Control of replication of FII plasmids: comparison of the basic replicons and of the copB systems of plasmids R100 and R1

The copy numbers of the FII plasmids R1 and R100 were determined in four different ways and found to be identical. Deletion of one of the copy number control genes, copB, together with its promoter gives rise to plasmid copy mutants with an increased copy number. The increase was found to be 8- and 3.5-fold for plasmids R1 and R100, respectively. These deletion derivatives were found to be extremely sensitive to the presence of CopB activity from their own parent plasmid but not to that of the other plasmid. Hence, the CopB protein and its target are plasmid-specific and not FII-group-specific. These results are consistent with the high degree of nonhomology between plasmids R1 and R100 in a 250-bp region covering the distal part of the copB gene and the repA promoter region, which contains the target for the CopB protein.     

The electrochemical H+ gradient in the yeastRhodotorula glutinis

The electrochemical gradient of protons, , was estimated in the obligatory aerobic yeastRhodotorula glutinis in the pH₀ range from 3 to 8.5. The membrane potential, , was measured by steady-state distribution of the hydrophobic ions, tetraphenylphosphonium (TPP⁺) for negative above pH₀ 4.5, and thiocyanate (SCN^–) for positive below pH₀ 4.5. The chemical gradient of H⁺ was determined by measuring the chemical shift of intracellular P_i by³¹P-NMR at given pH₀ values. The values of pH_i increased almost linearly from 7.3 at pH₀ 3 to 7.8 at pH₀ 8.5. In the physiological pH₀ range from 3.5 to 6, was fairly constant at values between 17–18 KJ mol^–1, gradually decreasing at pH₀ above 6. In deenergized cells, the intracellular pH_i decreased to values as low as 6, regardless of whether the cell suspension was buffered at pH₀ 4.5 or 7.5. There was no membrane potential detectable in deenergized cells.     

The steady-state rate equation for cytochrome c oxidase based on a minimal kinetic scheme

A minimal catalytic cycle for cytochrome c oxidase has been suggested, and the steady-state kinetic equation for this mechanism has been derived. This equation has been used to simulate experimental data for the pH dependence of the steady-state kinetic parameters, kcat and Km. In the simulations the rate constants for binding and dissociation of cytochrome c and for two internal electron-transfer steps have been allowed to vary, whereas fixed experimental values (for pH 7.4) have been used for the other rate constants. The results show that the dissociation of the product, ferricytochrome c, cannot be rate-limiting under all conditions, but that intramolecular electron-transfer steps also limit the rate. They also demonstrate that Km can differ considerably from the dissociation constant for the cytochrome c-oxidase complex. Published values for the rate constant for the dissociation of ferricytochrome c are too small to account for the steady-state rates. It is suggested that, at high concentrations, ferryocytochrome c transfers an electron to a cytochrome c molecule which remains bound to the oxidase. This can also explain the nonhyperbolic kinetics, which is observed at low substrate concentrations.     

Zinc supplements and reproduction in grazing ewes

Six experiments were carried out in Western Australia to further investigate the effect of zinc supplementation on the reproductive performance of grazing Merino ewes. There was a small increase (7%,P=0.1) in the number of lambs produced by zinc-supplemented ewes in only one of the six experiments. Plasma zinc levels just prior to lambing were increased by zinc supplementation in two experiments. These results are discussed and compared to the previously published reproductive responses to zinc supplementation by grazing Merino ewes.     

NADPH-specific and NADH-specific xylose reduction is catalyzed by two separate enzymes in Pachysolen tannophilus

Summary Pachysolen tannophilus contains — in addition to an NADPH-linked xylose reductase — a separate NADH-linked one, in this respect differing from the yeast Pichia stipitis. Both enzyme proteins can conveniently be separated from each other by either ion exchange chromatography or chromatofocusing.     

Fly photoreceptors and temperature: Relative UV-sensitivity is increased by cooling

Intracellular responses from blowfly photoreceptor cells were recorded at various temperatures in order to study the behaviour of the transduction system, with particular reference to spectral sensitivity. with decreased temperature the V-log I functions showed a reduction in amplitude and the responses showed a slowed time course. For double peaked spectral sensitivity function the UV or 350 nm peak was much less dependent on temperature than the peak in the visible region. The higher UV-sensitivity is interpreted in terms of the sensitizing pigment theory to indicate changes in the effectiveness of energy transfer between the two chromophores.     

Deurithitrema gingae n.g., n.sp. (Digenea: Plagiorchioidea) from the kidneys of a salt-water crocodile (Crocodylus porosus Schneider) from Australia

Summary The name Deurithitrema gingae n. g., n. sp. is proposed for worms from the kidneys of one Crocodylus porosus from Darwin, Northern Territory, Australia. The following combination of characters distinguishes the new genus from all others: prepharynx very short; oesophagus absent; caeca reaching to posterior end of body; testes tandem in middle third of body, contiguous or amost so; cirrus-sac large, over-reaches ventral sucker, contains bipartite seminal vesicle, prostatic regions and ejaculatory duct with spines around its opening into genital atrium; genital pore immediately in front of ventral sucker; uterus coiled to posterior end of body; vitellaria lateral from ventral sucker to slightly behind testes; excretory vesicle small, receiving a single ventro-lateral duct from each side behind end of uterine loops. Deurithitrema belongs to the superfamily Plagiorchioidea Dollfus, 1930. It superficially resembles a number of genera, but its relationships within the superfamily are obscure.     

A critical scanning electron microscope study of the scolex of Bothriocephalus acheilognathi Yamaguti, 1934, with a review of the taxonomic history of the genus Bothriocephalus parasitizing cyprinid fishes

Summary Bothriocephalus specimens from one Ctenopharyngodon idella (O+) were fixed using three different techniques and the scolex morphology was examined using scanning electron microscopy. The scolices were compared with figures of the five species of Bothriocephalus and one species of Schyzocotyle previously described from cyprinid fishes. The taxonomic history of the Bothriocephalus species parasitizing cyprinid fish is reviewed. It is concluded that there is only one Bothriocephalus species parasitizing cyprinid fishes, and that continued use should be made of the name B. acheilognathi Yamaguti, 1934. ac]19840326     

Demonstration of a beta-casomorphin immunoreactive material in the plasma of newborn calves after milk intake

Blood was collected from newborn calves before and after their first milk intake after birth; extracts of plasma were assayed by radioimmunoassay for the presence of beta-casomorphin-7 immunoreactive materials. No beta-casomorphin immunoreactivity was found in samples collected before milk ingestion; however, in samples collected after milk ingestion a beta-casomorphin-7 immunoreactive material was detected. Chromatographic characterization showed that this material was not identical with beta-casomorphin-7 but might rather represent a precursor thereof. The material proved resistant to enzymatic attack during a 30-min incubation period at 37 degrees C in the plasma of newborn calves, whereas beta-casomorphin-7 was degraded under these conditions. A physiological significance of beta-casomorphin-7 eventually cleaved from such a precursor material at any site in the newborn mammal is suggested.
