We have examined the growth behavior of small numbers of interstitial stem cells transplanted into tissue of genetically unrelated strains of Hydra magnipapillata. We show that such stem cells, which are at low density following transplantation, proliferate more rapidly than the stem cells of the host, which are at normal density. The rapid proliferation is similar to the proliferation rate of stem cells transplanted into interstitial cell free tissue. The results suggest that stem cells transplanted into heterotypic tissue are unable to "sense" the presence of host stem cells and to adopt their growth rate to that of the surrounding cells. Thus, the feedback signal which negatively regulates stem cell growth as a function of stem cell density must be strain specific.  相似文献

Summary Our current knowledge of the control of ventilation in fish is incomplete at all levels. The respiratory rhythm originates in a medullary central pattern generator (CPG), which has yet to be clearly identified and characterized. Its activity is directly modulated by inputs from elsewhere in the CNS and from peripheral mechanoreceptors. The central location of respiratory motoneurones, innervating the various respiratory muscles, has been described in detail for some fish, particularly elasmobranchs. We are still unclear, however, about the link between the CPG and the sequential firing of the motoneurones, which result in coordinated contractions of the respiratory muscles, and about the mechanisms that result in recruitment of feeding muscles into forced ventilation. In teleosts, ventilation is matched to oxygen requirements by stimulation of gill chemoreceptors, which seem to respond to oxygen content or supply. There is little evidence of a role for these receptors in elasmobranchs.Chemoreceptor stimulation evokes a number of reflex changes in the respiratory and cardiovascular systems of fish that are rapid in onset and seem adaptive (e.g. increased ventilation and a bradycardia in response to hypoxia). Conditions that result in hypoxaemia and the consequent ventilatory changes also cause an elevation in circulating catecholamine levels. We have explored the possibility of a causal relationship between these levels and the ventilatory response. Strong evidence for this relationship arises from experiments on hypoxia and acid infusion, which trigger a ventilatory increase and a rise in circulating catecholamines. Both ventilatory responses are blocked by an injection of propranolol, indicating that adrenoreceptors are involved in the response.The ventilatory response to hypoxia, in teleosts at least, occurs very rapidly, perhaps before any marked increase in circulating catecholamines and almost certainly before any blood-borne catecholamines could reach the respiratory neurones. This argues for an immediate neuronal reflex based on chemoreceptors in the gill region responding to hypoxia. Clearly, circulating catecholamines also affect ventilation through some action in the medulla and could act in concert with a direct neuronal chemoreceptive drive during hypoxia. The studies on acid infusion during hyperoxia, where there is an acidosis but no increase in ventilation or blood catecholamines, would argue against any hydrogen ion receptor, either peripheral or central, being involved in the reflex ventilatory response to acidotic conditions in fish.The release of catecholamines into the circulation, therefore, seems to be an absolute requirement for the ventilatory response to acidosis in fish. Present evidence supports a role for -adrenergic receptors on respiratory neurones, stimulated by changes in the levels of circulating catecholamines, in the control of ventilatory responses to marked changes in oxygen availability in fish, such as those occurring in the post-exercise acidotic state.  相似文献

The effects of chilling on tomato ( Lycopersicon esculentum Mill cv. Caruso) texture were investigated using fruit stored at 22°C (nonchilled) or 5°C (chilled) for 28 days. or at 5°C for 15 days before transfer to 22°C to facilitate ripening during and additional 13 days (prechilled). Prechilled fruit exhibited symptoms of slight chilling injury, i.e. development of mealiness, accelerated softening relative to that of nonchilled fruit and nonuniform surface colour development. The firmness of all fruit decreased during ripening and chilled storage when measured by flat plate compression and puncture, especially during the early stages of ripening of nonchilled and prechilled fruit. The compression firmness of pericarp tissue similarly decreased during ripening of nonchilled and prechilled fruit, but was maintained during chilling. Total moisture content (ca 94%) of tissue, uronide content (32-35% w/w) and extracted β-galactosidase activity did not differ significantly ( P > 0.05) among fruit during ripening and chilled storage. The degree of uronide methyl esterification in ethanol-insoluble solids prepared from pericarp tissue (EIS) was relatively low for all fruit. i.e. <40%. EIS from which greater levels of pectinesterase were extracted (i.e. nonchilled>chilled>prechilled) exhibited decreased levels of uronide methyl esterification. Markedly elevated levels of β-glucosidase activity were extracted from prechilled EIS. Total polygalacturonase activity (mainly as PGI) and autolysis of enzyme-extracted EIS were inversely correlated ( P ≤ 0.05) only with the loss of nonchilled fruit and tissue firmness and prechilled fruit firmness. Results suggest a possible role for β-glucosidase in textural changes of prechilled fruit and tissue (e.g. loss of firmness, development of mealiness) and also implicate loss of skin strength in the softening of whole fruit during chilling.  相似文献

Guard cell responses to light are mediated by guard cell chlorophyll and by a specific blue light photoreceptor. Gas exchange and epidermal peel techniques were employed to investigate these responses in the facultative Crassulacean acid metabolism (CAM) species, Portulacaria afra (L.) Jacq. In P. afra individuals performing C3 metabolism, red light stimulated an increase in leaf conductance in intact leaves and stomatal opening in isolated epidermal peels, indicating the presence in guard cells of the chlorophyll-mediated response to light. Under a background of continuous red illumination, conductance exhibited transient increases following pulses of blue but not red light, indicating that the specific stomatal response to blue light was also operative. In contrast, in CAM individuals, conductance in gas exchange experiments and stomatal opening in epidermal peel experiments were not stimulated by red light. In CAM plants, conductance did not increase following blue light pulses administered over a range of temperatures, vapor pressure differences (VPD), ambient CO2 concentrations and background red light intensities. These results indicate that P. afra does possess typical guard cell responses to light when performing C3 metabolism. The metabolic pathways mediating these responses are either lost or inhibited when CAM is induced.  相似文献

The metabolic cost of swimming for planktonic protists is calculated, on theoretical grounds, from a simple model based upon Stokes' law. Energetic expenditure is scaled over both typically encountered size ranges (1–100 µm) and swimming speeds (100–5,000 µm/sec). In agreement with previous estimates for typical flagellates, these estimates generally suggest a low (<1%) cost for motility, related to total metabolic rate of growing cells. However, the cost of motility in small, fast-moving forms, such as some ciliates and flagellates, may be significant (1–10%) and even substantial (10–100%+) for certain species. In accordance with these predictions, many fast-moving ciliates restrict motility to bursts of activity or jumps. In the absence of a reduction in swimming speed or in the frequency of jumps, it is predicted that this relative cost of motility will be significantly increased in starving heterotrophs or light-limited autotrophs, if such cells reduce cell volumes and specific rates of respiration.  相似文献
