Mutual effects between Mn, Ca, and Mg were studied during steady-state absorption experiments with excised barley roots. Calcium appeared to enhance the rate of Mn absorption; whereas, Mg had a highly depressive effect. The combination of both Ca and Mg was even more inhibitory to Mn absorption than Mg alone. Manganese had no effect on the usual negligible Ca absorption by this tissue, but effectively inhibited the absorption of Mg. Although divalent cation absorption from the Ca-Mg-Mn system was essentially nil, K absorption was greatly stimulated in the presence of these cations.These mutual effects and others reported in the literature are explained by the hypothesis that selectivity in ion absorption results from cation-induced conformational changes in the structure of the carrier molecule.  相似文献

Summary Observations on the inhibition of root elongation and cell division in Allium cepa showed that the toxic effects of scandium and aluminium were very similar. Tracer uptake studies using 46Sc indicated that the rate of uptake in the apical 3.0 mm of the axis was more rapid than elsewhere in the root and proceeded in two distinct phases; Phase 1, probably superficial adsorption, was characterised by a rapid initial rate which was little affected by low temperature, the rate of Phase 2 was slower but remained constant for 24 hours and was highly dependent on temperature.Autoradiographs from roots treated for 30 min with 46Sc showed that most of the isotope in the root tip was concentrated in a peripheral belt corresponding with the mucigel layer of the root cap and it is suggested that this is the site of Phase 1 adsorption. The underlying root cap and epidermal cells retained little scandium but interior to them some isotope was associated with dividing cells; this increased steadily over 6 hour to an estimated concentration of 30 mM, and possibly represents Phase 2 uptake. Differentiation and secondary wall formation in the cortex restricted the rate of radial penetration of scandium. The primary endodermis restricted the entry of scandium into the stele at a very early stage in its development, which leads to the conclusion that migration of the ion across the root is primarily in the free space.Scandium enters the dividing cells in advance of observable effects on cell division, a situation compatible with the direct involvement of this ion in the inhibition of the mitotic cycle. Suggestions are made on the mechanisms by which polyvalent cations might disturb cell division and extension.  相似文献

Zusammenfassung In normalen Leberzellen der Maus wurde das quantitative Verhalten des perimitochondrialen granulären endoplasmatischen Retikulums untersucht. 74% der Mitochondrien zeigen Beziehungen zum granulären endoplasmatischen Retikulum. Die einzelnen Mitochondrien werden zu 52%19,5 von den Membranen des granulären endoplasmatischen Retikulums bedeckt. Je Mikrometer Membranstrecke sind auf der mitochondriennahen Seite des granulären endoplasmatischen Retikulums 21 und auf der mitochondrienfernen Seite 20 Ribosomen zu finden, was der Zahl im übrigen granulären endoplasmatischen Retikulum entspricht. Die Befunde stellen die Grundlage für Untersuchungen des perimitochondrialen granulären endoplasmatischen Retikulums unter pathologischen Bedingungen dar.
Structure and quantitative behaviour of the perimitochondrial granular endoplasmic reticulum in the liver cells of the mouse
Summary The perimitochondrial granular endoplasmic reticulum in normal mouse liver cells has been investigated quantitatively. 74% of the mitochondria are in association with the granular endoplasmic reticulum. The individual mitochondrion is covered by the membranes of the granulated E. R. in 52%19.5. The outer surface of the endoplasmic membrane, facing the mitochondrion, is occupied by 21 ribosomes per m; the corresponding surface of the membrane facing the free cytoplasm is occupied by 20 ribosomes per m. These data are in agreement with those of that fraction of the E. R., which is not in association with mitochondria. These findings represent a basis for investigations of the perimitochondrial endoplasmic reticulum under pathological conditions.

