The microcirculation is the site of gas and nutrient exchange. Control of central or local signals acting on the myocytes, pericytes and endothelial cells within it, is essential for health. Due to technical problems of accessibility, the mechanisms controlling Ca2+ signalling and contractility of myocytes and pericytes in different sections of microvascular networks in situ have not been investigated. We aimed to investigate Ca2+ signalling and functional responses, in a microcirculatory network in situ. Using live confocal imaging of ureteric microvascular networks, we have studied the architecture, morphology, Ca2+ signalling and contractility of myocytes and pericytes. Ca2+ signals vary between distributing arcade and downstream transverse and precapillary arterioles, are modified by agonists, with sympathetic agonists being ineffective beyond transverse arterioles. In myocytes and pericytes, Ca2+ signals arise from Ca2+ release from the sarcoplasmic reticulum through inositol 1,4,5-trisphosphate-induced Ca2+ release and not via ryanodine receptors or Ca2+ entry into the cell. The responses in pericytes are less oscillatory, slower and longer-lasting than those in myocytes. Myocytes and pericytes are electrically coupled, transmitting Ca2+ signals between arteriolar and venular networks dependent on gap junctions and Ca2+ entry via L-type Ca2+ channels. Endothelial Ca2+ signalling inhibits intracellular Ca2+ oscillations in myocytes and pericytes via L-arginine/nitric oxide pathway and intercellular propagating Ca2+ signals via EDHF. Increases of Ca2+ in pericytes and myocytes constrict all vessels except capillaries. These data reveal the structural and signalling specializations allowing blood flow to be regulated by myocytes and pericytes. 相似文献

Within the last fifteen years Spain and Portugal have begun to experience a phenomenon that occurred in northern Europe three or four decades earlier. Rising prosperity, an ageing population, and growing labour shortages have triggered a demand for foreign labour in the Iberian economies. This survey article identifies the changing trends and developments of recent immigration to two southern European countries, and the problems of labour market integration. It compares and contrasts the insertion process in Spain and Portugal and highlights the policy dilemma that, like most of the rest of the EU, the Iberian economies require immigrant workers, but have yet to develop effective mechanisms for integrating them. It argues that the labour market is not only the principal mechanism by which immigrants are integrated, but is beginning to transform attitudes towards immigration. Employer organizations have played a lead role in pressing for a shift from restriction and control to the management of immigrant integration. However, the needs of employers, who require a supply of cheap, flexible labour clashes with the government's exclusion policy, as the case study based on Lorca, Spain demonstrates. 相似文献

Cloning is widely considered only to be a biological discourse. Few, however, have paid attention to the cultural contexts that have made cloning conceivable. The relation between the biological and cultural considerations of cloning are revealed by the anxieties conjured up by the prospects of cloning human beings. By cloning we understand the reproduction of sameness which is deeply ingrained in the organization and reproduction of culture. The ease with which cloning has been taken up in contemporary thinking has been made possible by the widespread saturation of the normative assumption of socio-cultural sameness underpinning much of mainstream modern thinking around politics, law, education, management, aesthetics, the military and processes of production. Oxydative cyclization of lapachol was attempted by feeding this compound into the culture broth of Beauveria sulfurescens and two ionophore-producing strains of Streptomyces albus and Streptomyces griseus. As a result, three compounds were obtained, lomatiol, lomatic acid and lomatiol acetate, only with B. sulfurescens and S. albus. We use stable isotope data to investigate the role of winter habitat use in altering the breeding phenology of yellow warblers Setophaga petechia. We first confirm that δ13C and δ15N isotopic signatures vary with winter habitat use in this species. We then examine the relationship between winter habitat use, breeding phenology and productivity within four age‐sex‐classes, since life history theory would predict that carry‐over effects should vary with age and gender. The δ13C signatures of yellow warblers using riparian habitats over winter were more depleted than the signatures of those using agricultural or scrub habitat. Individuals on the Pacific coast of Mexico were also more δ15N enriched than those on the southern Gulf of Mexico. δ13C and δ15N signatures were only correlated with earlier clutch initiation and subsequent higher productivity in first‐breeding‐season females. We estimate that shifts in δ13C equivalent to a shift from scrub to riparian winter habitat would be associated with the production of 0.8 more fledglings by yearling females. Pre‐breeding events that influence the timing of breeding could also influence the reproductive performance of older males and females, but we found little evidence that winter habitat use influenced breeding season phenology in these birds. 相似文献

Glucokinase (hexokinase IV) continues to be a compelling target for the treatment of type 2 diabetes given the wealth of supporting human genetics data and numerous reports of robust clinical glucose lowering in patients treated with small molecule allosteric activators. Recent work has demonstrated the ability of hepatoselective activators to deliver glucose lowering efficacy with minimal risk of hypoglycemia. While orally administered agents require a considerable degree of passive permeability to promote suitable exposures, there is no such restriction on intravenously delivered drugs. Therefore, minimization of membrane diffusion in the context of an intravenously agent should ensure optimal hepatic targeting and therapeutic index.
