Two forms of glutamine synthetase in free-living root-nodule bacteria.   总被引:23,自引:0,他引:23  
Cell-free extracts of Rhizobium japonicum 61A76 contain two forms of glutamine synthetase (EC 6.3.1.2) which can be easily separated by isoelectric focusing. The more acid form (pI = 5.4), like the enzyme from E. coli, is stable at 50° and catalyses an ADP-dependent transferase reaction, whose inhibition by excess Mg++ can be relieved by snake venom phosphodiesterase. The more alkaline form (pI = 6.1) is labile at 50°, and catalyses and ADP-dependent transferase reaction which is strongly inhibited by Mg++ regardless of phosphodiesterase treatment. We have also observed the two forms of the enzyme in a nitrogenaseless mutant of 61A76, and in other strains of rhizobia, but only the acid form in E. coli W, A. vinelandii OP, and K. pneumoniae M 51A.  相似文献

Acyl-CoA:diacylglycerol acyltransferase (DGAT) catalyzes the terminal step in triglyceride (TG) synthesis using diacylglycerol (DAG) and fatty acyl-CoA as substrates. In the liver, the production of VLDL permits the delivery of hydrophobic TG from the liver to peripheral tissues for energy metabolism. We describe here a novel high-content, high-throughput LC/MS/MS-based cellular assay for determining DGAT activity. We treated endogenous DGAT-expressing cells with stable isotope-labeled [13C18]oleic acid. The [13C18]oleoyl-incorporated TG and DAG lipid species were profiled. The TG synthesis pathway assay was optimized to a one-step extraction, followed by LC/MS/MS quantification. Further, we report a novel LC/MS/MS method for tracing hepatic TG synthesis and VLDL-TG secretion in vivo by administering [13C18]oleic acid to rats. The [13C18]oleic acid-incorporated VLDL-TG was detected after one-step extraction without conventional separation of TG and recovery by derivatizing [13C18]oleic acid for detection. Using potent and selective DGAT1 inhibitors as pharmacological tools, we measured changes in [13C18]oleoyl-incorporated TG and DAG and demonstrated that DGAT1 inhibition significantly reduced [13C18]oleoyl-incorporated VLDL-TG. This DGAT1-selective assay will enable researchers to discern differences between the roles of DGAT1 and DGAT2 in TG synthesis in vitro and in vivo.  相似文献

A new red basic dye, which has specificities similar to blue basic dyes and which exhibits metachromasia, has been named Darrow red, and is now commercially available. In a staining solution, prepared as indicated, it is stable for about 1 mo. Procedure: Dissolve 50 mg of Darrow red in 200 ml of 0.2 M acetic acid. Boil gently for 10 min; cool and filter. Stain frozen sections or decerated and hydrated mounted paraffin sections 20-30 min. Differentiate and dehydrate rapidly in 50, 70 and 95% ethyl alcohols followed by n-butyl alcohol, xylene and covering in synthetic resin.  相似文献

Protease-activated receptor (PAR)-4 is a low affinity thrombin receptor with slow activation and desensitization kinetics relative to PAR-1. This study provides novel evidence that cardiomyocytes express functional PAR-4 whose signaling phenotype is distinct from PAR-1 in cardiomyocytes. AYPGKF, a modified PAR-4 agonist with increased potency at PAR-4, activates p38-mitogen-activated protein kinase but is a weak activator of phospholipase C, extracellular signal-regulated kinase, and cardiomyocyte hypertrophy; AYPGKF and thrombin, but not the PAR-1 agonist SFLLRN, activate Src. The observation that AYPGKF and thrombin activate Src in cardiomyocytes cultured from PAR-1(-/-) mice establishes that Src activation is via PAR-4 (and not PAR-1) in cardiomyocytes. Further studies implicate Src and epidermal growth factor receptor (EGFR) kinase activity in the PAR-4-dependent p38-mitogen-activated protein kinase signaling pathway. Thrombin phosphorylates EGFRs and ErbB2 via a PP1-sensitive pathway in PAR-1(-/-) cells that stably overexpress PAR-4; the Src-mediated pathway for EGFR/ErbB2 transactivation underlies the protracted phases of thrombin-dependent extracellular signal-regulated kinase activation in PAR-1(-/-) cells that overexpress PAR-4 and in cardiomyocytes. These studies identify a unique signaling phenotype for PAR-4 (relative to other cardiomyocyte G protein-coupled receptors) that is predicted to contribute to cardiac remodeling and influence the functional outcome at sites of cardiac inflammation.  相似文献