1. The frequency of occurrence, under defined conditions, of the different human pepsins in the gastric juices of 50 normal subjects was investigated by agar-gel electrophoresis. 2. From a total of eight proteolytic zones located in the zymograms, no significant differences of occurrence existed between the sexes, or between subjects with or without gastric symptoms. 3. Two zones, numbered 3 and 5, occurred in all normal gastric juices. Zone 3 always exhibited the greatest proteolytic activity, then zone 5. The remaining enzymic zones were less well-marked and occurred less frequently. 4. A minor zone, 3a, was demonstrated within zone 3. The corresponding pepsin, 3a, has a mobility towards the anode 6-7% greater than has pepsin 3. 5. Of the eight zones, 1,2,3,3a and 5, at least, represent unique pepsins.  相似文献

1. Glycerol kinase (EC 2.7.1.30) activity was measured in crude extracts of skeletal muscles by a radiochemical method. The properties of the enzyme from a number of different muscles are very similar to those of the enzyme from rat liver. Glycerol kinase from locust flight muscle was inhibited competitively by l-3-glycerophosphate with a K(i) of 4.0x10(-4)m. 2. The activity of glycerol kinase was measured in a variety of muscles from vertebrates and invertebrates in an attempt to explain the large variation in the activity of this enzyme in different muscles. 3. In vertebrates glycerol kinase activities were generally higher in red muscle than in white muscle; the highest activities (approx. 0.2mumole/min./g. fresh wt.) were found in the red breast muscle of some birds (e.g. pigeon, duck, blue tit) whereas the activities in the white breast muscle of the pheasant and domestic fowl were very low (approx. 0.02mumole/min./g.). 4. On the basis of glycerol kinase activities, muscles from insects can be classified into three groups: muscles that have a low enzyme activity, i.e. <0.3mumole/min./g. (leg muscles of all insects studied and the flight muscles of cockroaches and the tsetse fly); muscles that have an intermediate enzyme activity, i.e. 0.3-1.5mumoles/min./g. (e.g. locusts, cockchafers, moths, water-bugs); and muscles that have a high enzyme activity, i.e. >1.5mumoles/min./g. (e.g. bees, wasps, some blowflies). 5. The function of glycerol kinase in vertebrate and insect muscles that possess a low or intermediate activity is considered to be the removal of glycerol that is produced from lipolysis of triglyceride or diglyceride by the muscle. Therefore in these muscles the activity of glycerol kinase is related to the metabolism of fat, which is used to support sustained muscular activity. A possible regulatory role of glycerol kinase in the initiation of triglyceride or diglyceride lipolysis is discussed. 6. The function of glycerol kinase in the insect muscles that possess a high activity of the enzyme is considered to be related to the high rates of glycolysis that these muscles can perform. The oxidation of extramitochondrial NADH, and therefore the maintenance of glycolysis, is dependent on the functioning of the glycerophosphate cycle; if at any stage of flight (e.g. at the start) the rate of mitochondrial oxidation of l-3-glycerophosphate was less than the activity of the extramitochondrial glycerophosphate dehydrogenase, this compound would accumulate, inhibit the latter enzyme and inhibit glycolysis. It is suggested that such excessive accumulation of l-3-glycerophosphate is prevented by hydrolysis of this compound to glycerol; the latter would have to be removed from the muscle when the accumulation of l-3-glycerophosphate had stopped, and this would explain the presence of glycerol kinase in these muscles and its inhibition by l-3-glycerophosphate.  相似文献

The role of ribonucleic acid (RNA) synthesis in the development of sporangia in the saprolegniaceous mold Achlya was studied. Methods were developed for growing and treating large populations of mycelia so that the hyphal tips would differentiate into sporangia with considerable synchrony. Under the starvation conditions imposed for the differentiation of sporangia, net RNA, deoxyribonucleic acid (DNA), and protein synthesis ceased. However, incorporation of radioactive precursors into RNA continued at a high rate throughout the period of differentiation, showing that the enzymatic mechanism for RNA synthesis was still in an active state. Actinomycin D inhibited the differentiation of sporangia and the incorporation of labeled precursors into RNA. The level of actinomycin used did not inhibit the normal outgrowth and branching of the mycelia that occurred during differentiation. Thus, DNA-dependent RNA synthesis was required for the differentiation of sporangia. Sucrose gradient analysis of newly synthesized RNA showed that only the ribosomal and soluble fractions of RNA were labeled during vegetative growth. During the differentiation of sporangia, ribosomal and soluble RNA fractions were also labeled, and, in addition, a heterodisperse fraction of labeled RNA which was heavier than ribosomal RNA appeared; this fraction was not evident in the newly synthesized RNA from vegetative mycelia.  相似文献
