Morphology and end product patterns were used to group 112 bacterial isolates from the ceca and colons of pigs fed unmedicated and chlortetracycline-containing diets. Most of the isolates (68%) were resistant to chlortetracycline (greater than 4 micrograms/ml). Chlortetracycline resistance was associated with all groups of anaerobic and facultative bacteria.  相似文献

Somatic cell genetic mapping of marsupial and monotreme species will greatly extend the power of comparative gene mapping to detect ancient mammalian gene arrangements. The use of eutherian-marsupial cell hybrids for such mapping is complicated by the frequent retention of deleted and rearranged marsupial chromosomes. We used staining techniques, involving the fluorochromes Hoechst 33258 and chromomycin A3, to facilitate rapid and unequivocal identification of marsupial chromosomes and chromosome segments and to make chromosome assignment and regional localization of marsupial genes possible. Chromosome segregation in rodent-macropod hybrids was consistent with preferential loss of the marsupial complement. The extent of loss was very variable. Some hybrids retained 30% of the marsupial complement; some retained small centric fragments; and some, no cytologically identifiable marsupial material. We examined the chromosomes and gene products of a number of rodent-grey kangaroo Macropus giganteus hybrids, and have assigned the genes Pgk-A (phosphoglycerate kinase-A), Hpt (Hypoxanthine phosphoribosyl transferase), and Gpd (Glucose-6-phosphate dehydrogenase) to the long arm of the kangaroo X chromosome, and provisionally established the gene order Pgk-A -Hpt -Gpd.  相似文献

Crude mitochondrial fractions containing a substantial amount of microsomes accumulate Ca2+ in the presence of ATP, ruthenium red and oligomycin. A proportion of this accumulated Ca2+ is released by the addition of low concentrations (ca. 1 microM) of inositol (1,4,5) trisphosphate . Under some conditions the release is transient, and evidence is presented which suggests that this is due to inhomogeneity in the vesicle population. (1,4,5)inositol trisphosphate -induced Ca2+ release can also be demonstrated, under appropriate experimental conditions, in a more purified microsomal fraction essentially free of mitochondria.  相似文献

Approximately 20,000 different short, linear, macronuclear DNA molecules are derived from micronuclear sequences of Oxytricha fallax after conjugation. These macronuclear DNAs are terminated at both ends by 20 base pairs of the sequence 5'-dC4A4-3'. Sequences homologous to this repeat (C4A4+) are also abundant in the micronuclear chromosomes, but most reside at their telomeres. Here we show that nontelomeric C4A4 clusters of 20 base pairs or longer exist in only a few hundred copies per micronuclear genome. This demonstrates that nearly none of the 20,000 sequence blocks of micronuclear DNA destined to be macronuclear DNA molecules can be flanked by full-length (20-base pair) C4A4 clusters, and therefore C4A4 repeats must be added to most, if not all, macronuclear telomeres during macronuclear development. Six internal micronuclear C4A4+ loci were cloned, and their structural relationships with macronuclear and micronuclear sequences were examined. The possible origins and functions of these rare, micronuclear internal C4A4 loci are discussed.  相似文献

Summary We present here a familial case of a paracentric inversion in man with a short review of the literature. A paracentric inversion of chromosome 10(q11q26) was found in the amniocytes drawn for advanced maternal age. The presence of the inversion was investigated in 35 family members in three generations. No recombinants were recognized. Summary Insulin release and membrane potential fluctuations in response to increased extracellular potassium [K+] o have been measured in single perifused islets of Langerhans from normal mice. An increase in [K+] o from 5mm to 50mm induced a transient insulin release with a peak at about 1 min. The peak value was [K+] o -dependent but the half-timet 1/2 for the decline was constant at nearly 1 min. 2.5mm cobalt completely inhibited the potassium-induced stimulation of insulin release. The insulin release elicited by 28 and 50mm [K+] o was similar in terms of peak, total release and half-time from maximum release. Stepwise increase in [K+] o from 10 to 28 to 50mm resulted in a normal response to 28mm but no peak of release after the 28 to 50mm increase. The results indicate good correlation between excess voltage noise, thought to reflect calcium channel activity, and insulin release evoked by changing extracellular potassium.  相似文献

The structures of the diabolic acid-containing phospholipids of Buryrivibrio S2 grown in the presence of palmitic acid have been investigated. Generally they consist of two conventional bacterial phospholipid or galactolipid structures linked by esterification through a single diabolic acid residue. The main lipid consists of the butyroyl ester of sn-1-alkenylglycero-3-phospho-1'-sn-glycerol joined in this way to the butyroyl ester of sn-1-alkenyl-3-galactosylglycerol by esterification of the vacant 2-hydroxy groups of the alkenyl-substituted glycerol molecules. A lipid that possess a palmitoyl group rather than one of the butyroyl groups of the latter structure has also been detected. The lipid occurring in the second highest concentration consists of two molecules of sn-1-alkenylglycero-3-phospho-sn-1'-glycerol butyroyl ester linked through diabolic acid in a similar manner to the main lipid. Other lipids with the latter structure either minus a butyroyl group or with palmitoyl group, instead of one of the buryroyl groups, exist as minor components.  相似文献