The musculature of the shoulder, arm, and forearm was studied in 19 genera of squirrels, representing the Pteromyinae (flying squirrels) and all 7 tribes of the Sciurinae (tree and ground squirrels). The objective was to locate derived anatomical features of functional or phylogenetic significance and to determine how much morphological variation underlies the diverse locomotor behavior of squirrels, which includes terrestrial and arboreal bounding, climbing, digging, and gliding. The fossil evidence suggests that arboreality is primitive for squirrels, and in fact tree squirrels appear to represent the primitive sciurid morphology. Ground squirrels are less uniform and exhibit a few derived features, including a clavobrachialis muscle not seen in other squirrels. Pygmy tree squirrels, which have evolved independently in three tribes, exhibit convergence of forelimb anatomy, including the loss or reduction of several muscles in the shoulder and forearm. The forelimb anatomy of flying squirrels is the most derived and differs from that of tree squirrels in details of shoulder, arm, and forearm musculature. Some of these muscular differences among squirrels have phylogenetic significance, being shared by closely related genera, but none has significance above the tribal level. We examined the role of prostaglandins or prostaglandin-producing cells in the regulation of proliferation and generation of specific cytotoxicity in one-way mixed lymphocyte cultures of mouse spleen cells. Cultures treated with indomethacin or other prostaglandin synthesis inhibitors resulted in enhanced proliferation and cytotoxicity. The level of prostaglandins produced in vitro, as measured by RIA, was 10?8M and was found to be completely blocked by indomethacin. Adding back 10?8M PG restored baseline (control) proliferative responses. Kinetics of the enhanced MLC response were unchanged from controls as were the specificities of the cytotoxic cells. Cells from indomethacin-treated cultures were more efficient at killing targets than those from control cultures. Prostaglandins appear to have a preferential effect on the induction of cytotoxic cells.  相似文献

Predictability in the composition of tropical assemblages of insect herbivores was studied using a sample of 35,952 caterpillars (Lepidoptera) from 534 species, feeding on 69 woody species from 45 genera and 23 families in a lowland rainforest in Papua New Guinea. Caterpillar assemblages were strongly dominated by a single species (median 48% of individuals and 49% of biomass). They were spatially and temporally constant (median normalized expected species shared (NESS) similarity between assemblages from the same host was greater than or equal to 0.85 for three sites 8-17 km apart as well as for three four-month periods of the year). Further, the median presence of species was 11 months per year. Assemblages on hosts from different families and genera were virtually disjunct (NESS similarity less than 0.05) as the caterpillars were mostly specialized to a single plant family (77% of species) and, within families, to a single genus (66% of species), while capable of feeding on multiple congeneric hosts (89% of species). The dominance of caterpillar assemblages by a small number of specialized species, which also exhibited low spatial and temporal variability, permitted robust and reliable estimates of assemblage composition and between-assemblage similarity from small samples, typically less than 300 individuals per host plant. By contrast, even considerably larger samples were insufficient for estimates of species richness. A sample of 300 individuals was typically obtained from 1,050 m(2) of foliage sampled during 596 tree inspections (i.e. a particular tree sampled at a particular time) in the course of 19 sampling days (median values from 69 assemblages). These results demonstrate that, contrary to some previous suggestions, insect herbivore assemblages in tropical rainforests have a predictable structure and, as such, are amenable to study.  相似文献
