterations in kinin-mediated migration of circulating angio-supportive cells and dysfunction of kinin-sensitive cells in type-1 diabetic (T1D) patients before the onset of cardiovascular disease.

Methods

Total mononuclear cells (MNC) were isolated from peripheral blood of 28 T1D patients free from cardiovascular complications except mild background retinopathy (age: 34.8±1.6 years, HbA1C: 7.9±0.2%) and 28 age- and sex-matched non-diabetic controls (H). We tested expression of kinin receptors by flow cytometry and migratory capacity of circulating monocytes and progenitor cells towards bradykinin (BK) in transwell migration assays. MNC migrating towards BK (BKmig) were assessed for capacity to support endothelial cell function in a matrigel assay, as well as generation of nitric oxide (NO) and superoxide (O2 *) by using the fluorescent probes diaminofluorescein and dihydroethidium.

Results

CD14hiCD16neg, CD14hiCD16pos and CD14loCD16pos monocytes and circulating CD34pos progenitor cells did not differ between T1D and H subjects in their kinin receptor expression and migration towards BK. T1D BKmig failed to generate NO upon BK stimulation and supported endothelial cell network formation less efficiently than H BKmig. In contrast, O2 * production was similar between groups. High glucose disturbed BK-induced NO generation by MNC-derived cultured angiogenic cells.

Conclusions/Interpretation

Our data point out alterations in kinin-mediated functions of circulating MNC from T1D patients, occurring before manifest macrovascular damage or progressed microvascular disease. Functional defects of MNC recruited to the vessel wall might compromise endothelial maintenance, initially without actively promoting endothelial damage, but rather by lacking supportive contribution to endothelial regeneration and healing.  相似文献

Alkylresorcinols are members of an extensive family of bioactive compounds referred to as phenolic lipids, which occur primarily in plants, fungi and bacteria. In plants, alkylresorcinols and their derivatives are thought to serve important roles as phytoanticipins and allelochemicals, although direct evidence for this is still somewhat lacking. Specialized type III polyketide synthases (referred to as ‘alkylresorcinol synthases’), which catalyze the formation of 5-alkylresorcinols using fatty acyl-CoA starter units and malonyl-CoA extender units, have been characterized from several microbial species; however, until very recently little has been known concerning their plant counterparts. Through the use of sorghum and rice EST and genomic data sets, significant inroads have now been made in this regard. Here we provide additional information concerning our recent report on the identification and characterization of alkylresorcinol synthases from Sorghum bicolor and Oryza sativa, as well as a brief consideration of the emergence of this intriguing subfamily of enzymes.Key words: alkylresorcinol, polyketide synthase, alkylresorcinol synthase, phenolic lipid, antifungal  相似文献

Lipase (EC.3.1.1.3) from Candida sp. 99-125 was separated into four isoforms (isoform A, isoform B, isoform C, and isoform D) by two steps of ion exchange chromatography. As analyzed on SDS- and non-denaturing PAGE, the four isoforms were homogenous and had the same molecular weight of approximate 38 kDa. MALDI-TOF peptide mass fingerprinting maps and circular dichroism spectra showed the isoforms had similar peptide patterns belonging to the same protein encoded by the YLlip2 gene and different secondary structures. The isoforms had a little distinct optimum temperature in the range of 20–35 °C, and the same optimum pH (8.0). They remained to be active in methanol, ethanol and ethylene glycol at the concentration of 10% and 20% (v/v) and acetone at the concentration of 10% (v/v), and sensitive to EDTA. Triton X-100, Sodium cholate and CHAPS slightly increased their activities. The metal ion Ca2+ and Mg2+ had mild effect on lipase activity. The isoforms showed a preference for long chain fatty acid triglyceride (triolein and olive). The lipase purified by one step of ion exchange chromatography and isoforms were less active than crude enzyme to catalyze cetyl alcohol and oleic acid in n-hexane, whereas the presence of small concentration of added water dramatically activated crude lipase but less the purified preparations.  相似文献
