In mature rats an area on the head has been subjected to a single radiation for 1.5 sec with microwaves in the continuous regimen of generation, frequency 2.4 GHz level of the specific absorbed power 5 W/g, that is accompanied with appearance of convulsions. Under anesthesia specimens of the superficial layer of the cerebral superlateral part are taken and subjected to electron microscopical investigation. Immediately after radiation and in 2 h certain disorders in microcirculation and reactive changes of mitochondria in perikaryons, axons, dendrites, synapses of the neurons and in gliocytes are revealed. The mitochondrial changes are designated as "edematous". In 2 and 6 h in karyoplasm of some neurons membranous structures appear; they are interpreted as a result of heat denaturation of the nuclear proteins. In synapses, together with lesions of mitochondria, synaptic complexes undergo destruction and osmiophilic substance is accumulated in the subsynaptic zone along the whole length of the contact. In one day, essential destructive changes are revealed as severe lesions of some neurons, vacuolization and destruction of mitochondria, localized in all the structures. Pathogenesis of the neurological disturbances is based on disturbances of interneuronal interactions, connected with an immediate heat effect of the electromagnetic radiation on the structures responsible for the synaptic transmission and with a rapidly developing tissue hypoxia as a consequence of microcirculatory disturbance and a sharp inhibition of energetic metabolism.  相似文献

Kinetics of glycogen binding by glycogen phosphorylase b has been studied by stopped flow and temperature jump methods. This reaction is followed by increase in light scattering whose amplitude depends upon the enzyme binding sites concentration of glycogen particles occupied by the enzyme. It has been shown that the complex formation has the first order with respect to enzyme and glycogen concentrations. Relaxation kinetics is compatible with proposed bimolecular reaction scheme. Microscopic rate constants of the forward and reverse reactions of glycogen binding by glycogen phosphorylase b are determined in temperature range from 12,7 to 30 degrees C. The possibility of diffusional control of the binding rate is discussed.  相似文献

Low temperature (77 degrees K) absorption spectra of nonequilibrium states of cytochrome c oxidase produced by reduction of oxidases form protein by thermolysed electrons at 77 degrees K was studied. During reduction of cytochrome oxidase water-glycerol solution by thermolysed electrons at 77 degrees K a nonequilibrium reduced protein is formed. Low temperature (77 degrees K) absorption spectra of the nonequilibrium cytochrome oxidase differs from those reduced by ditionite. It was shown that the oxidation state of cytochrome a3 or addition of cytochrom c have no influence on these spectral changes. It is assumed, that the observed effects are conditioned by structural differences of reduced and oxidased cytochrome oxidase active center. Similar spectral changes were observed for cytochrome oxidase, bound to the mitochondrial membrane. At temperature increasing the low temperature reduced protein is relaxed to a corresponding equilibrium state. The spectral properties of bacterial cytochrome oxidase M. lysodeicticus do not depend on the way of reduction (by dytionite or thermolysed electrons at 77 degrees K).  相似文献

Study of the motor dominant in man has shown that in aftereffect of rhythmic electrocutaneous stimulation of hand, a latent focus of excitation is formed in the CNS. When using sensory stimuli (light, sound) and also stimuli, addressed to the second signal system, a hand motor reaction appears in the rhythm of earlier electrocutaneous stimulation, what testifies to preservation of rhythmic nature of excitation in the dominant focus. Summation process in the dominant focus is not reflected in human consciousness. Study of the brain biopotentials has shown that at formation of human motor dominant, typical changes of spectral-coherent characteristics of the neocortex electrical activity appear: within the "dominant" hemisphere, the spectrum power, increases in the range of low frequencies; highly coherent connections of electrical processes appear in the delta-range of the frontal-parietal area at conjugate decrease of combination of processes in the parietal-occipital area of the cerebral cortex. Formation of the dominant focus in one hemisphere causes general reconstruction of the structure of intercentral relations of both hemispheres electrical activity.  相似文献

Recombinant plasmids coding for hybrid proteins between human interferon gamma and human tumour necrosis factor alpha or beta have been constructed using site-directed mutagenesis. The genes were fused via a synthetic oligonucleotide linker coding for tetrapeptide Pro-Val-Gly-Pro. The fused genes were expressed in Escherichia coli under control of early promoters of bacteriophage T7. E. coli cells harbouring the plasmids with the hybrid genes gave rather high level of the fused proteins biosynthesis. The hybrid recombinant proteins proved to be unstable in E. coli cells.  相似文献

The effect of high pressure on the spectral properties of cytochrome P-450 LM2(Fe2+)-CO complex was studied. The application of high pressure was shown to induce the conversion of cytochrome P-450 to P-420. In the solution when P-450 was oligomeric only about 65% of the total converted to P-420. The remaining portion of cytochrome P-450 was stable at pressures up to 6 kbar. When P-450 was incorporated into membranes or when it was succinylated, the proportion of the pressure sensitive fraction was slightly higher (about 75%). Dissociation of P-450 oligomers into monomers was made by addition of 0.2% Triton N-101. Monomers were the most sensitive to pressure; they could be completely converted to P-420. These results have been interpreted as evidence for the existence of two different conformers of P-450 LM2, which differ in pressure stability. Splitting between these two states appears to be a result of the oligomeric organization of cytochrome P-450 in solution and in the membrane.  相似文献

The use of the methods of multifactor, orthogonal and composition planning in studying the behavioural disturbances in rats after gamma irradiation with doses of 0.258 to 1.29 C/kg and the application of the proposed method of discrimination of effects by empirical models permitted to establish the informative and adequate dependences of the probability of these disturbances on dose of nonuniform irradiation and the degree of strengthening of the conditioned reflex. It was shown that the effect of radiation decreased, in a discrete manner, the probability of making the first decision by rats in a behavioural task (here we deal with the "dose-response" function). The average time of making the first decision after irradiation was invariable within the dose range under study. Within the range of the studied factors both the value of the dose of whole-body irradiation and the degree of strengthening of the conditioned reflex significantly affected the probability of fulfilling the task by the animals the significance of the radiation dose being several times higher. The effects of the interaction of the two factors, that is, irradiation and the degree of the radiation affection, were insignificant in changing the behavioural reactions under study.  相似文献
