To determine and compare the direct effects of prostaglandin F2a (PGF2a) and human chorionic gonadotropin (hCG) on luteal cell progesterone production in vitro, 9 human corpora lutea obtained at tubal ligation were minced and treated with collagenase to disaggregate luteal cells. Dispersed luteal cells (80% viable) were incubated in air at 37 degrees C in a shaking water bath for 3 h and total progesterone in the media and cells was determined by radioimmunoassay. Optimum progesterone production was obtained using 25,000 or more cells per incubate and an incubation time of 2-4 h. hCG-stimulated progesterone production increased significantly with 0.01 IU to as high as 100 IU. In the early luteal phase (days 1-5 post ovulation or days 15-20 of the luteal phase), PGF2a (10-1000 ng) significantly inhibited progesterone production but significantly stimulated progesterone production in the mid-luteal phase (days 21-25). PGF2a had no effect on luteal cell progesterone production in the late luteal phase (days 26-30). This age-dependent direct effect of PGF2a on human luteal cell progesterone production in vitro indicates a role for PGF2a in the total intragonadal regulation of progesterone output, possibly through a paracrine or autocrine manner directed towards synchronizing luteal progesterone secretion and endometrial preparation for nidation.  相似文献

Samples of two calcareous soils from central Iraq were amended with 2, 6 and 10 mg g−1 soil of agriculturegrade sulphur and incubated for 4, 8 and 12 weeks at 30°C and 70% water-holding capacity. At the end of each incubation period, soils were analyzed for sulphate, CaCO3 and CaSO4. The sulphate content of the soils increased, whereas the CaCO3 content decreased, with increasing S levels and incubation time. The gypsum formed was in proportion to the sulphur oxidised and the calcium released from CaCO3.  相似文献

Dracaena leaf proliferosis is a newly reported disease affecting Dracaena sanderiana in Egypt. A cause and effect relationship between this disease and the fungus,Fusarium proliferatum var. minus has been established. In addition to D. sanderiana the fungus was found to be pathogenic, when tested in the laboratory, to several other members of the family Liliaceae. While the in vitro growth of the fungus is optimum at 25 °C, symptom expression is best at 30°C. Twelve fungicides were tested for their in vitro effect on fungal growth. Benlate, Rubigan, Saprol, Cercobin and Vitavax-200 came first on the list and inhibited growth at 2.5, 12.0, 55.0, 75.0, and 94.0 μg/ml, respectively. Although, Benlate was the most effective fungicide in this respect it failed to demonstrate similar effect on disease development when applied to plants artificially inoculated with the fungus. Fungal growth was completely inhibited on PDA medium by a bacterium belonging to Bacillus sp. but when the bacterium at a concentration of 1 × 1011 cell/ml was applied 24 h before, at the same time with, or 24 h after inoculation no control of the disease was achieved. Naturally-infected plants could, however, be freed from infection when subjected to a hot air treatment at 35 ± 5 °C during day time and 25 ± 5° C at night for 3 months.  相似文献

Treatment of 2-bromoacetylbenzofuran with 1H-benzotriazole afforded 1-(benzofuran-2-yl)-2-(benzotriazol-1-yl)ethanone which reacted with phenylisothiocyanate to give the corresponding thioacetanilide derivatives. Treatment of the latter ethanone and thioacetanilide derivatives with hydrazonoyl chlorides afforded the corresponding pyrazole and 1,3,4-thiadiazole derivatives. The thioacetanilide derivative reacted with alpha-haloketones and alpha-halodiketones to afford thiophene and thiazole derivatives, respectively. The newly synthesized compounds were found to possess anticonvulsant and anti-inflammatory activities with the same mechanism of action of selective COX-2 inhibitors.  相似文献

Although recent research has increasingly focused on human sexual selection, fundamental questions remain concerning the relative influence of individual traits on success in competition for mates and the mechanisms, form, and direction of these sexual selective pressures. Here, we explore sexual selection on men's traits by ascertaining men's dominance and attractiveness from male and female acquaintances. On a large American university campus, 63 men from two social fraternities provided anthropometric measurements, facial photographs, voice recordings, and reported mating success (number of sexual partners). These men also assessed each other's dominance, and 72 women from two socially affiliated sororities assessed the men's attractiveness. We measured facial masculinity from inter-landmark distances and vocal masculinity from acoustic parameters. We additionally obtained facial and vocal attractiveness and dominance ratings from unfamiliar observers. Results indicate that dominance and the traits associated with it predict men's mating success, but attractiveness and the traits associated with it do not. These findings point to the salience of contest competition on men's mating success in this population.  相似文献

Low molecular weight phenolics are suggested to have a role in mediating diet selection in mammalian herbivores. However, very little is known about low molecular weight phenolic profiles of African savanna woody species. We determined low molecular weight phenolic profiles of six woody species with different life history, morphological and functional traits. We investigated interspecific phytochemical variation between species and found that: (1) related Acacia species were chemically dissimilar; (2) similarity percentage analysis revealed that Acacia grandicornuta was most dissimilar from other species and that the evergreen and unpalatable Euclea divinorum had a qualitatively similar chemical profile to the deciduous and palatable Acacia exuvialis and Combretum apiculatum; (3) C. apiculatum had the highest chemical diversity; (4) relative to spineless plants, spinescent plants contained significantly less HPLC phenolics and condensed tannins; and (5) the major quantitative difference between the evergreen and unpalatable E. divinorum and other species was its high myricitrin concentration.  相似文献

Members of the NDR (nuclear Dbf2-related) protein-kinase family are essential for cell differentiation and polarized morphogenesis. However, their functions in plant pathogenic fungi are not well understood. Here, we characterized the NDR kinase FgCot1 and its activator FgMob2 in Fusarium graminearum, a major pathogen causing Fusarium head blight (FHB) in wheat. FgCot1 and FgMob2 formed a NDR kinase–MOB protein complex. Localization assays using FgCot1-GFP or FgMob2-RFP constructs showed diverse subcellular localizations, including cytoplasm, septum, nucleus and hyphal tip. ΔFgcot1 and ΔFgmob2 exhibited serious defects in hyphal growth, polarity, fungal development and cell wall integrity as well as reduced virulence in planta. In contrast, lipid droplet accumulation was significantly increased in these two mutants. Phosphorylation of FgCot1 at two highly conserved residues (S462 and T630) as well as five new sites synergistically contributed its role in various cellular processes. In addition, non-synonymous mutations in two MAPK (mitogen-activated protein kinase) proteins, FgSte11 and FgGpmk1, partially rescued the growth defect of ΔFgmob2, indicating a functional link between the FgCot1–Mob2 complex and the FgGpmk1 signalling pathway in regulating filamentous fungal growth. These results indicated that the FgCot1–Mob2 complex is critical for polarity, fungal development, cell wall organization, lipid metabolism and virulence in F. graminearum.  相似文献
