A study of thin sections of hyphae of Streptomyces violaceoruber in the electron microscope showed that the structure of the walls and the mode of formation of cross-walls are similar to those of Gram-positive bacteria. A beaded structure was seen in some regions of the wall, and the significance of this observation is discussed in relation to previous studies of the fine structure of bacterial cell walls. Elements of the intracytoplasmic membrane system appear to be involved in the process of cross-wall formation. The walls of the hyphae of the aerial mycelium divide into two layers before the spores are formed, and only the inner component of the wall grows inwards to form the cross-walls and so delimit the spores. The outer component remains intact for a time and acts as a sheath around the developing spores. Finally the sheath breaks and the spores are liberated. This process is contrasted with the formation of endospores in eubacteria. When the spores germinate, the walls of the germ tubes are continuous with those of the spores. 相似文献

The structure of the flight muscle of a dragonfly (Aeshna sp.) has been studied with the light and electron microscopes, and the organization of this specialized tubular muscle is described. This tissue is characterized by the great development of the sarcosomes, which are slab-like and are arranged within the fiber opposite each sarcomere of the radially oriented lamellar myofibrils. A well developed and highly ordered sarcoplasmic reticulum is present, consisting of perforated curtain-like cisternae extending across the face of each fibril, together with tubular invaginations of the fiber plasma membrane situated within indentations in the sarcosomes and traversing the fibril surface midway between the Z and M levels. The structure of these fibers, and notably the organization of the reticulum, is compared with that of other types of muscle, and the possible role of the two components of the sarcoplasmic reticulum in the contraction physiology of the dragonfly muscle fiber is discussed. 相似文献

Illus. 1959.—A total of 118 leaf whorls and corresponding nodes from a total of 9 species of Equisetum were studied in serial cross-section. The number of whorls having the same leaf number as the 2 adjacent ones was 67. The size of the arc measured in degrees of circumference occupied by each leaf, as well as its position relative to leaves of adjacent whorls were measured. The disposition of the internodal and trans-nodal vascular strands was determined for each of the nodes. The average per cent deviations from theoretical leaf size within whorls were: whorls not involved in change in leaf number, 3.9; those with fewer leaves than the one below, 9.5; those with more leaves than one above, 7.1; those with more leaves than one below, 9.4; those with different leaf number from the 2 adjacent ones, 12.5. In Equisetum, it is a general rule that (1) a leaf which falls directly above a leaf in the next whorl where the younger whorl possesses fewer leaves than the older one is usually the largest leaf within its whorl, (2) a leaf which falls directly below a leaf in the next younger whorl where the younger whorl possesses fewer leaves than the older one is usually the smallest leaf within its whorl, (3) a leaf which falls directly above a leaf in the next older whorl where the younger whorl possesses a greater number of leaves than the older one is usually the smallest leaf in its whorl, and (4) a leaf which falls directly below a leaf in the next younger whorl where the younger whorl possesses a greater number of leaves than the older one is usually the largest leaf within its whorl. The numerous variations in the disposition of vascular tissue associated with changes in leaf number are described. Double leaf traces which originate in various ways are common, as well as vertical strands traversing the nodes. The leaf trace system is considered to be determined by the number, position, and relative sizes of leaf primordia. The enucleation technique has been applied to Amoeba proteus by several laboratories in attempts to determine whether the cytoplasm is capable of nucleus-independent ribonucleic acid synthesis. This cell is very convenient for micrurgy, but its use requires a thorough starvation period to eliminate the possibility of metabolic influence by food vacuoles and frequent washings and medium renewal to maintain asepsis. In the experiments described here, amoebae were starved for periods of 24 to 96 hours, cut into nucleated and enucleated halves, and exposed to either C-14 uracil, C-14 adenine, C-14 orotic acid, or a mixture of all three. When the starvation period was short (less than 72 hours), organisms (especially yeast cells) contained within amoeba food vacuoles frequently showed RNA synthesis in both nucleated and enucleated amoebae. When the preperiod of starvation was longer than 72 hours, food vacuole influence was apparently negligible, and a more meaningful comparison between enucleated and nucleated amoebae was possible. Nucleated cells incorporated all three precursors into RNA; enucleated cells were incapable of such incorporation. The experiments indicate a complete dependence on the nucleus for RNA synthesis. The conflict with the experimental results of others on this problem could possibly stem from differences in culture conditions, starvation treatment, or experimental conditions. For an unequivocal answer in experiments of this design, ideally the cells should be capable of growth on an entirely synthetic medium under aseptic conditions. The use of a synthetic medium (experiments with A. proteus are done under starvation conditions) would permit, moreover, a more realistic comparison of metabolic capacities of nucleated and enucleated cells. 相似文献
